of solids and is significantly enriched in metals relative to its solar-metallicity host star. We use these new constraints to explore the broader question of how systems like Kepler-167 form in the pebble accretion framework for giant planet core formation. We utilize simple disk evolution models to demonstrate that more massive and metal-rich disks, which are the most favorable sites for giant planet formation, can also deliver enough solids to the inner disk to form systems of super-Earths. We use these same models to constrain the nature of Kepler-167's protoplanetary disk and find that it likely contained ≳300
M
⊕
of dust and was ≳40 au in size. These values overlap with the upper end of the observed dust mass and size distributions of Class 0 and I disks and are also consistent with the observed occurrence rate of Jupiter analogs around Sun-like stars.

Proteins are not equally digestible-their proteolytic susceptibility varies by their source and processing method. Incomplete digestion increases colonic microbial protein fermentation ...(putrefaction), which produces toxic metabolites that can induce inflammation in vitro and have been associated with inflammation in vivo. Individual humans differ in protein digestive capacity based on phenotypes, particularly disease states. To avoid putrefaction-induced intestinal inflammation, protein sources, and processing methods must be tailored to the consumer's digestive capacity. This review explores how food processing techniques alter protein digestibility and examines how physiological conditions alter digestive capacity. Possible solutions to improving digestive function or matching low digestive capacity with more digestible protein sources are explored. Beyond the ileal digestibility measurements of protein digestibility, less invasive, quicker and cheaper techniques for monitoring the extent of protein digestion and fermentation are needed to personalize protein nourishment. Biomarkers of protein digestive capacity and efficiency can be identified with the toolsets of peptidomics, metabolomics, microbial sequencing and multiplexed protein analysis of fecal and urine samples. By monitoring individual protein digestive function, the protein component of diets can be tailored via protein source and processing selection to match individual needs to minimize colonic putrefaction and, thus, optimize gut health.

The alignment of planetary orbits with respect to the stellar rotation preserves information on their dynamical histories. Measuring this angle for young planets helps illuminate the ...mechanisms that create misaligned orbits for older planets, as different processes could operate over timescales ranging from a few megayears to a gigayear. We present spectroscopic transit observations of the young exoplanet V1298 Tau b; we update the age of V1298 Tau to be 28 ± 4 Myr based on Gaia EDR3 measurements. We observed a partial transit with Keck/HIRES and LBT/PEPSI, and detected the radial velocity anomaly due to the Rossiter–McLaughlin effect. V1298 Tau b has a prograde, well-aligned orbit, with

Alendronate and raloxifene are among the most popular anti-osteoporosis medications. However, there is a lack of head-to-head comparative effectiveness studies comparing the two treatments. We ...conducted a retrospective large-scale multicenter study encompassing over 300 million patients across nine databases encoded in the Observational Medical Outcomes Partnership (OMOP) Common Data Model (CDM). The primary outcome was the incidence of osteoporotic hip fracture, while secondary outcomes were vertebral fracture, atypical femoral fracture (AFF), osteonecrosis of the jaw (ONJ), and esophageal cancer. We used propensity score trimming and stratification based on an expansive propensity score model with all pre-treatment patient characteritistcs. We accounted for unmeasured confounding using negative control outcomes to estimate and adjust for residual systematic bias in each data source. We identified 283,586 alendronate patients and 40,463 raloxifene patients. There were 7.48 hip fracture, 8.18 vertebral fracture, 1.14 AFF, 0.21 esophageal cancer and 0.09 ONJ events per 1,000 person-years in the alendronate cohort and 6.62, 7.36, 0.69, 0.22 and 0.06 events per 1,000 person-years, respectively, in the raloxifene cohort. Alendronate and raloxifene have a similar hip fracture risk (hazard ratio HR 1.03, 95% confidence interval CI 0.94-1.13), but alendronate users are more likely to have vertebral fractures (HR 1.07, 95% CI 1.01-1.14). Alendronate has higher risk for AFF (HR 1.51, 95% CI 1.23-1.84) but similar risk for esophageal cancer (HR 0.95, 95% CI 0.53-1.70), and ONJ (HR 1.62, 95% CI 0.78-3.34). We demonstrated substantial control of measured confounding by propensity score adjustment, and minimal residual systematic bias through negative control experiments, lending credibility to our effect estimates. Raloxifene is as effective as alendronate and may remain an option in the prevention of osteoporotic fracture.
Burkholderia pseudomallei is the causative agent of melioidosis and a potential bioterrorism agent. In the development of medical countermeasures against B. pseudomallei infection, the US Food and ...Drug Administration (FDA) animal Rule recommends using well-characterized strains in animal challenge studies. In this study, whole genome sequence data were generated for 6 B. pseudomallei isolates previously identified as candidates for animal challenge studies; an additional 5 isolates were sequenced that were associated with human inhalational melioidosis. A core genome single nucleotide polymorphism (SNP) phylogeny inferred from a concatenated SNP alignment from the 11 isolates sequenced in this study and a diverse global collection of isolates demonstrated the diversity of the proposed Animal Rule isolates. To understand the genomic composition of each isolate, a large-scale blast score ratio (LS-BSR) analysis was performed on the entire pan-genome; this demonstrated the variable composition of genes across the panel and also helped to identify genes unique to individual isolates. In addition, a set of ~550 genes associated with pathogenesis in B. pseudomallei were screened against the 11 sequenced genomes with LS-BSR. Differential gene distribution for 54 virulence-associated genes was observed between genomes and three of these genes were correlated with differential virulence observed in animal challenge studies using BALB/c mice. Differentially conserved genes and SNPs associated with disease severity were identified and could be the basis for future studies investigating the pathogenesis of B. pseudomallei. Overall, the genetic characterization of the 11 proposed Animal Rule isolates provides context for future studies involving B. pseudomallei pathogenesis, differential virulence, and efficacy to therapeutics.

planets transiting a pre-main-sequence star presents an important, if stress-inducing, opportunity to observe and measure directly the properties of infant planets. Suárez Mascareño et al. published radial-velocity-derived masses for two of the V1298 Tau planets using a state-of-the-art Gaussian process regression framework. The planetary densities computed from these masses were surprisingly high, implying extremely rapid contraction after formation in tension with most existing planet-formation theories. In an effort to constrain further the masses of the V1298 Tau planets, we obtained 36 RVs using Keck/HIRES, and analyzed them in concert with published RVs and photometry. Through performing a suite of cross-validation tests, we found evidence that the preferred model of Suárez Mascareño et al. suffers from overfitting, defined as the inability to predict unseen data, rendering the masses unreliable. We detail several potential causes of this overfitting, many of which may be important for other RV analyses of other active stars, and recommend that additional time and resources be allocated to understanding and mitigating activity in active young stars such as V1298 Tau.

and period between 1700 and 7200 days, based on a joint analysis of RVs and astrometry from Gaia and Hipparcos. As a bright star hosting multiple planets with well-measured masses, HD 191939 presents many options for comparative planetary astronomy, including characterization with JWST.
Recent efforts to identify treatments for chronic diabetic complications have resulted in the discovery of a novel series of highly potent and selective 3-(benzothiazol-2-yl)methylindole-N-alkanoic ...acid aldose reductase inhibitors. The lead candidate, 3-(4,5,7-trifluorobenzothiazol-2-yl)methylindole-N-acetic acid (lidorestat, 9) inhibits aldose reductase with an IC(50) of 5 nM, while being 5400 times less active against aldehyde reductase, a related enzyme involved in the detoxification of reactive aldehydes. It lowers nerve and lens sorbitol levels with ED(50)'s of 1.9 and 4.5 mg/kg/d po, respectively, in the 5-day STZ-induced diabetic rat model. In a 3-month diabetic intervention model (1 month of diabetes followed by 2 months of drug treatment at 5 mg/kg/d po), it normalizes polyols and reduces the motor nerve conduction velocity deficit by 59% relative to diabetic controls. It has a favorable pharmacokinetic profile (F, 82%; t(1/2), 5.6 h; Vd, 0.694 L/kg) with good drug penetration in target tissues (C(max) in sciatic nerve and eye are 2.36 and 1.45 mug equiv/g, respectively, when dosed with (14)Clidorestat at 10 mg/kg po).

. TOI-2337b has the shortest period (
P
= 2.99432 ± 0.00008 days) of any planet discovered around a red giant star to date. Both TOI-4329b and TOI-2669b appear to be inflated, but TOI-2337b does not show any sign of inflation. The large radii and relatively low masses of TOI-4329b and TOI-2669b place them among the lowest density hot Jupiters currently known, while TOI-2337b is conversely one of the highest. All three planets have orbital eccentricities of below 0.2. The large spread in radii for these systems implies that planet inflation has a complex dependence on planet mass, radius, incident flux, and orbital properties. We predict that TOI-2337b has the shortest orbital decay timescale of any planet currently known, but do not detect any orbital decay in this system. Transmission spectroscopy of TOI-4329b would provide a favorable opportunity for the detection of water, carbon dioxide, and carbon monoxide features in the atmosphere of a planet orbiting an evolved star, and could yield new information about planet formation and atmospheric evolution.
