Two experiments were conducted to evaluate the response of sows to high levels of dietary riboflavin. Responses evaluated were erythrocyte glutathione reductase activity coefficient (EGRAC, a ...biochemical indicator of riboflavin status) and reproductive performance. In Exp. 1, eight sows were fed diets either slightly deficient or superadequate in riboflavin throughout a reproductive cycle, beginning in midgestation. The EGRAC values confirmed the slight deficiency (P < 0.01) but did not clearly show E time when the sow is most likely to be deficient in riboflavin. In Exp. 2, 118 sows at two farms were fed diets providing 10 (control, 1.4 times the NRC 1988 requirement estimate), 60, 110, or 160 mg of riboflavin/d for 21 d beginning at breeding, and the control level through the remainder of the reproductive cycle. The percentage of sows farrowing tended to be lower (P < 0.10) for the control sows than for sow fed the other treatments (66.7, 85.7, 93.3, and 86.7% for 10, 60, 110, and 160 mg/d, respectively). This response is similar to other results reported previously. No other measure of reproductive performance, nor EGRAC measured at any of five stages of the reproductive cycle, was affected by treatment. These results suggest that massive riboflavin supplementation of the sows' diet during early pregnancy may increase the percentage of farrowing but does not increase litter size.
We announce the discovery of 1-100 GeV gamma-ray emission from the archetypal TeV pulsar wind nebula (PWN) HESS J1825-137 using 20 months of survey data from the Fermi-Large Area Telescope (LAT). The ...gamma-ray emission detected by the LAT is significantly spatially extended, with a best-fit rms extension of {sigma} = 0.{sup 0}56 {+-} 0.{sup 0}07 for an assumed Gaussian model. The 1-100 GeV LAT spectrum of this source is well described by a power law with a spectral index of 1.38 {+-} 0.12 {+-} 0.16 and an integral flux above 1 GeV of (6.50 {+-} 0.21 {+-} 3.90) x 10{sup -9} cm{sup -2} s{sup -1}. The first errors represent the statistical errors on the fit parameters, while the second ones are the systematic uncertainties. Detailed morphological and spectral analyses bring new constraints on the energetics and magnetic field of the PWN system. The spatial extent and hard spectrum of the GeV emission are consistent with the picture of an inverse Compton origin of the GeV-TeV emission in a cooling-limited nebula powered by the pulsar PSR J1826-1334.
A new spinning oil film (SOF) solid-in-oil-in-oil emulsion process was developed to produce uniform-sized protein-loaded biodegradable microspheres. A thin SOF on a cylindrical rotor was used to ...shear droplets from a nozzle tip to control droplet size. The resulting microspheres with low polydispersity (6%) produced a low burst (6%-11%) release even at high loadings (13%-18% encapsulated solids, 8%-12% protein). The SOF process had a high yield and did not require the presence of water, which can cause protein denaturation, or surfactants, which may be unwanted in the final product. Amorphous protein and crystalline excipient solids were encapsulated into 3 different polymers, giving a homogenous drug distribution throughout the microspheres, and an essentially complete protein encapsulation efficiency (average = 99%). In contrast, large burst release was observed for polydisperse microspheres produced by a conventional emulsification technique, particularly for microspheres smaller than 25 mum in diameter, which gave 93% burst at 15% loading. The uniform encapsulation of high loadings of proteins into microspheres with low polydispersity in an anhydrous process is of practical interest in the development of controlled-release protein therapeutics.
In order to investigate the analgesic effect of timing of administration of ketorolac 10 mg i.v., we recorded patient-controlled use of diamorphine at 2, 4 and 12 h after abdominal hysterectomy. In a ...randomized, double-blind trial, 30 patients received ketorolac before skin incision and 28 after skin closure. A control group of 32 patients did not receive ketorolac. We measured operative blood loss and assessed nausea, vomiting and pruritus. After 2 h of patient-controlled analgesia, the median cumulative diamorphine dose in the group given ketorolac before operation was less than that of the control group (95% confidence interval 8-66 micrograms kg-1; P = 0.01). There were no other statistically significant differences in diamorphine consumption between the groups. The frequency of nausea and vomiting was similar in all groups Median blood loss in the group given ketorolac before operation exceeded that of the patients who did not receive ketorolac before operation (95% confidence interval 20-149 ml; P = 0.01). We conclude that the diamorphine-sparing effect of ketorolac attributable to timing of administration was small, conferred no clinical benefit and was accompanied by increased bleeding. No patient given ketorolac complained of pruritus.
As part of an effort to develop a low-volatile, essentially dry, carbon fiber tape from a phenylethynyl-terminated imide for use in heated head automated tow placement, a 70:30 blend of LaRCTM PETI-5 ...and LaRCTM PETI-IAX was prepared in N-methyl-2-pyrrolidinone by mixing the phenylethynylterminated amide acid solutions together. Solutions of the amide acids and the blend were characterized by size exclusion chromatography after aging at room temperature and at 5 °C over a 30-day period. Thin films were periodically cast from the solutions, thermally cured, and the tensile properties determined. Unoriented thin-film tensile properties were unaffected by aging conditions over the 30-day period whereas the weight average molecular weights of the phenylethynyl-terminated amide acids decreased. Imide oligomers were prepared from the amide acids, characterized, and thermally cured. A low-volatile tape prepared from prepreg of the 70:30 blend was fabricated into composites by processing in a vacuum press. The thin-film tensile and composite properties of the blend generally followed a rule of mixtures.
Thirty-five new cases of gastrinomas were diagnosed in N. Ireland between 1970 and 1996. Over this period, patient care has improved, with advances in imaging techniques and therapeutic regimens. ...Patients are now no longer presenting in the classical way with severe ulcer diathesis. Diarrhoea is often a major feature, occurring in 46% of patients. Thirty-one percent of patients presented with mixed amine precursor, uptake and decarboxylation (APUD) tumours. Survival has improved, most likely as a result of better detection of tumours, as well as treatment that is aimed at resection and removal of the gastrinoma. The advent of proton pump inhibitors has ensured symptom control in those for whom total tumour removal is impossible. Owing to improved survival, metastatic complications are often associated with patient mortality.
We report the discovery of two millisecond pulsars in a search for radio pulsations at the positions of Fermi-Large Area Telescope sources with no previously known counterparts, using the Nancay ...Radio Telescope. The two millisecond pulsars, PSRs J2017+0603 and J2302+4442, have rotational periods of 2.896 and 5.192 ms and are both in binary systems with low-eccentricity orbits and orbital periods of 2.2 and 125.9 days, respectively, suggesting long recycling processes. Gamma-ray pulsations were subsequently detected for both objects, indicating that they power the associated Fermi sources in which they were found. The gamma-ray light curves and spectral properties are similar to those of previously detected gamma-ray millisecond pulsars. Detailed modeling of the observed radio and gamma-ray light curves shows that the gamma-ray emission seems to originate at high altitudes in their magnetospheres. Additionally, X-ray observations revealed the presence of an X-ray source at the position of PSR J2302+4442, consistent with thermal emission from a neutron star. These discoveries along with the numerous detections of radio-loud millisecond pulsars in gamma rays suggest that many Fermi sources with no known counterpart could be unknown millisecond pulsars.
We describe a novel fungal expression system which utilizes the Quorn myco-protein fungus Fusarium graminearum A 3/5. A transformation system was developed for F. graminearum and was used to ...introduce the coding and regulatory regions of a trypsin gene from Fusarium oxysporum. The protein was efficiently expressed, processed and secreted by the recombinant host strain. In addition, the promoter and terminator of the F. oxysporum trypsin gene have been successfully utilized to drive the expression of a cellulase gene from Scytalidium thermophilum and a lipase gene from Thermomyces lanuginosus in F. graminearum.
