Fibrosis is a common lesion in different pathologic diseases and is defined by the excessive accumulation of collagen. Different approaches have been used to treat different conditions characterized ...by fibrosis. FDA and EMA approved collagenase to treat palmar fibromatosis, Dupuyten disease. EMA approved additionally its use in severe Peyronie disease, but it has been used off label in other conditions.1, 2. Approved treatment includes up to 3, in palmar fibromatosis or up to 8, in penile fibromatosis, injections followed by finger extension or penile modelling procedures, typically causing severe pain. Frequently single injections are enough to treat palmar fibromatosis. 3, The need to inject repeatedly doses of this enzyme can be originated by by the labile nature of collagenase which exhibits a complete activity loss after short periods of time. Herein, a novel strategy to manage this enzyme based on the synthesis of polymeric nanocapsules which contains collagenase housed within their matrix is presented. These nanocapsules have been engineered for achieving a gradual release of the encapsulated enzyme for longer times which can be up to ten days. The efficacy of these nanocapsules have been tested in murine model of local dermal fibrosis yielding higher fibrosis reduction in comparison with the injection of free enzyme which represent a significant improvement over conventional therapy.

La epidemia de obesidad afecta en forma transversal a todo el ciclo vital. En particular, en las últimas décadas se ha observado un incremento de la obesidad severa en adolescentes. En esta etapa de ...la vida caracterizada por profundos cambios físicos, emocionales y gran vulnerabilidad, la obesidad severa tiene pocas opciones efectivas de tratamiento. En adolescentes, los resultados del tratamiento centrado en modificaciones del estilo de vida son modestos y el arsenal farmacológico muy acotado y de poca efectividad. La Cirugía Bariátrica (CB) ha surgido como una opción terapéutica viable para un grupo seleccionado de adolescentes. El objetivo de esta revisión es presentar una visión actualizada respecto a indicaciones, contraindicaciones, complicaciones y resultados de estos procedimientos en población adolescente.
Resumen: En los últimos años, la obesidad severa en adolescentes ha aumentado a nivel mundial y Chile no es la excepción a este fenómeno. Es conocido que esta condición aumenta exponencialmente los ...riesgos para la salud y se asocia a mortalidad prematura. Desde el año 2008, diversas guías de tratamiento de obesidad pediátrica han incluido a la cirugía bariátrica como una estrategia de tratamiento para adolescentes obesos severos seleccionados. Estos procedimientos han mostrado ser seguros y eficaces en adultos. Un cuerpo emergente de evidencia demuestra que, en centros especializados, en el corto y mediano plazo se obtendrían resultados similares en adolescentes. Sin embargo, en este grupo de pacientes, la cirugía bariátrica tiene implícitos otros riesgos inherentes a la etapa de desarrollo en que se encuentran, y los resultados y complicaciones especialmente de largo plazo en gran medida son aún desconocidos. Por lo anterior y para el logro de los resultados esperados, es muy importante que la selección de pacientes, la cirugía y el seguimiento, sean realizados por equipos multidisciplinarios calificados, en centros hospitalarios que cuenten con la infraestructura adecuada, siendo imprescin dible la adherencia de por vida al seguimiento médico y nutricional. El objetivo de este documento es presentar la postura de la Rama de Nutrición de la Sociedad Chilena de Pediatría (SOCHIPE) frente a las diversas aristas a considerar para la adecuada indicación de estos procedimientos en adolescentes obesos severos.
Mitochondrial DNA sequences of the two hypervariable regions HVS-I and HVS-II were determined for 233 unrelated autochthonous individuals from East, Central and West Pyrenees. Resulting haplogroups ...were confirmed with RFLPs. Although the distribution of mitochondrial DNA haplogroups in the Pyrenees presents differences between populations, the results obtained may well support the hypothesis that the mountain chain did not act as a barrier to gene flow among Pyrenean populations. These preliminary data could be helpful for the understanding of the present distribution of mitochondrial DNA haplogroups in Iberian Peninsula and its phylogeny.
The effect of N-isopropyl oxamate on the activity of α-hydroxyacid dehydrogenase-isozyme II (HADH-isozyme II) from Trypanosoma cruzi was investigated. The kinetic studies showed that this substance ...was a competitive inhibitor of this isozyme. The attachment of the nonpolar isopropylic branched chain to the nitrogen of oxamate increased 12-fold the affinity of N-isopropyl oxamate for the active site of T. cruzi HADH-isozyme II. N-isopropyl oxamate was a selective inhibitor of HADH-isozyme II, since other T. cruzi dehydrogenases were not inhibited by this substance. Since HADH-isozyme II participates in the energy metabolism of T. cruzi, a trypanocidal effect can be expected with inhibitors of this isozyme. However, although it was not possible to detect any trypanocidal activity with N-isopropyl oxamate when the ethyl ester was tested as a possible trypanocidal prodrug, the expected trypanocidal effect was obtained, comparable to that obtained with nifurtimox and benznidazole.
