The present paper describes fundamentals, advantages and limitations of the Box-Behnken design (BBD) for the optimization of analytical methods. It establishes also a comparison between this design ...and composite central, three-level full factorial and Doehlert designs. A detailed study on factors and responses involved during the optimization of analytical systems is also presented. Functions developed for calculation of multiple responses are discussed, including the desirability function
, which was proposed by Derringer and Suich in 1980. Concept and evaluation of robustness of analytical methods are also discussed. Finally, descriptions of applications of this technique for optimization of analytical methods are presented.
The potential use of amino acids by ruminal microorganisms converting them into microbial protein for ruminants makes it challenging to supplement these nutrients in an accessible form in animals' ...diets. Several strategies to protect amino acids from ruminal degradation were reported, producing amino acids available for the protein used in the intestine called "bypass." The intercalation of biomolecules in clay mineral minerals has gained notoriety due to its ability to support, protect, transport, physicochemical properties and non-toxicity. This study aimed to investigate the incorporation of L-lysine (Lys), L-methionine (Met), and L-tryptophan (Trp) amino acids in the clay minerals sepiolite (Sep) and Veegum
(Veg) using the adsorption method. The characterization techniques of X-ray diffraction and infrared spectroscopy indicated the presence of biomolecules in the inorganic matrices. Elemental and thermal analyzes monitored the percentages of incorporated amino acids. They showed better incorporation capacities for Veg, such as Met-Veg < Lys-Veg < Trp-Veg and Lys-Sep < Met-Sep < Trp-Sep for sepiolite, except for the incorporation of Met. Matrices provide a promising alternative for planning the administration of biomolecules, using essential amino acids as models, and may offer an alternative to improve functional diet strategies.
Nutritional strategies that optimize immunity of feedlot cattle are warranted due to increasing regulations with the use of feed-grade antimicrobials. This study evaluated physiological, health, and ...performance responses of cattle receiving a synbiotic supplement (yeast-derived prebiotic + Bacillus subtilis probiotic), which replaced feed-grade antimicrobials or were fed in conjunction with monensin during the initial 45 days in the feedlot. Angus-influenced steers (n = 256) were acquired from an auction facility on day −2, and transported (800 km) to the feedlot. Shrunk BW was recorded upon arrival (day −1). Steers were allocated to 1 of 18 pens (day 0), and pens were assigned to receive (n = 6/treatment) a free-choice diet containing: (1) monensin and tylosin (RT; 360 mg/steer daily from Rumensin and 90 mg/steer daily from Tylan; Elanco Animal Health, Greenfield, IN, USA), (2) yeast-derived ingredient and B. subtilis probiotic (CC; 18 g/steer daily of Celmanax and 28 g/steer daily of Certillus; Church and Dwight Co., Inc., Princeton, NJ, USA), or (3) monensin in addition to yeast-derived and B. subtilis ingredients (RCC) as in RT and CC. Steers were assessed for bovine respiratory disease (BRD) and DMI daily. Steer BW was recorded on days 45 and 46, and averaged for final BW. Blood samples were collected on days 0, 7, 17, 31, and 45. Feed intake was greater (P ≤ 0.05) in CC vs. RCC and RT during the initial 3 weeks upon feedlot arrival. No treatment differences were noted (P ≥ 0.41) for average daily gain, BW, and feed efficiency. Incidence of BRD did not differ (P = 0.77) between treatments (average 80.1%). A greater proportion (P ≤ 0.03) of RT steers diagnosed with BRD required a second antimicrobial treatment compared with CC and RCC (57.3, 37.3, and 38.6%, respectively). Removal of steers from the trial due to severe morbidity + mortality was greater (P = 0.02) in RT vs. CC (22.4 and 7.0%), and did not differ (P ≥ 0.16) among RCC (12.9%) vs. RT and CC. Plasma glucose concentrations were greater (P ≤ 0.02) in CC vs. RCC and RT on day 7. Plasma concentrations of nonesterified fatty acids were greater (P ≤ 0.02) in RT and RCC vs. CC on day 7, and in RT vs. CC on day 17. Steers receiving the synbiotic supplement had improved response to BRD treatment, suggesting heightened immunocompetence from partially enhanced metabolism and the nutraceutical effects of B. subtilis and yeast compounds.
Many public social housing building stocks were constructed before the introduction of national thermal regulations, and, as a result, in some situations, energy poverty conditioning during severe ...winter seasons results in little to no heating habits involving active systems in order to improve building thermal performances. Besides rigorous summer seasons, climate change predictions also indicate rigorous winter seasons will occur that will prevail in some Iberia Peninsula locations, worsening this scenario for this Southern European region. Among others, understanding the extension of discomfort in social housing buildings during heating seasons is therefore essential so as to perceive the suitability of the building stock to deal with present and future climate scenarios. Thus, this article presents a thermal comfort assessment during a winter season period applied to two social housing dwellings located in Covilhã, Portugal, inhabited by elderly residents, under realistic heating habits. An experimental campaign was performed and the results show that discomfort was found to be extremely significant for the majority of the occupied time. Passive means alone and resident heating habits were not enough to achieve proper indoor thermal and humidity conditions, resulting in important losses of well-being to the risk group of the elderly.
The present research is related to the study of p-cresol oxidation reaction in aqueous phase. Firstly, the conventional advanced oxidation process (AOP) in a lab-scale batch reactor was used, seeking ...to identify the most impacting process variables and then to propose an optimization approach for ensuring the complete p-cresol degradation and the highest total organic carbon (TOC) conversion. In the AOP with the use of hydrogen peroxide as the oxidizing agent, the oxidation reaction was optimized with the aid of a factorial design, and a maximum TOC conversion of 63% was obtained. The Lumped Kinetic Model (LKM) was used to describe the profile of residual TOC concentration due to chemical species, which were categorized into two groups (refractory and non-refractory compounds). The model was able to satisfactorily describe the profile of the residual fractions of these two classes of organic compounds and allowed estimating the related kinetic constants (k) at two different temperatures, namely (a) 3.19 × 10−1 and 2.82 × 10−3 min−1 for non-refractory and refractory compounds at 80 °C and (b) 4.73 × 10−1 and 5.09 × 10−3 min−1 for the same compound classes at 90 °C, while the activation energy (Ea) of the process was 42.02 and 62.09 kJ mol−1, respectively. The kinetic modeling of organic pollutants oxidation in liquid effluents would allow to perform in situ seawater treatment on vertical reactors installed in offshore platforms and to properly release treated water into the oceans. In this way, ocean contamination caused by the exploration on offshore platforms of oil and natural gas, the main energy sources and vectors in the current world, may be remarkably reduced, thus favoring a more eco-friendly energy production.
The goal of most studies published on sand contaminants is to gather and discuss knowledge to avoid faecal contamination of water by run-offs and tide-retractions. Other life forms in the sand, ...however, are seldom studied but always pointed out as relevant. The Mycosands initiative was created to generate data on fungi in beach sands and waters, of both coastal and freshwater inland bathing sites. A team of medical mycologists and water quality specialists explored the sand culturable mycobiota of 91 bathing sites, and water of 67 of these, spanning from the Atlantic to the Eastern Mediterranean coasts, including the Italian lakes and the Adriatic, Baltic, and Black Seas. Sydney (Australia) was also included in the study. Thirteen countries took part in the initiative. The present study considered several fungal parameters (all fungi, several species of the genus Aspergillus and Candida and the genera themselves, plus other yeasts, allergenic fungi, dematiaceous fungi and dermatophytes). The study considered four variables that the team expected would influence the results of the analytical parameters, such as coast or inland location, urban and non-urban sites, period of the year, geographical proximity and type of sediment. The genera most frequently found were Aspergillus spp., Candida spp., Fusarium spp. and Cryptococcus spp. both in sand and in water. A site-blind median was found to be 89 Colony-Forming Units (CFU) of fungi per gram of sand in coastal and inland freshwaters, with variability between 0 and 6400 CFU/g. For freshwater sites, that number was 201.7 CFU/g (0, 6400 CFU/g (p = 0.01)) and for coastal sites was 76.7 CFU/g (0, 3497.5 CFU/g). For coastal waters and all waters, the median was 0 CFU/ml (0, 1592 CFU/ml) and for freshwaters 6.7 (0, 310.0) CFU/ml (p < 0.001). The results advocate that beaches should be monitored for fungi for safer use and better management.

Mayaro fever, caused by Mayaro virus (MAYV) is a sub-lethal disease with symptoms that are easily confused with those of dengue fever, except for polyarthralgia, which may culminate in physical ...incapacitation. Recently, outbreaks of MAYV have been documented in metropolitan areas, and to date, there is no therapy or vaccine available. Moreover, there is no information regarding the three-dimensional structure of the viral proteins of MAYV, which is important in the search for antivirals. In this work, we constructed a three-dimensional model of protein C of MAYV by homology modelling, and this was employed in a manner similar to that of receptors in virtual screening studies to evaluate 590 molecules as prospective antiviral agents.

bioassays were utilized to confirm the potential antiviral activity of the flavonoid epicatechin isolated from
Salacia crassifolia
(Celastraceae). The virtual screening showed that six flavonoids were promising ligands for protein C. The bioassays showed potent antiviral action of epicatechin, which protected the cells from almost all of the effects of viral infection. An effective concentration (EC
50
) of 0.247 μmol/mL was observed with a selectivity index (SI) of 7. The cytotoxicity assay showed that epicatechin has low toxicity, with a 50% cytotoxic concentration (CC
50
) greater than 1.723 µmol/mL. Epicatechin was found to be twice as potent as the reference antiviral ribavirin. Furthermore, a replication kinetics assay showed a strong inhibitory effect of epicatechin on MAYV growth, with a reduction of at least four logs in virus production. Our results indicate that epicatechin is a promising candidate for further testing as an antiviral agent against Mayaro virus and other alphaviruses.
Aging increases the risk of type 2 diabetes, and this can be prevented by dietary restriction (DR). We have previously shown that DR inhibits the downregulation of miRNAs and their processing enzymes ...- mainly Dicer - that occurs with aging in mouse white adipose tissue (WAT). Here we used fat-specific Dicer knockout mice (AdicerKO) to understand the contributions of adipose tissue Dicer to the metabolic effects of aging and DR. Metabolomic data uncovered a clear distinction between the serum metabolite profiles of Lox control and AdicerKO mice, with a notable elevation of branched-chain amino acids (BCAA) in AdicerKO. These profiles were associated with reduced oxidative metabolism and increased lactate in WAT of AdicerKO mice and were accompanied by structural and functional changes in mitochondria, particularly under DR. AdicerKO mice displayed increased mTORC1 activation in WAT and skeletal muscle, where Dicer expression is not affected. This was accompanied by accelerated age-associated insulin resistance and premature mortality. Moreover, DR-induced insulin sensitivity was abrogated in AdicerKO mice. This was reverted by rapamycin injection, demonstrating that insulin resistance in AdicerKO mice is caused by mTORC1 hyperactivation. Our study evidences a DR-modulated role for WAT Dicer in controlling metabolism and insulin resistance.
Aims: To investigate the in vitro antiviral activity of Distictella elongata (Vahl) Urb. ethanol extracts from leaves (LEE), fruits (FEE), stems and their main components. Methods and Results: The ...antiviral activity was evaluated against human herpesvirus type 1 (HSV‐1), murine encephalomyocarditis virus (EMCV), vaccinia virus Western Reserve (VACV‐WR) and dengue virus 2 (DENV‐2) by the 3‐(4, 5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) colorimetric assay. LEE presented anti‐HSV‐1 EC50 142·8 ± 5·3 μg ml−1; selectivity index (SI) 2·0 and anti‐DENV‐2 activity (EC50 9·8 ± 1·3 μg ml−1; SI 1·5). The pectolinarin (1) isolated from LEE was less active against HSV‐1 and DENV‐2. A mixture of the triterpenoids ursolic, pomolic and oleanolic acids was also obtained. Ursolic and oleanolic acids have shown antiviral activity against HSV‐1. A mixture of pectolinarin (1) and acacetin‐7‐O‐rutinoside (2) was isolated from FEE and has presented anti‐DENV‐2 activity (EC50 11·1 ± 1·6 μg ml−1; SI > 45). Besides the antiviral activity, D. elongata has disclosed antioxidant effect. Conclusions: These data shows that D. elongata has antiviral activity mainly against HSV‐1 and DENV‐2, besides antioxidant activity. These effects might be principally attributed to flavonoids isolated. Significance and Impact of the Study: Distictella elongata might be considered a promising source of anti‐dengue fever phytochemicals.
