Using the Gamma-Ray Spectrometer on the Mars Odyssey, we have identified two regions near the poles that are enriched in hydrogen. The data indicate the presence of a subsurface layer enriched in ...hydrogen overlain by a hydrogen-poor layer. The thickness of the upper layer decreases with decreasing distance to the pole, ranging from a column density of about 150 grams per square centimeter at -42° latitude to about 40 grams per square centimeter at -77°. The hydrogen-rich regions correlate with regions of predicted ice stability. We suggest that the host of the hydrogen in the subsurface layer is ice, which constitutes 35 ± 15% of the layer by weight.
We report maps of the concentrations of H, Si, Cl, K, Fe, and Th as determined by the Gamma Ray Spectrometer (GRS) on board the 2001 Mars Odyssey Mission for ±∼45° latitudes. The procedures by which ...the spectra are processed to yield quantitative concentrations are described in detail. The concentrations of elements determined over the locations of the various Mars landers generally agree well with the lander values except for Fe, although the mean of the GRS Fe data agrees well with that of Martian meteorites. The water‐equivalent concentration of hydrogen by mass varies from about 1.5% to 7.5% (by mass) with the most enriched areas being near Apollinaris Patera and Arabia Terra. Cl shows a distribution similar to H over the surface except that the Cl content over Medusae Fossae is much greater than elsewhere. The map of Fe shows enrichment in the northern lowlands versus the southern highlands. Silicon shows only very modest variation over the surface with mass fractions ranging from 19% to 22% over most of the planet, though a significant depletion in Si is noted in a region west of Tharsis Montes and Olympus Mons where the Si content is as low as 18%. K and Th show a very similar pattern with depletions associated with young volcanic deposits and enrichments associated with the TES Surface Type‐2 material. It is noted that there appears to be no evidence of significant globally distributed thick dust deposits of uniform composition.
On Mars, seasonal martian flow features known as recurring slope lineae (RSL) are prevalent on sun-facing slopes and are associated with salts. On Earth, subsurface interactions of gypsum with ...chlorides and oxychlorine salts wreak havoc: instigating sinkholes, cave collapse, debris flows, and upheave. Here, we illustrate (i) the disruptive potential of sulfate-chloride reactions in laboratory soil crust experiments, (ii) the formation of thin films of mixed ice-liquid water “slush” at −40° to −20°C on salty Mars analog grains, (iii) how mixtures of sulfates and chlorine
salts affect their solubilities in low-temperature environments, and (iv) how these salt brines could be contributing to RSL formation on Mars. Our results demonstrate that interactions of sulfates and chlorine salts in fine-grained soils on Mars could absorb water, expand, deliquesce, cause subsidence, form crusts, disrupt surfaces, and ultimately produce landslides after dust loading on these unstable surfaces.
The alveolar-capillary barrier is composed of epithelial and endothelial cells interacting across a fibrous extracelluar matrix (ECM). Although remodeling of the ECM occurs during several lung ...disorders, it is not known how fiber structure and mechanics influences cell injury during cyclic airway reopening as occurs during mechanical ventilation (atelectrauma). We have developed a novel in vitro platform that mimics the micro/nano-scale architecture of the alveolar microenvironment and have used this system to investigate how ECM microstructural properties influence epithelial cell injury during airway reopening. In addition to epithelial-endothelial interactions, our platform accounts for the fibrous topography of the basal membrane and allows for easy modulation of fiber size/diameter, density and stiffness. Results indicate that fiber stiffness and topography significantly influence epithelial/endothelial barrier function where increased fiber stiffness/density resulted in altered cytoskeletal structure, increased tight junction (TJ) formation and reduced barrier permeability. However, cells on rigid/dense fibers were also more susceptible to injury during airway reopening. These results indicate that changes in the mechanics and architecture of the lung microenvironment can significantly alter cell function and injury and demonstrate the importance of implementing in vitro models that more closely resemble the natural conditions of the lung microenvironment.
The Wilms tumor suppressor gene WT1 encodes a developmentally regulated transcription factor that is mutated in a subset of embryonal tumors. To test its functional properties, we developed ...osteosarcoma cell lines expressing WT1 under an inducible tetracycline‐regulated promoter. Induction of WT1 resulted in programmed cell death. This effect, which was differentially mediated by the alternative splicing variants of WT1, was independent of p53. WT1‐mediated apoptosis was associated with reduced synthesis of the epidermal growth factor receptor (EGFR), but not of other postulated WT1‐target genes, and it was abrogated by constitutive expression of EGFR. WT1 repressed transcription from the EGFR promoter, binding to two TC‐rich repeat sequences. In the developing kidney, EGFR expression in renal precursor cells declined with the onset of WT1 expression. Repression of EGFR and induction of apoptosis by mechanism that may contribute to its critical role in normal kidney development and to the immortalization of tumor cells with inactivated WT1 alleles.
The diuretic ethacrynic acid (EA) has been shown to inhibit signaling by the proinflammatory transcription factor nuclear factor-kappaB (NF-kappaB). Accordingly, we sought to determine whether this ...compound is capable of inhibiting the release of cytokines interleukin (IL)-6 and IL-10 and NO from RAW 264.7 murine macrophage-like cells stimulated with lipopolysaccharide (LPS). Additionally, we sought to determine whether EA can inhibit secretion of high-mobility group box 1 (HMGB1), a nuclear protein that is secreted by immunostimulated macrophages and functions in the extracellular milieu as a proinflammatory mediator. In a concentration-dependent manner, EA inhibited secretion of IL-6, IL-10, nitric oxide, and HMGB1. As expected, EA inhibited NF-kappaB DNA binding in LPS-stimulated RAW 264.7 cells. Treating these cells with pyrrolidine dithiocarbamate, SN50 (amino acid sequence AAVALLPAVLLALLAPVQRKRQKLMP) or 5-(thien-3-yl)-3-aminothiophene-2-carboxamide (SC-514) also inhibited LPS-induced NF-kappaB DNA binding, but these compounds failed to inhibit LPS-induced HMGB1 secretion. These findings suggested that inhibition of HMGB1 secretion by EA might occur via a mechanism unrelated to the NF-kappaB signaling pathway. Because EA is an electrophilic compound that is known to be capable of inducing expression of so-called phase 2 proteins, we sought to determine whether two other phase 2 enzyme inducers, oltipraz and DL-sulforaphane, also are capable of inhibiting HMGB1 release from immunostimulated macrophages. Incubating RAW 264.7 cells with either oltipraz or DL-sulforaphane inhibited LPS-induced HMGB1 secretion. Moreover, both EA and DL-sulforaphane inhibited relocalization of nuclear HMGB1 into the cytoplasm of LPS-stimulated RAW 264.7 cells. These data suggest that phase 2 inducers may exert anti-inflammatory effects by inhibiting secretion of the cytokine-like nuclear protein HMGB1.
The Ionospheric Connection Explorer, or ICON, is a new NASA Explorer mission that will explore the boundary between Earth and space to understand the physical connection between our world and our ...space environment. This connection is made in the ionosphere, which has long been known to exhibit variability associated with the sun and solar wind. However, it has been recognized in the 21st century that equally significant changes in ionospheric conditions are apparently associated with energy and momentum propagating upward from our own atmosphere. ICON’s goal is to weigh the competing impacts of these two drivers as they influence our space environment. Here we describe the specific science objectives that address this goal, as well as the means by which they will be achieved. The instruments selected, the overall performance requirements of the science payload and the operational requirements are also described. ICON’s development began in 2013 and the mission is on track for launch in 2018. ICON is developed and managed by the Space Sciences Laboratory at the University of California, Berkeley, with key contributions from several partner institutions.

Through an international collaboration, we analyzed seizure characteristics, EEG data, and genotypes of a cohort of patients with heterozygous variants in
. To gain insight into disease mechanisms, we performed ex vivo cortical electroporation in mouse embryos of 5 selected variants, 2 truncating and 3 missense, and evaluated on expression and quantified changes in axonal morphology.
We identified 35 patients (17 male, median age 10 years, range 2.5-23 years) carrying 32 different heterozygous variants in
, including 28 single-nucleotide variants or small insertions/deletions (12 missense, 12 frameshift or nonsense, 2 splice-site variants, and 2 in-frame deletions), and 4 microdeletions; de novo in 18 patients and inherited in 10. Seizures were reported in 31/35 (89%) patients, with a median age at onset of 3 years (range 4 months-12 years). Absence seizures occurred in 25 patients with epilepsy (81%). Nineteen patients experienced a single seizure type: absences, myoclonic absences, or absences with eyelid myoclonia and focal seizures. Nine patients had absence seizures combined with other generalized seizure types. One patient had presented with absences associated with photosensitive occipital seizures. Three other patients had generalized tonic-clonic seizures without absences. ID of variable degree was observed in 85% of the patients. Expression studies in cultured neurons showed shorter axons for the 5 tested variants, both truncating and missense variants, supporting an impaired protein function.
In most patients, the phenotype of the
-related disorder associates absence seizures with mild-to-moderate ID. In silico and in vitro evaluation of the variants in our cohort, including axonal morphogenetic experiments in cultured neurons, supports their pathogenicity, showing a hypomorphic effect.
The space shuttle launched for the last time on 8 July 2011. As with most shuttle launches, the three main engines injected about 350 t of water vapor between 100 and 115 km off the east coast of the ...United States during its ascent to orbit. We follow the motion of this exhaust with a variety of satellite and ground‐based data sets and find that (1) the shuttle water vapor plume spread out horizontally in all directions over a distance of 3000 to 4000 km in 18 h, (2) a portion of the plume reached northern Europe in 21 h to form polar mesospheric clouds (PMCs) that are brighter than over 99% of all PMCs observed in that region, and (3) the observed altitude dependence of the particle size is reversed with larger particles above smaller particles. We use a one‐dimensional cloud formation model initialized with predictions of a plume diffusion model to simulate the unusually bright PMCs. We find that eddy mixing can move the plume water vapor down to the mesopause near 90 km where ice particles can form. If the eddy diffusion coefficient is 400 to 1000 m2/s, the predicted integrated cloud brightness is in agreement with both satellite and ground‐based observations of the shuttle PMCs. The propellant mass of the shuttle is about 20% of that from all vehicles launched during the northern 2011 PMC season. We suggest that the brightest PMC population near 70°N is formed by space traffic exhaust.

To simulate planetary gamma-ray spectroscopy, gamma rays were measured during a series of five irradiations of up to 30-ton thick targets with protons beams of 1.5 and 2.5
GeV. The targets were steel ...(iron), basalt with structural steel, basalt with added S and Cl, and basalt with added H, S, and Cl. The pulsed proton beam was carefully monitored and counted. Spectra were collected with both proton beam on and beam off and with a lead shield both between the target and the germanium detector and with the lead shield removed. This set of four spectra was used to determine the fluxes of prompt gamma rays emitted from the target. Over 200 discrete gamma-ray lines per irradiation were observed and identified. Counting results for the more intense gamma rays were compiled, and gamma-ray fluxes determined for about 25 gamma rays of interest to planetary gamma-ray spectroscopy. The ratios of thermal and fast neutron induced gamma-ray fluxes between irradiations were similar. Thus the relative gamma-ray fluxes can be used in testing model calculations and interpreting planetary gamma-ray spectra.
