OBJETIVO: Verificar a prevalência da substituição do almoço e do jantar por lanches em adolescentes e a associação com estado nutricional e sexo, assim como avaliar os alimentos que substituem as ...refeições. MÉTODOS: Estudo transversal com 106 adolescentes do ensino médio de uma escola técnica localizada no município de São Paulo, que responderam a um questionário sobre atitudes alimentares a partir do qual foi avaliada a frequência de substituição do almoço e do jantar por lanche e os alimentos substitutos. Foram aferidos o peso e a altura para o cálculo do índice de massa corpórea por idade, e a classificação do estado nutricional de acordo com o critério da Organização Mundial da Saúde. Foi realizada análise descritiva com nível de significância de 5%. RESULTADOS: Um terço dos adolescentes substituiu o almoço e metade deles substituiu o jantar por lanches. Verificou-se que mais meninas costumavam substituir refeições. Observou-se maior prevalência de substituição do almoço (40%) e do jantar (70%) por lanche entre adolescentes obesos. Sanduíches, salgados, pizzas e hambúrgueres foram os principais substitutos das refeições. CONCLUSÕES: Verificou-se prevalência elevada de substituição de refeições principais, principalmente do jantar, por lanches. Os substitutos do almoço e do jantar apresentavam, em sua maioria, alta densidade energética e baixo valor nutritivo.
OBJECTIVE: To verify the prevalence of replacement of lunch and dinner with snacks among adolescents and its association with gender and nutritional status, and to evaluate the food that replaces such meals. METHODS: A cross-sectional study that enrolled 106 high-school students in the city of São Paulo, Brazil. The adolescents answered questions about how often they replaced lunch and dinner with snacks, and which foods replaced those meals. Their weight and height were measured in order to calculate the body mass index, and the nutritional classification was based on the World Health Organization criteria. Data were analyzed descriptively, with a significance level of 5%. RESULTS: One third of the adolescents replaced lunch and half of them replaced dinner with snacks. It was observed that girls replaced meals more than boys. The prevalence of meal replacement by snacks was higher among obese adolescents, being 40% for lunch and 70% for dinner. Sandwiches, baked or fried salty pastries, pizzas and hamburgers were the snacks that mostly replaced the meals. CONCLUSIONS: The replacement of meals, mainly dinner, with snacks was prevalent in the studied population. Most foods consumed in replacement of lunch and dinner had high energy density and low nutritional value.

Se realizó análisis descriptivo, con nivel de significancia de 5%. RESULTADOS: Un tercio de los adolescentes sustituyó el almuerzo y mitad de ellos la cena por meriendas. Se verificó que más muchachas solían sustituir comidas. Se observó mayor prevalencia de sustitución del almuerzo (40%) y de la cena (70%) por meriendas entre los adolescentes obesos. Sándwiches, pasteles salados, pizzas y hamburguesas fueron los principales sustitutos de las comidas. CONCLUSIONES: Se verificó la prevalencia elevada de sustitución de comidas principales, especialmente la cena, por meriendas. Los sustitutos del almuerzo y de la cena presentaban, en su mayoría, alta densidad energética y bajo valor nutritivo.
ABSTRACT
By drawing on the Schumpeterian distinction between invention (i.e., new ideas and knowledge creation) and commercialization of new ideas (i.e., innovation), this paper shows that knowledge ...and innovation are both important drivers of economic growth, but have heterogeneous spatial impacts. In particular, the growth benefits accruing from knowledge seem rather selective and concentrated across space whereas the growth benefits generated by innovation seem more diffusive, and regions innovating in the absence of a strong local knowledge base can be as successful as more knowledge‐intensive regions in turning innovation into a higher growth rate, possibly by exploiting local informal knowledge and/or knowledge spillovers. These results are of great importance for the design of research and innovation policies within the frame of the Europe 2020 strategy.
The RNA helicase SUV3 and the polynucleotide phosphorylase PNPase are involved in the degradation of mitochondrial mRNAs but their roles in vivo are not fully understood. Additionally, upstream ...processes, such as transcript maturation, have been linked to some of these factors, suggesting either dual roles or tightly interconnected mechanisms of mitochondrial RNA metabolism. To get a better understanding of the turn-over of mitochondrial RNAs in vivo, we manipulated the mitochondrial mRNA degrading complex in Drosophila melanogaster models and studied the molecular consequences. Additionally, we investigated if and how these factors interact with the mitochondrial poly(A) polymerase, MTPAP, as well as with the mitochondrial mRNA stabilising factor, LRPPRC. Our results demonstrate a tight interdependency of mitochondrial mRNA stability, polyadenylation and the removal of antisense RNA. Furthermore, disruption of degradation, as well as polyadenylation, leads to the accumulation of double-stranded RNAs, and their escape out into the cytoplasm is associated with an altered immune-response in flies. Together our results suggest a highly organised and inter-dependable regulation of mitochondrial RNA metabolism with far reaching consequences on cellular physiology.

the function of ZFP423 in neurogenesis, we analyzed allelic murine mutants in which distinct functional domains are deleted. One deletion impairs mitotic spindle orientation, leading to premature cell cycle exit and Purkinje cell (PC) progenitor pool deletion. The other deletion impairs PC differentiation. In both mutants, cell cycle progression is remarkably delayed and DDR markers are upregulated in cerebellar ventricular zone progenitors. Our
evidence sheds light on the domain-specific roles played by ZFP423 in different aspects of PC progenitor development, and at the same time strengthens the emerging notion that an impaired DDR may be a key factor in the pathogenesis of JS and other ciliopathies.
Myocardial injury after non-cardiac surgery (MINS) increases the risk of cardiovascular events and deaths, which anticoagulation therapy could prevent. Dabigatran prevents perioperative venous ...thromboembolism, but whether this drug can prevent a broader range of vascular complications in patients with MINS is unknown. The MANAGE trial assessed the potential of dabigatran to prevent major vascular complications among such patients.
In this international, randomised, placebo-controlled trial, we recruited patients from 84 hospitals in 19 countries. Eligible patients were aged at least 45 years, had undergone non-cardiac surgery, and were within 35 days of MINS. Patients were randomly assigned (1:1) to receive dabigatran 110 mg orally twice daily or matched placebo for a maximum of 2 years or until termination of the trial and, using a partial 2-by-2 factorial design, patients not taking a proton-pump inhibitor were also randomly assigned (1:1) to omeprazole 20 mg once daily, for which results will be reported elsewhere, or matched placebo to measure its effect on major upper gastrointestinal complications. Research personnel randomised patients through a central 24 h computerised randomisation system using block randomisation, stratified by centre. Patients, health-care providers, data collectors, and outcome adjudicators were masked to treatment allocation. The primary efficacy outcome was the occurrence of a major vascular complication, a composite of vascular mortality and non-fatal myocardial infarction, non-haemorrhagic stroke, peripheral arterial thrombosis, amputation, and symptomatic venous thromboembolism. The primary safety outcome was a composite of life-threatening, major, and critical organ bleeding. Analyses were done according to the intention-to-treat principle. This trial is registered with ClinicalTrials.gov, number NCT01661101.
Between Jan 10, 2013, and July 17, 2017, we randomly assigned 1754 patients to receive dabigatran (n=877) or placebo (n=877); 556 patients were also randomised in the omeprazole partial factorial component. Study drug was permanently discontinued in 401 (46%) of 877 patients allocated to dabigatran and 380 (43%) of 877 patients allocated to placebo. The composite primary efficacy outcome occurred in fewer patients randomised to dabigatran than placebo (97 11% of 877 patients assigned to dabigatran vs 133 15% of 877 patients assigned to placebo; hazard ratio HR 0·72, 95% CI 0·55–0·93; p=0·0115). The primary safety composite outcome occurred in 29 patients (3%) randomised to dabigatran and 31 patients (4%) randomised to placebo (HR 0·92, 95% CI 0·55–1·53; p=0·76).
Among patients who had MINS, dabigatran 110 mg twice daily lowered the risk of major vascular complications, with no significant increase in major bleeding. Patients with MINS have a poor prognosis; dabigatran 110 mg twice daily has the potential to help many of the 8 million adults globally who have MINS to reduce their risk of a major vascular complication.

In many countries, traditional data sources for collecting injuries of workers covered by compulsory accident insurance have recently been integrated by new observatories whose results may differ. A ...comparative analysis of the Italian data collection systems related to fatal tractor accidents in agriculture was performed focusing on tractor rollover fatalities with the aim of analyzing the accident scenario.
Data from the Operational Archives of the Italian Workers Compensation Authority (INAIL), which collects injuries of workers covered by compulsory accident insurance and those of the National Surveillance System (INAIL_ASL), which provides narrative text reports of work-related fatal accidents have been analyzed and compared to the information collected by the INAIL Observatory. The INAL Observatory was recently set up to complement the collection of fatal accidents involving agricultural machinery. Italian data were then compared to data available at an international level. Fatal tractor accidents vary considerably with respect to fatal accidents in agriculture, being 10.6 and 43.7% for the Operational Archives and Surveillance System, respectively. National Surveillance System records, implemented with narrative texts allowed the accident scenario to be defined.
71.7% of fatal tractor-related accidents refer to non-ROPS equipped vehicles and of these, 26.5% involved machines originally mounted with a ROPS that had been removed or was inoperative in the folded-down position during the rollover event. Just one fatal event from a collapsed ROPS on the overturned tractor was recorded. It is interesting that 16.6% of fatal accidents involved a clear environmental factor.
A campaign to train tractor drivers on the correct use of the combination ROPS and seatbelt can contribute to decreasing rollover events with fatal outcomes. Contemporarily a strict requirement to install ROPS and a seatbelt on tractors, combined to an official inspection at the farm level, can increase the chance of survival in a rollover accident.

Lipoprotein synthesis is controlled by estrogens, but the exact mechanisms underpinning this regulation and the role of the hepatic estrogen receptor α (ERα) in cholesterol physiology are unclear. ...Utilizing a mouse model involving selective ablation of ERα in the liver, we demonstrate that hepatic ERα couples lipid metabolism to the reproductive cycle. We show that this receptor regulates the synthesis of cholesterol transport proteins, enzymes for lipoprotein remodeling, and receptors for cholesterol uptake. Additionally, ERα is indispensable during proestrus for the generation of high-density lipoproteins efficient in eliciting cholesterol efflux from macrophages. We propose that a specific interaction with liver X receptor α (LXRα) mediates the broad effects of ERα on the hepatic lipid metabolism.

Mammalian mitochondrial DNA (mtDNA) is inherited uniparentally through the female germline without undergoing recombination. This poses a major problem as deleterious mtDNA mutations must be ...eliminated to avoid a mutational meltdown over generations. At least two mechanisms that can decrease the mutation load during maternal transmission are operational: a stochastic bottleneck for mtDNA transmission from mother to child, and a directed purifying selection against transmission of deleterious mtDNA mutations. However, the molecular mechanisms controlling these processes remain unknown. In this study, we systematically tested whether decreased autophagy contributes to purifying selection by crossing the C5024T mouse model harbouring a single pathogenic heteroplasmic mutation in the tRNAAla gene of the mtDNA with different autophagy-deficient mouse models, including knockouts of Parkin, Bcl2l13, Ulk1, and Ulk2. Our study reveals a statistically robust effect of knockout of Bcl2l13 on the selection process, and weaker evidence for the effect of Ulk1 and potentially Ulk2, while no statistically significant impact is seen for knockout of Parkin. This points at distinctive roles of these players in germline purifying selection. Overall, our approach provides a framework for investigating the roles of other important factors involved in the enigmatic process of purifying selection and guides further investigations for the role of BCL2L13 in the elimination of non-synonymous mutations in protein-coding genes.
The impact of the gut microbiota in human health is affected by several factors including its composition, drug administrations, therapeutic interventions and underlying diseases. Unfortunately, many ...human microbiota datasets available publicly were collected to study the impact of single variables, and typically consist of outpatients in cross-sectional studies, have small sample numbers and/or lack metadata to account for confounders. These limitations can complicate reusing the data for questions outside their original focus. Here, we provide comprehensive longitudinal patient dataset that overcomes those limitations: a collection of fecal microbiota compositions (>10,000 microbiota samples from >1,000 patients) and a rich description of the "hospitalome" experienced by the hosts, i.e., their drug exposures and other metadata from patients with cancer, hospitalized to receive allogeneic hematopoietic cell transplantation (allo-HCT) at a large cancer center in the United States. We present five examples of how to apply these data to address clinical and scientific questions on host-associated microbial communities.
Dopamine (DA) neurons of the midbrain are at risk to become affected by mitochondrial damage over time and mitochondrial defects have been frequently reported in Parkinson's disease (PD) patients. ...However, the causal contribution of adult-onset mitochondrial dysfunction to PD remains uncertain. Here, we developed a mouse model lacking Mitofusin 2 (MFN2), a key regulator of mitochondrial network homeostasis, in adult midbrain DA neurons. The knockout mice develop severe and progressive DA neuron-specific mitochondrial dysfunction resulting in neurodegeneration and parkinsonism. To gain further insights into pathophysiological events, we performed transcriptomic analyses of isolated DA neurons and found that mitochondrial dysfunction triggers an early onset immune response, which precedes mitochondrial swelling, mtDNA depletion, respiratory chain deficiency and cell death. Our experiments show that the immune response is an early pathological event when mitochondrial dysfunction is induced in adult midbrain DA neurons and that neuronal death may be promoted non-cell autonomously by the cross-talk and activation of surrounding glial cells.
