Bird migrations are impressive behavioral phenomena, representing complex spatiotemporal strategies to balance costs of living while maximizing fitness. The field of bird migration research has made ...great strides over the past decades, yet fundamental gaps remain. Technologies have sparked a transformation in the study of bird migration research by revealing remarkable insights into the underlying behavioral, cognitive, physiological and evolutionary mechanisms of these diverse journeys. Here, we aim to encourage broad discussions and promote future studies by highlighting research fields that are characterized by major knowledge gaps or conflicting evidence, namely the fields of navigation, social learning, individual development, energetics and conservation. We approach each topic by summarizing the current state of knowledge and provide a future outlook of ideas and state-of-the-art methods to further advance the field. Integrating knowledge across these disciplines will allow us to understand the adaptive abilities of different species and to develop effective conservation strategies in a rapidly changing world.

Although it is known that changes in land use and climate have an impact on ecological communities, it is unclear which of these factors is currently most important. We sought to determine the ...influence of land-use and climate alteration on changes in the abundance of Central European birds. We examined the impact of these factors by contrasting abundance changes of birds of different breeding habitat, latitudinal distribution, and migratory behavior. We examined data from the semiquantitative Breeding Bird Atlas of Lake Constance, which borders Germany, Switzerland, and Austria. Changes in the regional abundance of the 159 coexisting bird species from 1980-1981 to 2000-2002 were influenced by all three factors. Farmland birds, species with northerly ranges, and long-distance migrants declined, and wetland birds and species with southerly ranges increased in abundance. A separate analysis of the two decades between 1980-1981 and 1990-1992 and between 1990-1992 and 2000-2002 showed that the impact of climate change increased significantly over time. Latitudinal distribution was not significant in the first decade and became the most significant predictor of abundance changes in the second decade. Although the spatial scale and temporal resolution of our study is limited, this is the first study that suggests that climate change has overtaken land-use modification in determining population trends of Central European birds.
Autoimmune diseases are characterized by dendritic cell (DC)‐driven activation of pro‐inflammatory T cell responses. Therapeutic options for these severe diseases comprise small molecules such as ...dimethyl fumarate, or “gasotransmitters” such as CO. Herein we describe the synthesis of bifunctional enzyme‐triggered CO‐releasing molecules (ET‐CORMs) that allow the simultaneous intracellular release of both CO and methyl fumarate. Using bone‐marrow‐derived DCs the impressive therapeutic potential of these methyl fumarate‐derived compounds (FumET‐CORMs) is demonstrated by strong inhibition of lipopolysaccharide‐induced pro‐inflammatory signaling pathways and blockade of downstream interleukin‐12 or ‐23 production. The data also show that FumET‐CORMs are able to transform DCs into an anti‐inflammatory phenotype. Thus, these novel compounds have great clinical potential, for example, for the treatment of psoriasis or other inflammatory conditions of the skin.
Safe CO delivery: Linked ET‐CORM‐(di)methyl fumarates (FumET‐CORMs) were synthesized and investigated for their synergistic anti‐inflammatory and immune‐modulating properties. The results show a stronger HO‐1 induction with simultaneous inhibition of STAT1 phosphorylation in primary murine DCs relative to dimethyl fumarate. In addition, inhibited secretion of the pro‐inflammatory cytokines IL‐12 and IL‐23 could be detected.
Triple-negative breast cancer (TNBC), representing the most aggressive form of breast cancer with currently no targeted therapy available, is characterized by an inflammatory and hypoxic tumor ...microenvironment. To date, a broad spectrum of anti-tumor activities has been reported for phenanthroindolizidine alkaloids (PAs), however, their mode of action in TNBC remains elusive. Thus, we investigated six naturally occurring PAs extracted from the plant Tylophora ovata: O-methyltylophorinidine (1) and its five derivatives tylophorinidine (2), tylophoridicine E (3), 2-demethoxytylophorine (4), tylophoridicine D (5), and anhydrodehydrotylophorinidine (6). In comparison to natural (1) and for more-in depth studies, we also utilized a sample of synthetic O-methyltylophorinidine (1s). Our results indicate a remarkably effective blockade of nuclear factor kappa B (NFκB) within 2 h for compounds (1) and (1s) (IC50 = 17.1 ± 2.0 nM and 3.3 ± 0.2 nM) that is different from its effect on cell viability within 24 h (IC50 = 13.6 ± 0.4 nM and 4.2 ± 1 nM). Furthermore, NFκB inhibition data for the additional five analogues indicate a structure–activity relationship (SAR). Mechanistically, NFκB is significantly blocked through the stabilization of its inhibitor protein kappa B alpha (IκBα) under normoxic as well as hypoxic conditions. To better mimic the TNBC microenvironment in vitro, we established a 3D co-culture by combining the human TNBC cell line MDA-MB-231 with primary murine cancer-associated fibroblasts (CAF) and type I collagen. Compound (1) demonstrates superiority against the therapeutic gold standard paclitaxel by diminishing spheroid growth by 40% at 100 nM. The anti-proliferative effect of (1s) is distinct from paclitaxel in that it arrests the cell cycle at the G0/G1 state, thereby mediating a time-dependent delay in cell cycle progression. Furthermore, (1s) inhibited invasion of TNBC monoculture spheroids into a matrigel®-based environment at 10 nM. In conclusion, PAs serve as promising agents with presumably multiple target sites to combat inflammatory and hypoxia-driven cancer, such as TNBC, with a different mode of action than the currently applied chemotherapeutic drugs.
Conceptually, premature initiation of post-wound angiogenesis could interfere with hemostasis, as it relies on fibrinolysis. The mechanisms facilitating orchestration of these events remain poorly ...understood, however, likely due to limitations in discerning the individual contribution of cells and extracellular matrix. Here, we designed an in vitro Hemostatic-Components-Model (HCM) to investigate the role of the fibrin matrix as protein factor-carrier, independent of its cell-scaffold function. After characterizing the proteomic profile of HCM-harvested matrix releasates, we demonstrate that the key pro-/anti-angiogenic factors, VEGF and PF4, are differentially bound by the matrix. Changing matrix fibrin mass consequently alters the balance of releasate factor concentrations, with differential effects on basic endothelial cell (EC) behaviors. While increasing mass, and releasate VEGF levels, promoted EC chemotactic migration, it progressively inhibited tube formation, a response that was dependent on PF4. These results indicate that the clot's matrix component initially serves as biochemical anti-angiogenic barrier, suggesting that post-hemostatic angiogenesis follows fibrinolysis-mediated angiogenic disinhibition. Beyond their significance towards understanding the spatiotemporal regulation of wound healing, our findings could inform the study of other pathophysiological processes in which coagulation and angiogenesis are prominent features, such as cardiovascular and malignant disease.
Climate change has been associated with both latitudinal and elevational shifts in species' ranges. The extent, however, to which climate change has driven recent range shifts alongside other ...putative drivers remains uncertain. Here, we use the changing distributions of 378 European breeding bird species over 30 years to explore the putative drivers of recent range dynamics, considering the effects of climate, land cover, other environmental variables, and species' traits on the probability of local colonisation and extinction. On average, species shifted their ranges by 2.4 km/year. These shifts, however, were significantly different from expectations due to changing climate and land cover. We found that local colonisation and extinction events were influenced primarily by initial climate conditions and by species' range traits. By contrast, changes in climate suitability over the period were less important. This highlights the limitations of using only climate and land cover when projecting future changes in species' ranges and emphasises the need for integrative, multi-predictor approaches for more robust forecasting.
The annual migration cycle of waterbirds often involves several distinct movement stages, for example within‐winter movements or the moult migration during summer, which require a high degree of ...individual flexibility in migration direction. Here, we investigate whether such flexibility is a common characteristic of waterbird migration by analysing movement behaviour of a dabbling duck, the gadwall Anas strepera, during the little studied, intermediate autumn period. The tracking of individuals via satellite transmitters (n = 7) as well as the ring re‐encounter analysis of three European gadwall populations (Germany, England, Russia) revealed that autumn movements were multidirectional. Furthermore, the comparison with winter re‐encounters suggested that autumn movements were partly independent of the movements towards subsequently used south to southwestern wintering areas. Some individuals even travelled long distances north‐ or eastwards. Accordingly, some autumn locations were characterized by a harsh climate, thus serving as temporary staging sites but necessitating further movements when wetlands freeze during winter. The occurrence of such detours or reversals of migration was confirmed by the transmitter data. Inter‐individual variability in distance and direction of autumn movements was found for both sexes and age‐classes indicating that gadwalls, in general, followed flexible movement strategies. Based on the extent of multidirectional autumn movements, we hypothesize important benefits of such flights and suggest that the analysis of year‐round movement patterns of individual animals during their distinct life‐history stages is essential to understand how they can successfully reproduce and survive.
Abstract
Wetland bird species have been declining in population size worldwide as climate warming and land-use change affect their suitable habitats. We used species distribution models (SDMs) to ...predict changes in range dynamics for 64 non-passerine wetland birds breeding in Europe, including range size, position of centroid, and margins. We fitted the SDMs with data collected for the first European Breeding Bird Atlas and climate and land-use data to predict distributional changes over a century (the 1970s–2070s). The predicted annual changes were then compared to observed annual changes in range size and range centroid over a time period of 30 years using data from the second European Breeding Bird Atlas. Our models successfully predicted ca. 75% of the 64 bird species to contract their breeding range in the future, while the remaining species (mostly southerly breeding species) were predicted to expand their breeding ranges northward. The northern margins of southerly species and southern margins of northerly species, both, predicted to shift northward. Predicted changes in range size and shifts in range centroids were broadly positively associated with the observed changes, although some species deviated markedly from the predictions. towards the north (5% northeast, 45% north, and 40% northwest), compared to a slower observed average shift of ca. 3.9 km yr
−1
. Predicted changes in range centroids were generally larger than observed changes, which suggests that bird distribution changes may lag behind environmental changes leading to ‘climate debt’. We suggest that predictions of SDMs should be viewed as qualitative rather than quantitative outcomes, indicating that care should be taken concerning single species. Still, our results highlight the urgent need for management actions such as wetland creation and restoration to improve wetland birds’ resilience to the expected environmental changes in the future.
Summary
1. Since zebra mussel invaded Lake Constance in the 1960s the number of wintering waterbirds increased fourfold. We studied the impact of predation by waterbirds (tufted duck Aythya fuligula, ...pochard Aythya ferina and coot Fulica atra) on the population of Dreissena polymorpha in winter 2001/2002. These three species, with monthly peak numbers of approximately 230 000 individuals, currently comprise up to 80% of the waterbird population wintering at Lake Constance.
2. Four different study sites and four depths, that represent typical and characteristic habitats of mussels in Upper Lake Constance, were chosen.
3. Zebra mussels were sampled before, during and after predation by waterbirds. Their biomass in shallow areas decreased by >90%; the biomass reduction in deeper areas was highly variable and dependent on the substratum. With one exception, no changes could be detected at the greatest depth (11 m).
4. Concurrent exclosure studies revealed that the decrease in zebra mussels was caused by waterbird predation. A GIS‐based approximation revealed that in an area of 1 km2 a total of approximately 750 t mussel fresh mass was removed by birds, which is equivalent to 1390 g mussels per bird per day.
5. Wintering waterbirds have a strong structural impact on the littoral community of Lake Constance and could be the key predator of zebra mussels.
