Nuage are amorphous ultrastructural granules in the cytoplasm of male germ cells as divergent as Drosophila, Xenopus, and Homo sapiens. Most nuage are cytoplasmic ribonucleoprotein structures ...implicated in diverse RNA metabolism including the regulation of PIWI-interacting RNA (piRNA) synthesis by the PIWI family (i.e., MILI, MIWI2, and MIWI). MILI is prominent in embryonic and early post-natal germ cells in nuage also called germinal granules that are often associated with mitochondria and called intermitochondrial cement. We find that GASZ (Germ cell protein with Ankyrin repeats, Sterile alpha motif, and leucine Zipper) co-localizes with MILI in intermitochondrial cement. Knockout of Gasz in mice results in a dramatic downregulation of MILI, and phenocopies the zygotene-pachytene spermatocyte block and male sterility defect observed in MILI null mice. In Gasz null testes, we observe increased hypomethylation and expression of retrotransposons similar to MILI null testes. We also find global shifts in the small RNAome, including down-regulation of repeat-associated, known, and novel piRNAs. These studies provide the first evidence for an essential structural role for GASZ in male fertility and epigenetic and post-transcriptional silencing of retrotransposons by stabilizing MILI in nuage.

Stable intercellular bridges are a conserved feature of gametogenesis in multicellular animals observed more than 100 years ago, but their function was unknown. Many of the components necessary for ...this structure have been identified through the study of cytokinesis in Drosophila; however, mammalian intercellular bridges have distinct properties from those of insects. Mammalian germ cell intercellular bridges are composed of general cytokinesis components with additional germ cell-specific factors including TEX14. TEX14 is an inactive kinase essential for the maintenance of stable intercellular bridges in gametes of both sexes but whose loss specifically impairs male meiosis. TEX14 acts to impede the terminal steps of abscission by competing for essential component CEP55, blocking its interaction in nongerm cells with ALIX and TSG101. Additionally, TEX14-interacting protein RBM44, whose localization in stabile intercellular bridges is limited to pachytene and secondary spermatocytes, may participate in processes such as RNA transport but is nonessential to the maintenance of intercellular bridge stability.
We report the results of a ∼4 yr direct imaging survey of 104 stars to resolve and characterize circumstellar debris disks in scattered light as part of the Gemini Planet Imager (GPI) Exoplanet ...Survey. We targeted nearby ( 150 pc), young ( 500 Myr) stars with high infrared (IR) excesses (LIR/L > 10−5), including 38 with previously resolved disks. Observations were made using the GPI high-contrast integral field spectrograph in H-band (1.6 m) coronagraphic polarimetry mode to measure both polarized and total intensities. We resolved 26 debris disks and 3 protoplanetary/transitional disks. Seven debris disks were resolved in scattered light for the first time, including newly presented HD 117214 and HD 156623, and we quantified basic morphologies of five of them using radiative transfer models. All of our detected debris disks except HD 156623 have dust-poor inner holes, and their scattered-light radii are generally larger than corresponding radii measured from resolved thermal emission and those inferred from spectral energy distributions. To assess sensitivity, we report contrasts and consider causes of nondetections. Detections were strongly correlated with high IR excess and high inclination, although polarimetry outperformed total intensity angular differential imaging for detecting low-inclination disks ( 70°). Based on postsurvey statistics, we improved upon our presurvey target prioritization metric predicting polarimetric disk detectability. We also examined scattered-light disks in the contexts of gas, far-IR, and millimeter detections. Comparing H-band and ALMA fluxes for two disks revealed tentative evidence for differing grain properties. Finally, we found no preference for debris disks to be detected in scattered light if wide-separation substellar companions were present.
The HR 8799 system uniquely harbors four young super-Jupiters whose orbits can provide insights into the system's dynamical history and constrain the masses of the planets themselves. Using the ...Gemini Planet Imager, we obtained down to one milliarcsecond precision on the astrometry of these planets. We assessed four-planet orbit models with different levels of constraints and found that assuming the planets are near 1:2:4:8 period commensurabilities, or are coplanar, does not worsen the fit. We added the prior that the planets must have been stable for the age of the system (40 Myr) by running orbit configurations from our posteriors through N-body simulations and varying the masses of the planets. We found that only assuming the planets are both coplanar and near 1:2:4:8 period commensurabilities produces dynamically stable orbits in large quantities. Our posterior of stable coplanar orbits tightly constrains the planets' orbits, and we discuss implications for the outermost planet b shaping the debris disk. A four-planet resonance lock is not necessary for stability up to now. However, planet pairs d and e, and c and d, are each likely locked in two-body resonances for stability if their component masses are above 6 MJup and 7 MJup, respectively. Combining the dynamical and luminosity constraints on the masses using hot-start evolutionary models and a system age of 42 5 Myr, we found the mass of planet b to be 5.8 0.5 MJup, and the masses of planets c, d, and e to be each.
We present a new matched-filter algorithm for direct detection of point sources in the immediate vicinity of bright stars. The stellar point-spread function (PSF) is first subtracted using a ...Karhunen-Loéve image processing (KLIP) algorithm with angular and spectral differential imaging (ADI and SDI). The KLIP-induced distortion of the astrophysical signal is included in the matched-filter template by computing a forward model of the PSF at every position in the image. To optimize the performance of the algorithm, we conduct extensive planet injection and recovery tests and tune the exoplanet spectra template and KLIP reduction aggressiveness to maximize the signal-to-noise ratio (S/N) of the recovered planets. We show that only two spectral templates are necessary to recover any young Jovian exoplanets with minimal S/N loss. We also developed a complete pipeline for the automated detection of point-source candidates, the calculation of receiver operating characteristics (ROC), contrast curves based on false positives, and completeness contours. We process in a uniform manner more than 330 data sets from the Gemini Planet Imager Exoplanet Survey and assess GPI typical sensitivity as a function of the star and the hypothetical companion spectral type. This work allows for the first time a comparison of different detection algorithms at a survey scale accounting for both planet completeness and false-positive rate. We show that the new forward model matched filter allows the detection of 50% fainter objects than a conventional cross-correlation technique with a Gaussian PSF template for the same false-positive rate.
Cytokinesis in somatic cells concludes with the formation of a midbody, which is abscised to form individual daughter cells. In contrast, germ cell cytokinesis results in a permanent intercellular ...bridge connecting the daughter cells through a large cytoplasmic channel. During spermatogenesis, proposed roles for the intercellular bridge include germ cell communication, synchronization, and chromosome dosage compensation in haploid cells. Although several essential components of the midbody have recently been identified, essential components of the vertebrate germ cell intercellular bridge have until now not been described. Herein, we show that testis-expressed gene 14 (TEX14) is a novel protein that localizes to germ cell intercellular bridges. In the absence of TEX14, intercellular bridges are not observed by using electron microscopy and other markers. Spermatogenesis in$Tex14^{-/-}$mice progresses through the transit amplification of diploid spermatogonia and the expression of early meiotic markers but halts before the completion of the first meiotic division. Thus, TEX14 is required for intercellular bridges in vertebrate germ cells, and these studies provide evidence that the intercellular bridge is essential for spermatogenesis and fertility.

and show algorithm stability with varying KLIP parameters. We report ...new and re-reduced spectrophotometry of HR 8799 c, d, and e in the
H
and
K
bands. We discuss a strategy for choosing optimal KLIP PSF subtraction parameters by injecting simulated sources and recovering them over a range of parameters. The
K1
/
K2
spectra for HR 8799 c and d are similar to previously published results from the same data set. We also present a
K
-band spectrum of HR 8799 e for the first time and show that our
H
-band spectra agree well with previously published spectra from the VLT/SPHERE instrument. We show that HR 8799 c and d show significant differences in their
H
and
K
spectra, but do not find any conclusive differences between d and e, nor between c and e, likely due to large error bars in the recovered spectrum of e. Compared to M-, L-, and T-type field brown dwarfs, all three planets are most consistent with mid- and late-L spectral types. All objects are consistent with low gravity, but a lack of standard spectra for low gravity limit the ability to fit the best spectral type. We discuss how dedicated modeling efforts can better fit HR 8799 planets’ near-IR flux, as well as how differences between the properties of these planets can be further explored.
We present new observations of the planet β Pictoris b from 2018 with the Gemini Planet Imager (GPI), the first GPI observations following conjunction. Based on these new measurements, we perform a ...joint orbit fit to the available relative astrometry from ground-based imaging, the Hipparcos Intermediate Astrometric Data (IAD), and the Gaia DR2 position, and demonstrate how to incorporate the IAD into direct imaging orbit fits. We find a mass consistent with predictions of hot-start evolutionary models and previous works following similar methods, though with larger uncertainties: 12.8+5.3−3.2 MJup. Our eccentricity determination of disfavors circular orbits. We consider orbit fits to several different imaging data sets, and find generally similar posteriors on the mass for each combination of imaging data. Our analysis underscores the importance of performing joint fits to the absolute and relative astrometry simultaneously, given the strong covariance between orbital elements. Time of conjunction is well-constrained within 2.8 days of 2017 September 13, with the star behind the planet's Hill sphere between 2017 April 11 and 2018 February 16 ( 18 days). Following the recent radial velocity detection of a second planet in the system, β Pic c, we perform additional two-planet fits combining relative astrometry, absolute astrometry, and stellar radial velocities. These joint fits find a significantly smaller mass (8.0 2.6 MJup) for the imaged planet β Pic b, in a somewhat more circular orbit. We expect future ground-based observations to further constrain the visual orbit and mass of the planet in advance of the release of Gaia DR4.
We present new H-band scattered light images of the HD 32297 edge-on debris disk obtained with the Gemini Planet Imager. The disk is detected in total and polarized intensity down to a projected ...angular separation of 0 15, or 20 au. On the other hand, the large-scale swept-back halo remains undetected, likely a consequence of its markedly blue color relative to the parent body belt. We analyze the curvature of the disk spine and estimate a radius of 100 au for the parent body belt, smaller than past scattered light studies but consistent with thermal emission maps of the system. We employ three different flux-preserving post-processing methods to suppress the residual starlight and evaluate the surface brightness and polarization profile along the disk spine. Unlike past studies of the system, our high-fidelity images reveal the disk to be highly symmetric and devoid of morphological and surface brightness perturbations. We find the dust scattering properties of the system to be consistent with those observed in other debris disks, with the exception of HR 4796. Finally, we find no direct evidence for the presence of a planetary-mass object in the system.
