There is currently an ambiguity in what controls polar mesospheric cloud (PMC) periodicities near 83 km altitude. This is primarily because satellite and ground‐based data sets cannot resolve global ...mesospheric temperature variability over the diurnal cycle. To address this limitation, we employ a global meteorological analysis and forecast system that assimilates mesospheric satellite data with two significant advances. The first is that we use output at a more rapid one hourly cadence, allowing for a quantitative description of diurnal (24 h), semidiurnal (12 h), and terdiurnal oscillations. The second is that the output drives a simple PMC parameterization which depends only on the local temperature, pressure, and water vapor concentrations. Our study focuses on results from July 2009 in the Northern Hemisphere and January 2008 in the Southern Hemisphere. We find that the 24 h migrating temperature tide as well as the 12 h and 24 h nonmigrating tides dominate northern PMC oscillations whereas the 12 h and 24 h nonmigrating tides dominate southern oscillations. Monthly averaged amplitudes for each of these components are generally 2–6 K with the larger amplitudes at lower PMC latitudes (50°). The 2 day and 5 day planetary waves also contribute in both hemispheres, with monthly averaged amplitudes from 1 to 3 K although these amplitudes can be as high as 4–6 K on some days. Over length scales of ~1000 km and timescales of ~1 week, we find that local temperature oscillations adequately describe midlatitude PMC observations.

Climate change presents a new challenge for the management of invasive exotic species that threaten both biodiversity and agricultural productivity. The invasion of exotic perennial grasses ...throughout the globe is particularly problematic given their impacts on a broad range of native plant communities and livelihoods. As the climate continues to change, pre-emptive long-term management strategies for exotic grasses will become increasingly important. Using species distribution modelling we investigated potential changes to the location of climatically suitable habitat for some exotic perennial grass species currently in Australia, under a range of future climate scenarios for the decade centred around 2050. We focus on eleven species shortlisted or declared as the Weeds of National Significance or Alert List species in Australia, which have also become successful invaders in other parts of the world. Our results indicate that the extent of climatically suitable habitat available for all of the exotic grasses modelled is projected to decrease under climate scenarios for 2050. This reduction is most severe for the three species of Needle Grass (genus
Nassella
) that currently have infestations in the south-east of the continent. Combined with information on other aspects of establishment risk (e.g. demographic rates, human-use, propagule pressure), predictions of reduced climatic suitability provide justification for re-assessing which weeds are prioritised for intensive management as the climate changes.

It is well-documented that fungicides can affect crustacean leaf shredders via two effect pathways, namely waterborne exposure and their diet (i.e., via dietary uptake of fungicides adsorbed to leaf material and an altered microorganism-mediated food quality). As a consequence of different life history strategies, the relevance of these effect pathways for aquatic shredders belonging to other taxonomic classes, for instance insects, remains unclear. Therefore, we investigated waterborne and diet-related effects in larvae of the caddisfly leaf shredder Chaetopteryx villosa (Insecta: Trichoptera) and compared our observations to previous reports on effects in adults of the crustacean leaf shredder Gammarus fossarum (Malacostraca: Amphipoda). We assessed acute waterborne effects of an organic fungicide mixture (OFM) and the inorganic fungicide copper (Cu) on the leaf consumption (n = 30) of the fourth-/fifth-instar larvae of C. villosa and their food choice (n = 49) when offered leaf material, which was either conditioned in presence or in absence of the respective fungicide(s). Moreover, the larval leaf consumption (n = 50) and physiological fitness (i.e., growth as well as lipid and protein content) were examined after subjecting C. villosa for 24 days towards the combination of both effect pathways at environmentally relevant concentrations. G. fossarum and C. villosa exhibited similar sensitivities and the same effect direction when exposed to the OFM (either waterborne or dietary pathways). Both shredders also showed the same effect direction when exposed to dietary Cu, while with regards to mortality and leaf consumption C. villosa was less sensitive to waterborne Cu than G. fossarum. Finally, as observed for G. fossarum, the combined exposure to OFM over 24 days negatively affected leaf consumption and the physiology (i.e., growth and lipid reserves) of C. villosa. While no combined Cu effects were observed for larval leaf consumption, contrasting to the observations for G. fossarum, the physiology of both shredders was negatively affected, despite partly differing effect sizes and directions. Our results suggest that C. villosa and G. fossarum are of comparable sensitivity towards waterborne and diet-related organic fungicide exposure, whereas the trichopteran is less sensitive to Cu-based waterborne fungicide exposure. However, when both pathways act jointly, organic and inorganic fungicides can affect the physiology of shredder species with completely different life history strategies. As caddisflies represent a subsidy for terrestrial consumers, these observations indicate that fungicide exposure might not only affect aquatic ecosystem functioning but also the flux of energy across ecosystem boundaries.
ABSTRACT
We present extensive ultraviolet (UV) and optical photometric and optical spectroscopic follow-up of supernova (SN) 2021gno by the ‘Precision Observations of Infant Supernova Explosions’ ...(POISE) project, starting less than 2 d after the explosion. Given its intermediate luminosity, fast photometric evolution, and quick transition to the nebular phase with spectra dominated by Ca ii lines, SN 2021gno belongs to the small family of Calcium-rich transients. Moreover, it shows double-peaked light curves, a phenomenon shared with only four other Calcium-rich events. The projected distance from the centre of the host galaxy is not as large as other objects in this family. The initial optical light-curve peaks coincide with a very quick decline of the UV flux, indicating a fast initial cooling phase. Through hydrodynamical modelling of the bolometric light curve and line velocity evolution, we found that the observations are compatible with the explosion of a highly stripped massive star with an ejecta mass of $0.8\, M_\odot$ and a 56Ni mass of 0.024 M⊙. The initial cooling phase (first light-curve peak) is explained by the presence of an extended circumstellar material comprising ∼$10^{-2}\, {\rm M}_{\odot }$ with an extension of $1100\, R_{\odot }$. We discuss if hydrogen features are present in both maximum-light and nebular spectra, and their implications in terms of the proposed progenitor scenarios for Calcium-rich transients.

This was a retrospective cross-sectional study of patients referred for multigene panel testing between March 18, 2016, and August 3, 2020, with outcomes reported between May and November 2020. The study setting included a commercial genetic testing laboratory and multicenter clinical practices. Patients with epilepsy, regardless of sociodemographic features, who received a pathogenic/likely pathogenic (P/LP) variant were included in the study. Case report forms were completed by all health care professionals.

Clinical management changes after a genetic diagnosis (ie, 1 P/LP variant in autosomal dominant and X-linked diseases; 2 P/LP variants in autosomal recessive diseases) and subsequent patient outcomes as reported by health care professionals on case report forms.
Among 418 patients, median (IQR) age at the time of testing was 4 (1-10) years, with an age range of 0 to 52 years, and 53.8% (n = 225) were female individuals. The mean (SD) time from a genetic test order to case report form completion was 595 (368) days (range, 27-1673 days). A genetic diagnosis was associated with changes in clinical management for 208 patients (49.8%) and usually (81.7% of the time) within 3 months of receiving the result. The most common clinical management changes were the addition of a new medication (78 21.7%), the initiation of medication (51 14.2%), the referral of a patient to a specialist (48 13.4%), vigilance for subclinical or extraneurological disease features (46 12.8%), and the cessation of a medication (42 11.7%). Among 167 patients with follow-up clinical information available (mean SD time, 584 365 days), 125 (74.9%) reported positive outcomes, 108 (64.7%) reported reduction or elimination of seizures, 37 (22.2%) had decreases in the severity of other clinical signs, and 11 (6.6%) had reduced medication adverse effects. A few patients reported worsening of outcomes, including a decline in their condition (20 12.0%), increased seizure frequency (6 3.6%), and adverse medication effects (3 1.8%). No clinical management changes were reported for 178 patients (42.6%).
Results of this cross-sectional study suggest that genetic testing of individuals with epilepsy may be materially associated with clinical decision-making and improved patient outcomes.
Postsynaptic density protein-95 (PSD-95), encoded by DLG4, regulates excitatory synaptic function in the brain. Here we present the clinical and genetic features of 53 patients (42 previously ...unpublished) with DLG4 variants.
The clinical and genetic information were collected through GeneMatcher collaboration. All the individuals were investigated by local clinicians and the gene variants were identified by clinical exome/genome sequencing.
The clinical picture was predominated by early onset global developmental delay, intellectual disability, autism spectrum disorder, and attention deficit-hyperactivity disorder, all of which point to a brain disorder. Marfanoid habitus, which was previously suggested to be a characteristic feature of DLG4-related phenotypes, was found in only nine individuals and despite some overlapping features, a distinct facial dysmorphism could not be established. Of the 45 different DLG4 variants, 39 were predicted to lead to loss of protein function and the majority occurred de novo (four with unknown origin). The six missense variants identified were suggested to lead to structural or functional changes by protein modeling studies.
The present study shows that clinical manifestations associated with DLG4 overlap with those found in other neurodevelopmental disorders of synaptic dysfunction; thus, we designate this group of disorders as DLG4-related synaptopathy.
Copper (Cu) exposure can increase leaf-associated fungal biomass, an important food component for leaf-shredding macroinvertebrates. To test if this positive nutritional effect supports the ...physiological fitness of these animals and to assess its importance compared to waterborne toxicity, we performed a 24-day-bioassay in combination with a 2×2 factorial design using the amphipod shredder Gammarus fossarum and a field-relevant Cu concentration of 25 μg/L (n = 65). Waterborne toxicity was negligible, while gammarids fed leaves exposed to Cu during microbial colonization exhibited a near-significant impairment in growth (∼30%) and a significantly reduced lipid content (∼20%). These effects appear to be governed by dietary uptake of Cu, which accumulated in leaves as well as gammarids and likely overrode the positive nutritional effect of the increased fungal biomass. Our results suggest that for adsorptive freshwater contaminants dietary uptake should be evaluated already during the registration process to safeguard the integrity of detritus-based ecosystems.

Calibrated Automated Thrombography (CAT) is a versatile and sensitive method for analyzing coagulation reactions culminating in thrombin generation (TG). Here, we present a CAT method for analyzing ...TG in murine whole blood by adapting the CAT assay used for measuring TG in human plasma. The diagnostically used artificial and physiologic factor XII (FXII) contact activators kaolin, ellagic acid and polyphosphate (polyP) stimulated TG in murine blood in a dose-dependent manner resulting in a gradual increase in endogenous thrombin potential and peak thrombin, with shortened lag times and times to peak. The activated FXII inhibitor rHA-Infestin-4 and direct oral anticoagulants (DOACs) interfered with TG triggered by kaolin, ellagic acid and polyP and TG was completely attenuated in blood of FXII- (
) and FXI-deficient (
) mice. Moreover, reconstitution of blood from
mice with human FXII restored impaired contact-stimulated TG. HEK293 cell-purified polyP also initiated FXII-driven TG in mouse whole blood and addition of the selective inhibitor PPX_Δ12 ablated natural polyP-stimulated TG. In conclusion, the data provide a method for analysis of contact activation-mediated TG in murine whole blood. As the FXII-driven intrinsic pathway of coagulation has emerged as novel target for antithrombotic agents that are validated in mouse thrombosis and bleeding models, our novel assay could expedite therapeutic drug development.
To examine the safety and effectiveness of assisted reproduction using sperm washing for HIV-1-serodiscordant couples wishing to procreate where the male partner is infected.
A retrospective ...multicentre study at eight centres adhering on the European network CREAThE and involving 1036 serodiscordant couples wishing to procreate. Sperm washing was used to obtain motile spermatozoa for 3390 assisted reproduction cycles (2840 intrauterine inseminations, 107 in-vitro fertilizations, 394 intra-cytoplasmic sperm injections and 49 frozen embryo transfers). An HIV test was performed in female partners at least 6 months after assisted reproduction attempt. The outcome measures recorded were number of assisted reproduction cycles, pregnancy outcome and HIV test on women post-treatment.
A total of 580 pregnancies were obtained from 3315 cycles. Pregnancy outcome was unknown in 47 cases. The 533 pregnancies resulted in 410 deliveries and 463 live births. The result of female HIV testing after assisted reproduction was known in 967 out of 1036 woman (7.1% lost to follow-up). All tests recorded were negative. The calculated probability of contamination was equal to zero (95% confidence interval, 0-0.09%).
This first multicentre retrospective study of assisted reproduction following sperm washing demonstrates the method to be effective and to significantly reduce HIV-1 transmission risk to the uninfected female partner. These results support the view that assisted reproduction with sperm washing could not be denied to serodiscordant couples in developed countries and, where possible, could perhaps be integrated into a global public health initiative against HIV in developing countries.
