The permanent epithelial cell line from the dipteran insect Chironomus tentans grows as homogeneous monolayered multicellular vesicles. Moulting hormones induce morphogenetic changes in this cell ...line while stopping cell proliferation. Electron microscopic investigations revealed bundles of microtubuli after administration of the moulting hormone 20-OH-ecdysone (20E), suggesting changes in the orientation and/or amount of cytoskeletal proteins. In order to discriminate between these two possibilities, the present investigation was started to determine the amounts of actin and tubulin mRNAs and proteins.
Reversible deprotonation of fascaplysin (1) was achieved with non‐nucleophilic bases (Scheme 1). Under basic aqueous conditions, opening of ring D of 1 occurred, yielding zwitter‐ionic reticulatine ...2a, whereas, in a methoxide‐containing MeOH solution, an unexpected addition of three molecules of MeOH to the pyridinium ring produced an isomer mixture 3 of a trimethoxy‐substituted compound (Scheme 2). Transformation of the keto group of 1 to the oxime 4A took place in the presence of pyridine as base (Scheme 3). Grignard and alkyllithium reagents added as expected to the keto group of 1, providing tertiary alcohols 5 and 6 (Scheme 4).
A 7-year-old Belgian stallion hemorrhaged excessively after castration; the hemostatic mechanism was investigated. The horse had normal one-stage prothrombin time and markedly prolonged activated ...partial thromboplastin time (APTT). Results of intrinsic coagulation factor assays were all normal with the exception of prekallikrein activity, which was markedly reduced (less than 1% activity; value for control population, 63 to 150%). Two of this horse's full siblings, a brother and sister, had markedly prolonged APTT and low prekallikrein values (2.5% and less than 1%, respectively). The addition of plasma from a normal equine plasma pool corrected the prolonged APTT in the 3 Belgian sibling with low prekallikrein activity. Prekallikrein activity in 10 other closely related Belgian horses ranged between 12.5 and 64% (mean, 29.3%), compared with 63 to 150% (mean, 91%) in 10 mixed-breed horses. In the 3 Belgian siblings with low prekallikrein activity, the APTT approached normal after prolonged incubation (15 minutes) with the contact activator and in response to addition of an ellagic acid activator. The 3 Belgian siblings with low prekallikrein activity may be homozygous for prekallikrein deficiency, whereas the other close relatives may be heterozygous for the genetic defect.
The morphogenetic changes in an epithelial cell line from Chironomus tentans that are evoked by molting hormones and molting hormone agonists are accompanied by transient changes in the concentration ...of actin and beta-tubulin protein and mRNA. As compared to controls, actin protein and mRNA concentrations increase by about 50%, whereas tubulin reaches maxima of 100% increase. The proportion between globular and filamentous actin remains constant after hormone treatment.
Studies were conducted to determine the effects of delaying the separation of serum from the clot and of long‐term storage of serum samples on the measurement of thyroid hormones in blood from horses ...using a fluorescence polarization immunoassay. The measured concentrations of T3 and T4 were not affected by leaving serum on the clot for as long as 24 hours at room temperatures. Storage of serum for 19 to 22 months at ‐20°C resulted in significant increases of measured T4, but not T3. These studies support previous work demonstrating that thyroid hormones are resistant to degradation, immunologically stable, and reasonably insensitive to potential problems of routine specimen handling when measured with an immunoassay.

Wir berichten über den Fall einer 41-jährigen Patientin, welche sich 3 Jahre nach einer Hämorrhoidenoperation nach Longo mit rezidivierenden Unterbauchschmerzen in der zentralen ...Notaufnahme unseres Spitals vorstellte. Klinisch wurde eine Raumforderung im Septum rectovaginalis palpiert und magnetresonanztomographisch ergab sich ein stuhlgefülltes Rektumdivertikel. Über einen perinealen Zugang konnte das Divertikel an der Basis abgetragen und der Mukosadefekt transanal mittels Naht verschlossen werden. Ein Rektumdivertikel ist eine seltene Komplikation (2,5%) nach Stapler-Hämorrhoidopexie
