A short description of the muon tomography demonstrator at the INFN Laboratori Nazionali di Legnaro near Padua, Italy, is given and the principal achievements owing to the data collected at that ...experimental facility are presented. In particular, the feasibility studies for several applications based on the muon-tomographic technology, within national and European projects, are discussed. The experimental problems and the procedures used to improve the performance are underlined. Material recognition by measuring simultaneously the transmission of neutron and gamma ray produced by a
252Cf source has been studied, determining the average atomic number resolving power. In ...addition, it is demonstrated the possibility to derive direct signatures able to identify light elements (C, N, O) using the measured transmission versus neutron time-of-flight. This allows one to determine the relevant elemental ratios (C/O and C/N) normally used to identify threat organic materials such as explosives and drugs.
The muon tomography technique, based on multiple Coulomb scattering of cosmic ray muons, has been proposed as a tool to detect the presence of high density objects inside closed volumes. In this ...paper a new and innovative method is presented to handle the density fluctuations (noise) of reconstructed images, a well known problem of this technique. The effectiveness of our method is evaluated using experimental data obtained with a muon tomography prototype located at the Legnaro National Laboratories (LNL) of the Istituto Nazionale di Fisica Nucleare (INFN). The results reported in this paper, obtained with real cosmic ray data, show that with appropriate image filtering and muon momentum classification, the muon tomography technique can detect high density materials, such as lead, albeit surrounded by light or medium density material, in short times. A comparison with algorithms published in literature is also presented.
Data reported in this study concern 56 women who were subjected, 1 and 6 months after delivery, to a perineal examination including, in addition to the functional evaluation of the levator ani ...muscles, the "Q-tip test". A positive Q-tip test is associated with a greater incidence of stress urinary incontinence, especially when, at the same time, the score for the functional evaluation of the levator ani muscles ("Perineal Testing") is inferior to 3. The results of this study seem to indicate a greater gravity and ventral extension of obstetric perineal injuries.
In this retrospective study, the role of Gravidic Maternal Ponderal Increase (GMPI) within pathogenetic factors of perineal obstetric injuries, is analysed. TO this purpose, clinical data of 58 ...puerperae, who underwent a functional evaluation of perineum 30 days after delivery, were examined. The results of this study seem to indicate an autonomous pathogenetic role of GMPI, besides the indirect relation deriving from the association with other risk factors (age, parity, diabetes, fetal megalosomia, ecc).
Background: The pancreatic ductal adenocarcinoma (PDAC) microenvironment is highly fibrotic and hypoxic, with poor immune cell infiltration. Recently, we showed that nucleolin (NCL) inhibition ...normalizes tumour vessels and impairs PDAC growth. Methods: Immunocompetent mouse models of PDAC were treated by the pseudopeptide N6L, which selectively inhibits NCL. Tumour-infiltrating immune cells and changes in the tumour microenvironment were analysed. Results: N6L reduced the proportion of regulatory T cells (Tregs) and myeloid-derived suppressor cells (MDSCs) and increased tumour-infiltrated T lymphocytes (TILs) with an activated phenotype. Low-dose anti-VEGFR2 treatment normalized PDAC vessels but did not modulate the immune suppressive microenvironment. RNAseq analysis of N6L-treated PDAC tumours revealed a reduction of cancer-associated fibroblast (CAF) expansion in vivo and in vitro. Notably, N6L treatment decreased IL-6 levels both in tumour tissues and in serum. Treating mPDAC by an antibody blocking IL-6 reduced the proportion of Tregs and MDSCs and increased the amount of TILs, thus mimicking the effects of N6L. Conclusions: These results demonstrate that NCL inhibition blocks the amplification of lymphoid and myeloid immunosuppressive cells and promotes T cell activation in PDAC through a new mechanism of action dependent on the direct inhibition of the tumoral stroma.
Pancreatic ductal adenocarcinoma (PDAC) is a highly aggressive and resistant cancer with no available effective therapy. We have previously demonstrated that nucleolin targeting by N6L impairs tumor ...growth and normalizes tumor vessels in PDAC mouse models. Here, we investigated new pathways that are regulated by nucleolin in PDAC. We found that N6L and nucleolin interact with β-catenin. We found that the Wnt/β-catenin pathway is activated in PDAC and is necessary for tumor-derived 3D growth. N6L and nucleolin loss of function induced by siRNA inhibited Wnt pathway activation by preventing β-catenin stabilization in PDAC cells. N6L also inhibited the growth and the activation of the Wnt/β-catenin pathway in vivo in mice and in 3D cultures derived from MIA PaCa2 tumors. On the other hand, nucleolin overexpression increased β-catenin stabilization. In conclusion, in this study, we identified β-catenin as a new nucleolin interactor and suggest that the Wnt/β-catenin pathway could be a new target of the nucleolin antagonist N6L in PDAC.
En mobilisant certaines catégories élaborées dans le cadre de ma conception de l’Analyse du Discours philosophique, je m’attache à montrer, sur un objet apparemment secondaire, les préfaces et texte ...liminaires en philosophie et sciences humaines, la fécondité de cette méthode. Je déplace l’analyse descriptive et typologique des seuils, élaborée par Genette, au profit de l’étude pragmatique d’un « agir préfacier » déterminant pour l’auto-constitution des discours théoriques. Les préfaces opèrent certaines transactions discursives permettant de surmonter le risque de tensions entre les contraintes liées aux visées de vérité et celles qui relèvent de visées communicationnelles. Elles relient en boucle le texte à son contexte et à son cotexte, négocient le mode d’inscription simultané de l’auteur et du lecteur ainsi que les conditions de la lecture, configurent l’organisation interne du livre, relient opus et corpus et contribuent à donner à une doctrine la forme de l’œuvre, voire du chef-d’œuvre.
