Vaccination is one of the most important measures to contain the COVID-19 pandemic, especially for frail patients. VACCINATE is a multicentre prospective observational study promoted by the European ...Reference Network on Rare and Complex Connective Tissue and Musculoskeletal Diseases (ERN ReCONNET) aimed at assessing the long-term outcomes of COVID-19 vaccination in patients with rare and complex connective tissue diseases (rcCTDs) in terms of efficacy and safety.
Adult rcCTDs patients were eligible for recruitment. Demographic, clinical and vaccination data were collected at enrolment. Follow-up visits were scheduled 4, 12, 24, 36 and 48 weeks after completion of the first vaccination cycle; data on adverse events, disease exacerbations and the occurrence of new SARS-CoV-2 infections were collected at these time-points.
365 rcCTDs patients (87 % female, mean age 51.8 ± 14.6 years) were recruited. Overall, 200 patients (54.8 %) experienced at least one adverse event, generally mild and in most cases occurring early after the vaccination. During follow-up, 55 disease exacerbations were recorded in 39 patients (10.7 %), distributed over the entire observation period, although most frequently within 4 weeks after completion of the vaccination cycle. The incidence of new SARS-CoV-2 infections was 8.9 per 1000 person-months, with no cases within 12 weeks from vaccine administration and an increasing trend of infections moving away from the primary vaccination cycle. Only one case of severe COVID-19 was reported during the study period.
COVID-19 vaccination seems effective and safe in rcCTDs patients. The rate of new infections was rather low and serious infections were uncommon in our cohort. No increased risk of disease flares was observed compared to previous disease history; however, such exacerbations may be potentially severe, emphasising the need for close monitoring of our patients.
Plasma electrolytic oxidation (PEO) has been a promising surface coating with better mechanical and antimicrobial parameters comparing to conventional treatment surfaces. This study evaluated the ...peri-implant bone repair using (PEO) surface coatings compared with sandblasted acid (SLA) treatment. For this purpose, 44 Wistar rats were ovariectomized (OVX-22 animals) or underwent simulated surgery (SS-22 animals) and received implants in the tibia with each of the surface coatings. The peri-implant bone subsequently underwent molecular, microstructural, bone turnover, and histometric analysis. Real-time PCR showed a higher expression of osteoprotegerin (OPG), receptor activator of nuclear kappa-B ligand (RANKL), and osteocalcin (OC) proteins in the SLA/OVX and PEO/SS groups (
< 0.05). Computed microtomography, confocal microscopy, and histometry showed similarity between the PEO and SLA surfaces, with a trend toward the superiority of PEO in OVX animals. Thus, PEO surfaces were shown to be promising for enhancing peri-implant bone repair in ovariectomized rats.

Pro- and anti-inflammatory cytokine serum levels were higher in AS patients than in controls. RANKL neutrophil expression was higher in AS patients when compared to healthy donors, but CD51/CD61 expression was lower in the classical monocyte subpopulation. Concerning osteoclastogenesis, we found no differences in the
osteoclast differentiating potential of these cells when compared to healthy donors. However, we observed low expression of CSF1R, RANK, and NFATc1 in AS OCPs.
Despite the high levels of pro-inflammatory cytokines present in AS patients, no differences in the number of OC or resorbed area were found between AS patients and healthy donors. Moreover, we observed that OCPs have low OC-specific gene expression. These findings support our hypothesis of an impaired response of OCPs to pro-osteoclastogenic stimuli
in AS patients.
Over recent decades, the world has experienced the adverse consequences of uncontrolled development of multiple human activities. In recent years, the total production of chemicals has been composed ...of environmentally harmful compounds, the majority of which have significant environmental impacts. These emerging contaminants (ECs) include a wide range of man-made chemicals (such as pesticides, cosmetics, personal and household care products, pharmaceuticals), which are of worldwide use. Among these, several ECs raised concerns regarding their ecotoxicological effects and how to assess them efficiently. This is of particular interest if marine diatoms are considered as potential target species, due to their widespread distribution, being the most abundant phytoplankton group in the oceans, and also being responsible for key ecological roles. Bio-optical ecotoxicity methods appear as reliable, fast, and high-throughput screening (HTS) techniques, providing large datasets with biological relevance on the mode of action of these ECs in phototrophic organisms, such as diatoms. However, from the large datasets produced, only a small amount of data are normally extracted for physiological evaluation, leaving out a large amount of information on the ECs exposure. In the present paper, we use all the available information and evaluate the application of several machine learning and deep learning algorithms to predict the exposure of model organisms to different ECs under different doses, using a model marine diatom (Phaeodactylum tricornutum) as a test organism. The results show that 2D convolutional neural networks are the best method to predict the type of EC to which the cultures were exposed, achieving a median accuracy of 97.65%, while Rocket is the best at predicting which concentration the cultures were subjected to, achieving a median accuracy of 100%.
As one of the major functions of bone is to provide structural support for the musculoskeletal system, it is important to evaluate its mechanical strength. Bones may be subjected to multiaxial ...stresses due to bone pathologies, accidental loads which may lead to hip, wrist fracture, or to a prosthetic joint replacement. Twist loading may lead to fractures, especially involving long bones from lower limbs. The aim of this work was to study the effect of the strain rate on the shear properties of trabecular bone samples from women with hip fracture (from 65 to 100 years). Cylindrical samples were core drilled from human femoral heads along the primary trabecular direction. The cylinder's ends were polished and embedded in blocks of polymeric material which fit the grips of the testing device. Deformation rates of 0.005, 0.01, 0.015, and 0.05 s⁻¹ were applied. Twisting tests were conducted with or without an applied axial load of 500 N. From the torque-angular displacement curves, the shear stress-strain curves were obtained. The maximum shear strength and the shear modulus (i.e. the slope of the linear region) were determined. A large scatter of the results of the shear strength and the shear modulus was found, which is probably related to the heterogeneity of nonhealthy human bone samples. There is no significant effect of the strain rate on the maximum shear stress and the shear modulus, either in tests undertaken with or without the application of an axial load. The effect of strain rate on nonhealthy bone trabecular twisting properties did not follow the trend observed on the effect of strain rate in healthy bone, where an increase is detected.
Rheumatoid arthritis (RA) is associated with higher levels of inflammatory mediators and with a more atherogenic lipid profile. Dyslipidemia can be present years before arthritis develops. ...Lymphotoxin-α (LTA) is a cytokine that mediates proinflammatory responses while also participating in lipid homeostasis, and its transcriptional activity is in part genetically determined. We examined the role of the single-nucleotide polymorphism at position 252 of the LTA gene in the genetic background of RA and dyslipidemia.
The association between the LTA 252 A>G polymorphism and disease status was examined in a nested case-control study of 388 patients with RA and 269 unrelated healthy controls, all white. Demographics and disease features were assessed, fasting lipids measured, and the use of lipid-lowering agents evaluated.
The LTA 252 A allele was more frequent in cases compared to controls (70.5% and 64.3%, respectively; p = 0.018, OR 1.325, 95% CI 1.049-1.675), as well as the A/A genotype (50.8% vs 43.5%; p = 0.025). The A/A genotype was independently associated with dyslipidemia in patients, but not in controls. Patients with RA who had the LTA 252 G/G genotype were younger at disease onset and had higher C-reactive protein (CRP) levels.
We found the LTA 252 A allele to be associated with an increased risk for developing RA in whites. The LTA 252 A/A genotype translates to a phenotype more prone to dyslipidemia, and the G/G genotype to a phenotype with earlier onset of disease and higher levels of CRP, when RA does occur. These observations highlight a possible common genetic predisposition to RA and dyslipidemia.
