The function of tropical peatland as a carbon sink is a balance between peat accumulation and peat loss; however, various interacting factors are involved affecting this process. In this study, we ...collected and intensively radiocarbon dated peat cores from two peat domes, visualized their cross-sectional profiles of geochemical properties, and developed three macrocharcoal records from each peat dome. We find that the young (4500 y calBP) and shallow (6 m) coastal peat has a simple and linear age–depth relationship, showing stable accumulation of carbon during the late Holocene. In contrast, the older (ca. 40,000 y cal BP) and deeper (15 m) inland peat shows a more complex history, where we observed age reversals and hiatuses, likely caused by climate variability from the Last Glacial Maximum (LGM) to the Holocene. The charcoal record reveals a continuous presence of low-severity fire as indicated by charcoal morphotypes, though fire frequency increased after agriculture was established. An age reversal during the LGM was likely caused by a flood. Two periods of hiatuses occurred, each several millennia in length, at the end of the LGM and during the early Holocene. One cause of the hiatuses may have been a climatically halted peat formation from low precipitation and cooler climate during the LGM. Another cause may have been that severe fires consumed thousands of years of accumulated peat. If the hiatuses were entirely due to fire, the carbon released from these paleo-fire events (600 t C ha
−1
) suggests several times the impact of the most intense modern peat fires.

Understanding the causes and consequences of wildfires in forests of the western United States requires integrated information about fire, climate changes, and human activity on multiple temporal ...scales. We use sedimentary charcoal accumulation rates to construct long-term variations in fire during the past 3,000 y in the American West and compare this record to independent fire-history data from historical records and fire scars. There has been a slight decline in burning over the past 3,000 y, with the lowest levels attained during the 20th century and during the Little Ice Age (LIA, ca. 1400–1700 CE Common Era). Prominent peaks in forest fires occurred during the Medieval Climate Anomaly (ca. 950–1250 CE) and during the 1800s. Analysis of climate reconstructions beginning from 500 CE and population data show that temperature and drought predict changes in biomass burning up to the late 1800s CE. Since the late 1800s , human activities and the ecological effects of recent high fire activity caused a large, abrupt decline in burning similar to the LIA fire decline. Consequently, there is now a forest "fire deficit" in the western United States attributable to the combined effects of human activities, ecological, and climate changes. Large fires in the late 20th and 21st century fires have begun to address the fire deficit, but it is continuing to grow.
Malaria vaccine design and prioritization has been hindered by the lack of a mechanistic correlate of protection. We previously demonstrated a strong association between protection and ...merozoite-neutralizing antibody responses following vaccination of non-human primates against Plasmodium falciparum reticulocyte binding protein homolog 5 (PfRH5). Here, we test the mechanism of protection. Using mutant human IgG1 Fc regions engineered not to engage complement or FcR-dependent effector mechanisms, we produce merozoite-neutralizing and non-neutralizing anti-PfRH5 chimeric monoclonal antibodies (mAbs) and perform a passive transfer-P. falciparum challenge study in Aotus nancymaae monkeys. At the highest dose tested, 6/6 animals given the neutralizing PfRH5-binding mAb c2AC7 survive the challenge without treatment, compared to 0/6 animals given non-neutralizing PfRH5-binding mAb c4BA7 and 0/6 animals given an isotype control mAb. Our results address the controversy regarding whether merozoite-neutralizing antibody can cause protection against P. falciparum blood-stage infections, and highlight the quantitative challenge of achieving such protection.
Detecting latitudinal range shifts of forest trees in response to recent climate change is difficult because of slow demographic rates and limited dispersal but may be facilitated by spatially ...compressed climatic zones along elevation gradients in montane environments. We resurveyed forest plots established in 1964 along elevation transects in the Green Mountains (Vermont) to examine whether a shift had occurred in the location of the northern hardwood-boreal forest ecotone (NBE) from 1964 to 2004. We found a 19% increase in dominance of northern hardwoods from 70% in 1964 to 89% in 2004 in the lower half of the NBE. This shift was driven by a decrease (up to 76%) in boreal and increase (up to 16%) in northern hardwood basal area within the lower portions of the ecotone. We used aerial photographs and satellite imagery to estimate a 91- to 119-m upslope shift in the upper limits of the NBE from 1962 to 2005. The upward shift is consistent with regional climatic change during the same period; interpolating climate data to the NBE showed a 1.1°C increase in annual temperature, which would predict a 208-m upslope movement of the ecotone, along with a 34% increase in precipitation. The rapid upward movement of the NBE indicates little inertia to climatically induced range shifts in montane forests; the upslope shift may have been accelerated by high turnover in canopy trees that provided opportunities for ingrowth of lower elevation species. Our results indicate that high-elevation forests may be jeopardized by climate change sooner than anticipated.
Fire is a primary mode of natural disturbance in the forests of the Pacific Northwest. Increased fuel loads following fire suppression and the occurrence of several large and severe fires have led to ...the perception that in many areas there is a greatly increased risk of high-severity fire compared with presettlement forests. To reconstruct the variability of the fire regime in the Siskiyou Mountains, Oregon, we analyzed a 10-m, 2,000-y sediment core for charcoal, pollen, and sedimentological data. The record reveals a highly episodic pattern of fire in which 77% of the 68 charcoal peaks before Euro-American settlement cluster within nine distinct periods marked by a 15-y mean interval. The 11 largest charcoal peaks are significantly related to decadal-scale drought periods and are followed by pulses of minerogenic sediment suggestive of rapid sediment delivery. After logging in the 1950s, sediment load was increased fourfold compared with that from the most severe presettlement fire. Less severe fires, marked by smaller charcoal peaks and no sediment pulses, are not correlated significantly with drought periods. Pollen indicators of closed forests are consistent with fire-free periods of sufficient length to maintain dense forest and indicate a fire-triggered switch to more open conditions during the Medieval Climatic Anomaly. Our results indicate that over millennia fire was more episodic than revealed by nearby shorter tree-ring records and that recent severe fires have precedents during earlier drought episodes but also that sediment loads resulting from logging and road building have no precedent in earlier fire events.
Extreme climatic events are key factors in initiating gradual or sudden changes in forest ecosystems through the promotion of severe, tree-killing disturbances such as fire, blowdown, and widespread ...insect outbreaks. In contrast to these climatically-incited disturbances, little is known about the more direct effect of drought on tree mortality, especially in high-elevation forests. Therefore projections of drought-induced mortality under future climatic conditions remain uncertain. For a subalpine forest landscape in the Rocky Mountains of northern Colorado (USA), we quantified lag effects of drought on mortality of Engelmann spruce Picea engelmannii, subalpine fir Abies lasiocarpa, and lodgepole pine Pinus contorta. For the period 1910-2004, we related death dates of 164 crossdated dead trees to early-season and late-season droughts. Following early-season droughts, spruce mortality increased over five years and fir mortality increased sharply over 11 years. Following late-season droughts, spruce showed a small increase in mortality within one year, whereas fir showed a consistent period of increased mortality over two years. Pine mortality was not affected by drought. Low pre-drought radial growth rates predisposed spruce and fir to drought-related mortality. Spruce and fir trees that died during a recent drought (2000-2004) had significantly lower pre-drought growth rates than live neighbour trees. Overall, we found large interspecific differences in drought-related mortality with fir showing the strongest effect followed by spruce and pine. This direct influence of climatic variability on differential tree mortality has the potential for driving large-scale changes in subalpine forests of the Rocky Mountains.
There is intense interest in induction and characterization of strain-transcending neutralizing Ab against antigenically variable human pathogens. We have recently identified the human malaria ...parasite Plasmodium falciparum reticulocyte-binding protein homolog 5 (PfRH5) as a target of broadly neutralizing Abs, but there is little information regarding the functional mechanism(s) of Ab-mediated neutralization. In this study, we report that vaccine-induced polyclonal anti-PfRH5 Abs inhibit the tight attachment of merozoites to erythrocytes and are capable of blocking the interaction of PfRH5 with its receptor basigin. Furthermore, by developing anti-PfRH5 mAbs, we provide evidence of the following: 1) the ability to block the PfRH5-basigin interaction in vitro is predictive of functional activity, but absence of blockade does not predict absence of functional activity; 2) neutralizing mAbs bind spatially related epitopes on the folded protein, involving at least two defined regions of the PfRH5 primary sequence; 3) a brief exposure window of PfRH5 is likely to necessitate rapid binding of Ab to neutralize parasites; and 4) intact bivalent IgG contributes to but is not necessary for parasite neutralization. These data provide important insight into the mechanisms of broadly neutralizing anti-malaria Abs and further encourage anti-PfRH5-based malaria prevention efforts.
Tropical peatlands in Southeast Asia cover ∼25 million hectares and exert a strong influence on the global carbon cycle. Recent widespread peatland subsidence and carbon dioxide emissions in response ...to human activity and climate change have been well documented, but peatland genesis remains poorly understood. Unlike coastal peatlands that established following sea-level stabilization during the mid-Holocene, inland peatlands of Borneo are little studied and have no apparent environmental constraint on their formation. Here, we report radiocarbon dates from the Upper Kapuas Basin which show inland peat formation since at least 47.8 thousand calibrated radiocarbon years before present, ka. We provide a synthesis of new and existing peat basal dates across Borneo, which shows a hiatus in peat genesis during a cool and dry period from 30 to 20 ka. Despite likely peat degradation during that period, the Upper Kapuas is still exceptionally deep, reaching a maximum depth (determined from coring) of 18 m. Our best estimate of mean peat depth over 3833 km2 of the Upper Kapuas is 5.16 ± 2.66 m, corresponding to a carbon density of 2790 ± 1440 Mg C ha−1. This is one of the most carbon-dense ecosystems in the world. It withstood the glacial-interglacial climate transition and remains mostly intact, but is increasingly threatened by land-use change.
Amazonian Dark Earths (ADEs) are unusually fertile soils characterised by elevated concentrations of microscopic charcoal particles, which confer their distinctive colouration. Frequent occurrences ...of pre-Columbian artefacts at ADE sites led to their ubiquitous classification as Anthrosols (soils of anthropic origin). However, it remains unclear how indigenous peoples created areas of high fertility in one of the most nutrient-impoverished environments on Earth. Here, we report new data from a well-studied ADE site in the Brazilian Amazon, which compel us to reconsider its anthropic origin. The amounts of phosphorus and calcium-two of the least abundant macronutrients in the region-are orders of magnitude higher in ADE profiles than in the surrounding soil. The elevated levels of phosphorus and calcium, which are often interpreted as evidence of human activity at other sites, correlate spatially with trace elements that indicate exogenous mineral sources rather than in situ deposition. Stable isotope ratios of neodymium, strontium, and radiocarbon activity of microcharcoal particles also indicate exogenous inputs from alluvial deposition of carbon and mineral elements to ADE profiles, beginning several thousands of years before the earliest evidence of soil management for plant cultivation in the region. Our data suggest that indigenous peoples harnessed natural processes of landscape formation, which led to the unique properties of ADEs, but were not responsible for their genesis. If corroborated elsewhere, this hypothesis would transform our understanding of human influence in Amazonia, opening new frontiers for the sustainable use of tropical landscapes going forward.
Despite its perceived historical rarity, fire is an important disturbance in tropical rainforests. Very large rainforest fires have been observed multiple times in recent decades, often during years ...of strong El Niño‐Southern Oscillation droughts. Fire in rainforest has major short‐term consequences for humans and wildlife by converting forest to fire‐prone fern, shrub, and grass, but the long‐term effects remain to be seen. Borneo's indigenous groups have been using fire to clear land for centuries, yet the prevalence and spatial patterns of premodern fire across forest types in Borneo are not well understood. This research set out to reconstruct fire in a 1500‐ha primary rainforest spanning 800 m of elevation in Indonesian Borneo with the goal of elucidating the role humans have played in rainforest fire. We found that humans played an important role in the occurrence of fire in recent centuries. Evidence of fire—charcoal >2 mm—is more abundant in forest types where humans would be more likely to live and/or practice swidden agriculture. However, pyrogenic material is ubiquitous across the study area, showing that all forest types have experienced fire. A set of 50 radiocarbon dates showed that in lowland areas—where human‐caused fire is most likely—fire occurred throughout the last 3200 years, peaking 1300–1600 ce. The upland areas lacked evidence of fire before 1250 ce but otherwise had a similar pattern to the lowlands. The period of high fire coincides with regional demographic changes as well as regional droughts documented elsewhere in Southeast Asia. In upland areas, fires likely burned only under regional drought when fires could more easily spread upslope. Although forest plot studies at this site show little structural evidence of past fires, tree diversity is lower than expected in the most burned areas (alluvial benches). Thus, our results suggest that land clearance was a major source of fire, but the current intact state of these rainforests indicates that they were largely resilient to fires and land use hundreds of years ago. Recent fires mirror patterns of fire spread that occurred hundreds of years ago, though their severity and extent are likely much greater.
