Abstract
Purpose. Besides basic measurements as maximum standardized uptake value (SUV)max or SUVmean derived from 18F-FDG positron emission tomography (PET) scans, more advanced quantitative imaging ...features (i.e. "Radiomics" features) are increasingly investigated for treatment monitoring, outcome prediction, or as potential biomarkers. With these prospected applications of Radiomics features, it is a requisite that they provide robust and reliable measurements. The aim of our study was therefore to perform an integrated stability analysis of a large number of PET-derived features in non-small cell lung carcinoma (NSCLC), based on both a test-retest and an inter-observer setup. Methods. Eleven NSCLC patients were included in the test-retest cohort. Patients underwent repeated PET imaging within a one day interval, before any treatment was delivered. Lesions were delineated by applying a threshold of 50% of the maximum uptake value within the tumor. Twenty-three NSCLC patients were included in the inter-observer cohort. Patients underwent a diagnostic whole body PET-computed tomography (CT). Lesions were manually delineated based on fused PET-CT, using a standardized clinical delineation protocol. Delineation was performed independently by five observers, blinded to each other. Fifteen first order statistics, 39 descriptors of intensity volume histograms, eight geometric features and 44 textural features were extracted. For every feature, test-retest and inter-observer stability was assessed with the intra-class correlation coefficient (ICC) and the coefficient of variability, normalized to mean and range. Similarity between test-retest and inter-observer stability rankings of features was assessed with Spearman's rank correlation coefficient. Results. Results showed that the majority of assessed features had both a high test-retest (71%) and inter-observer (91%) stability in terms of their ICC. Overall, features more stable in repeated PET imaging were also found to be more robust against inter-observer variability. Conclusion. Results suggest that further research of quantitative imaging features is warranted with respect to more advanced applications of PET imaging as being used for treatment monitoring, outcome prediction or imaging biomarkers.
We present cosmological parameter constraints from a tomographic weak gravitational lensing analysis of ~450 deg super( 2) of imaging data from the Kilo Degree Survey (KiDS). For a flat ... cold dark ...matter (...CDM) cosmology with a prior on H sub( 0) that encompasses the most recent direct measurements, we find S sub( 8) ... = 0.745 plus or minus 0.039. This result is in good agreement with other low-redshift probes of large-scale structure, including recent cosmic shear results, along with pre-Planck cosmic microwave background constraints. A 2.3... tension in S sub( 8) and 'substantial discordance' in the full parameter space is found with respect to the Planck 2015 results. We use shear measurements for nearly 15 million galaxies, determined with a new improved 'self-calibrating' version of lensfit validated using an extensive suite of image simulations. Four-band ugri photometric redshifts are calibrated directly with deep spectroscopic surveys. The redshift calibration is confirmed using two independent techniques based on angular cross-correlations and the properties of the photometric redshift probability distributions. Our covariance matrix is determined using an analytical approach, verified numerically with large mock galaxy catalogues. We account for uncertainties in the modelling of intrinsic galaxy alignments and the impact of baryon feedback on the shape of the non-linear matter power spectrum, in addition to the small residual uncertainties in the shear and redshift calibration. The cosmology analysis was performed blind. PD-L1 immunohistochemistry correlates only moderately with patient survival and response to PD-(L)1 treatment. Heterogeneity of tumor PD-L1 expression might limit the predictive value of small ...biopsies. Here we show that tumor PD-L1 and PD-1 expression can be quantified non-invasively using PET-CT in patients with non-small-cell lung cancer. Whole body PD-(L)1 PET-CT reveals significant tumor tracer uptake heterogeneity both between patients, as well as within patients between different tumor lesions.
Five-year survival after resection of colorectal cancer liver metastasis (CRLCM) is <30%. We recently found that aurora kinase A (AURKA) drives 20q gain-associated tumour progression and is ...associated with disease recurrence. This study evaluates the prognostic value of AURKA expression in CRCLM of patients who underwent liver resection.
Tissue microarrays (TMAs) were generated using formalin-fixed paraffin-embedded CRCLM and matched primary tumour from a multi-institutional cohort of patients with CRCLM who underwent liver resection between 1990 and 2010. Tissue microarrays were stained for AURKA using immunohistochemistry, and a hazard rate ratio (HRR) for the association between overall survival (OS) and nuclear AURKA expression in CRCLM was calculated. Results were validated by 500-fold cross-validation.
The expression of AURKA was evaluated in CRCLM of 343 patients. High AURKA expression was associated with poor OS (HRR 1.55, P<0.01), with a cross-validated average HRR of 1.57 (P=0.02). Average HRR was adjusted for the established prognostic clinicopathological variables in a multivariate analysis (average HRR 1.66; P=0.02). The expression of AURKA in CRCLM was correlated to its expression in corresponding primary tumour (P<0.01).
The expression of AURKA protein is a molecular biomarker with prognostic value for patients with CRCLM, independent of established clinicopathological variables.
Virtual water is the volume of water used to produce a commodity or service. Hitherto, most virtual water ‘trade’ studies have focused on its potential contribution to saving water, especially in ...water short regions. Very little, however, has been said about the opportunity cost of the associated water. The present research critically evaluates the strategic importance of green water (soil water originating from rainfall) in relation to international commodity trade. Besides having a lower opportunity cost, the use of green water for the production of crops has generally less negative environmental externalities than the use of blue water (irrigation with water abstracted from ground or surface water systems). Although it is widely known that major grain exporters – the USA, Canada, France, Australia and Argentina – produce grain in highly productive rain-fed conditions, green water volumes in exports have rarely been estimated. The present study corroborates that green water is by far the largest share of virtual water in maize, soybean and wheat exports from its main exporting countries (USA, Canada, Australia and Argentina) during the period 2000–2004. Insofar virtual water is ‘traded’ towards water-scarce nations that heavily depend on their blue water resources, green virtual-water ‘trade’ related to these commodities plays a role in ensuring water and water-dependent food security and avoiding further potential damage to the water environments in both importing and exporting countries. This potential of international green virtual-water ‘trade’, however, is constrained by factors such as technology, the potential for further increases in the productivity of soil and irrigation water, the level of socio-economic development, national food policies and international trade agreements.

Hyposplenic patients are at risk of overwhelming post-splenectomy infection (OPSI), which carries mortality of up to 70%. Therefore, preventive measures are warranted. However, patients with ...diminished splenic function are difficult to identify. In this review we discuss immunological, haematological and scintigraphic parameters that can be used to measure splenic function. IgM memory B cells are a potential parameter for assessing splenic function; however, more studies are necessary for its validation. Detection of Howell–Jolly bodies does not reflect splenic function accurately, whereas determining the percentage of pitted erythrocytes is a well-evaluated method and seems a good first-line investigation for assessing splenic function. Tc-labelled, heat-altered, autologous erythrocyte scintigraphy with multimodality single photon emission computed tomography (SPECT)-CT technology is the best approach, as all facets of splenic function are evaluated. In conclusion, although scintigraphic methods are most reliable, they are not suitable for screening large populations. We therefore recommend using the percentage of pitted erythrocytes, albeit suboptimal, as a first-line investigation and subsequently confirming abnormal readings by means of scintigraphy. More studies evaluating the value of potentially new markers are needed.
The metabolic syndrome (MS) might increase the risk of cardiovascular disease in testicular cancer (TC) survivors. We investigated its prevalence, development, vascular implications, and the role of ...gonadal function.
TC survivors treated with chemotherapy and follow-up ≥3 years (N = 370, study I) were retrospectively evaluated for the development of cardiovascular risk factors. A subgroup followed 3–20 years (N = 173, study II) was compared with controls (N = 1085) for MS prevalence and evaluated for vascular function.
In TC survivors (study I), 24% developed overweight, 24% hypercholesterolemia, and 30% hypertension, after median follow-up of 1.7, 0.9, and 5.1 years, respectively. At the median follow-up of 5 years (study II), 25% of survivors have the MS {odds ratio (OR) 2.2, 95% confidence interval (CI) 1.5–3.3 compared with controls}. Survivors with MS have features of inflammation and prothrombotic state, increased carotid artery intima-media thickness. Survivors with testosterone levels <15 nmol/l (22%) have an increased risk of the MS (OR 4.1, 95% CI 1.8–9.3).
The current data suggest that the MS occurs at earlier age in TC survivors treated with chemotherapy compared with controls and is accompanied by early signs of atherosclerosis. As low testosterone may have a causal role, it is a target for interventions.
Extreme ultraviolet (EUV) lithography is currently entering high-volume manufacturing to enable the continued miniaturization of semiconductor devices. The required EUV light, at 13.5 nm wavelength, ...is produced in a hot and dense laser-driven tin plasma. The atomic origins of this light are demonstrably poorly understood. Here we calculate detailed tin opacity spectra using the Los Alamos atomic physics suite ATOMIC and validate these calculations with experimental comparisons. Our key finding is that EUV light largely originates from transitions between multiply-excited states, and not from the singly-excited states decaying to the ground state as is the current paradigm. Moreover, we find that transitions between these multiply-excited states also contribute in the same narrow window around 13.5 nm as those originating from singly-excited states, and this striking property holds over a wide range of charge states. We thus reveal the doubly magic behavior of tin and the origins of the EUV light.

. This result is insensitive to changes in the priors on nuisance parameters for intrinsic alignment, baryon feedback, and neutrino mass. KiDS shear measurements are calibrated with a new, more realistic set of image simulations and no significant B-modes are detected in the survey, indicating that systematic errors are under control. When calibrating our redshift distributions by assuming the 30-band COSMOS-2015 photometric redshifts are correct (following the Dark Energy Survey and the Hyper Suprime-Cam Survey), we find the tension with
Planck
is alleviated. The robust determination of source redshift distributions remains one of the most challenging aspects for future cosmic shear surveys.
BACKGROUND AND AIMS: We investigated whether spatial differences in belowground niches between shallow- and deep-rooted species result in increased resource uptake and drought resistance in ...agricultural grassland communities. MATERIAL AND METHODS: We injected the tracers ¹⁵N and rubidium (Rb) into the soil and measured tracer uptake from 5 and 35 cm soil depth in monocultures and mixtures containing shallow-rooted (Lolium perenne, non-legume; Trifolium repens, legume) and deep-rooted (Cichorium intybus, non-legume; Trifolium pratense, legume) species, grown under drought or control conditions. RESULTS: Deep-rooted species had a higher proportional uptake of both tracers from 35 cm soil depth than shallow-rooted species (p < 0.001). Under drought conditions, all species enhanced their proportional tracer uptake from the deeper soil layer (p < 0.004) and species with a high proportional ¹⁵N uptake from the deeper soil layer were more drought resistant. Total community uptake of Rb was significantly higher in mixed communities than monocultures (p < 0.007); however, this diversity effect was the result of mixing legumes with non-legumes and not the result of mixing deep- with shallow-rooted species. CONCLUSIONS: Clear evidence for spatial niche differences in resource uptake between shallow- and deep-rooted species did not translate into increased resource uptake in mixtures. Resource uptake from deeper soil layers was found to contribute to adaptation and resistance to drought stress.
