improved insulin sensitivity and glucose tolerance. The Ca 2+ ...âcalmodulin-dependent kinase II (CaMKII), calcineurin and AMPK pathways have been implicated in GLUT4 gene regulation based
on pharmacological evidence. Here, we have used a more specific genetic approach to establish the relative role of the three
pathways in fast and slow muscles. Plasmids coding for protein inhibitors of CaMKII or calcineurin were co-transfected in vivo with a GLUT4 enhancer-reporter construct either in normal mice or in mice expressing a kinase dead (KD) AMPK mutant. GLUT4
reporter activity was not inhibited in the slow soleus muscle by blocking either CaMKII or calcineurin alone, but was inhibited
by blocking both pathways. GLUT4 reporter activity was likewise unchanged in the soleus of KD-AMPK mice, but was significantly
reduced by incapacitation of either CaMKII or calcineurin in these mice. On the other hand, in the fast tibialis anterior (TA) muscle, calcineurin appears to exert a prominent role in the control of GLUT4 reporter activity, independent of CaMKII
and AMPK. The results point to a muscle type-specific and redundant regulation of GLUT4 enhancer based on the interplay of multiple signalling pathways, all of which are known to affect myocyte enhancing factor
2 (MEF2) transcriptional activity, a point of convergence of different pathways on muscle gene regulation.
The Path of Fertile Family (FFP) is a service for couples experiencing infertility, aimed to obtain natural conception, as an alternative to assisted reproductive techniques, developed in 2018 and ...provided by the Counseling Centre of Centro della Famiglia Foundation, an institute of culture and pastoral care of the Diocese of Treviso, in cooperation with the local socio-health unit. FFP offers to infertile couples a specific multidisciplinary path of care, including the involvement of specific gynecological, andrological, endocrinological, and psycho-relational competences, together with fertility awareness. It represents an “experiential laboratory” for the multidisciplinary diagnosis and treatment of infertility, for understanding relational processes, for the development of operational strategies and above all for the verification of the goodness of a lifestyle that is able to harmonize the couple’s project and the family’s project, couple satisfaction and parental openness. The setting of the service draws inspiration and reference from three paradigms from which significant ethical choices derive: the ecological perspective of fertility, the relational intimacy of the couple and a multidisciplinary approach. From the three founding paradigms, FFP model has been developed and progressively implemented in order to define a specific FFP Quality of Care Path (QCP) for couple infertility. Although further studies are needed to better clarify the role of each studied variable in a complex monitoring system, we identified for the first time a QCP Path based on 7 variables we present here two illustrative cases: the first one in which a natural conception has been obtained and the second one, in which the focus has been moved from natural conception to the reconstruction of relational competence. Further studies will demonstrate, on large scale, the efficacy of our protocol and of the analyzed variables.
Summary: This paper provides an overview of the conceptual and methodological process of implementing the monitoring of the path of infertile couples towards natural conception in harmony with their relational life project. A multi-professional team accompanies couples in overcoming the obstacles to fertility and in the progressive awareness that parenting is not only biological. The result is a range of goals to strive for. The reference paradigms of the service justify the ethical choices of the couple and the team. The operating protocol supports the couple commitment and guides professionals in their effective action if competent and concerted. The variables identified are the monitoring tool that gradually indicates the degree of the path progress.
A fully integrated GSM/GPRS quad-band transceiver is presented. Transplanted patients are susceptible to viral infections; thus, the aim of this study was to evaluate the features of acute rejections and the outcome of the renal graft in transplanted patients ...with herpes virus diseases. Renal biopsies from 30 renal transplanted patients undergoing early acute rejection (type IA and IB according to the Banff 97 classification) were evaluated. In total, 15 of these patients experienced cytomegalovirus (CMV) or Epstein–Barr virus disease within the first year following transplantation (group I) and 15 patients showed no evidence of viral infection (group II). No significant differences between the groups in terms of age, male/female ratio, living/cadaveric donor ratio, cold ischemia time, HLA A-B matching, pretransplant panel reactive antibody test, occurrence of post-transplant tubular necrosis, plasma levels of cyclosporin A and mean percent increase of serum creatinine at the time of the biopsy were observed. In group I biopsies, the mean number of interstitial plasma cells, as well as the mean number of CD79a-positive cells (B lymphocytes and plasma cells) was significantly higher than in group II (P<0.01 and <0.01, respectively). There was a positive correlation between the number of infections and the number of plasma cells (P<0.05). In transplanted patients, CMV can trigger the formation of anti-endothelial cell antibodies, which have been proposed to play a role in antibody-mediated rejections. We investigated whether a deposition of C4d, a marker of antibody-mediated reactions, was present in renal peritubular capillaries. In group I C4d deposition was found in five cases, while in group II it was not observed (P<0.05). In group I, 7/15 patients developed chronic allograft nephropathy vs 1/15 patients in group II (P<0.05). The estimated 1-, 5- and 8-year cumulative graft survival rates were 80, 66 and 57%, respectively, in group I, while in group II the estimated 8-year cumulative survival rate was 100% (P<0.05). In conclusion, acute rejection biopsies of patients with viral infections displayed plasma cell infiltrates and, in several cases, C4d deposition. Our study suggests a role of B lymphocytes in the pathology of these rejections and confirms the association between viral infections and poor graft survival.
Neutrinos are copiously emitted by neutron star mergers, due to the high temperatures reached by dense matter during the merger and its aftermath. Neutrinos influence the merger dynamics and shape ...the properties of the ejecta, including the resulting \(r\)-process nucleosynthesis and kilonova emission. In this work, we analyze neutrino emission from a large sample of merger radiation hydrodynamics simulations in Numerical Relativity, covering a broad range of initial masses, nuclear equation of state and viscosity treatments. We extract neutrino luminosities and mean energies, and compute quantities of interest such as the peak values, peak broadnesses, time averages and decrease time scales. We provide a systematic description of such quantities, including their dependence on the initial parameters of the system. We find that for equal-mass systems the total neutrino luminosity (several \(10^{53}{\rm erg~s^{-1}}\)) decreases for increasing reduced tidal deformability, as a consequence of the less violent merger dynamics. Similarly, tidal disruption in asymmetric mergers leads to systematically smaller luminosities. Peak luminosities can be twice as large as the average ones. Electron antineutrino luminosities dominate (initially by a factor of 2-3) over electron neutrino ones, while electron neutrinos and heavy flavour neutrinos have similar luminosities. Mean energies are nearly constant in time and independent on the binary parameters. Their values reflect the different decoupling temperature inside the merger remnant. Despite present uncertainties in neutrino modelling, our results provide a broad and physically grounded characterization of neutrino emission, and they can serve as a reference point to develop more sophisticated neutrino transport schemes.
Considering its relevance in the field of cryptography, integer factorization is a prominent application where Quantum computers are expected to have a substantial impact. Thanks to Shor's algorithm ...this peculiar problem can be solved in polynomial time. However, both the number of qubits and applied gates detrimentally affect the ability to run a particular quantum circuit on the near term Quantum hardware. In this work, we help addressing both these problems by introducing a reduced version of Shor's algorithm that proposes a step forward in increasing the range of numbers that can be factorized on noisy Quantum devices. The implementation presented in this work is general and does not use any assumptions on the number to factor. In particular, we have found noteworthy results in most cases, often being able to factor the given number with only one iteration of the proposed algorithm. Finally, comparing the original quantum algorithm with our version on simulator, the outcomes are identical for some of the numbers considered.
