Rates of biodiversity loss are higher in freshwater ecosystems than in most terrestrial or marine ecosystems, making freshwater conservation a priority. However, prioritization methods are impeded by ...insufficient knowledge on the distribution and conservation status of freshwater taxa, particularly invertebrates. We evaluated the extinction risk of the world's 590 freshwater crayfish species using the IUCN Categories and Criteria and found 32% of all species are threatened with extinction. The level of extinction risk differed between families, with proportionally more threatened species in the Parastacidae and Astacidae than in the Cambaridae. Four described species were Extinct and 21% were assessed as Data Deficient. There was geographical variation in the dominant threats affecting the main centres of crayfish diversity. The majority of threatened US and Mexican species face threats associated with urban development, pollution, damming and water management. Conversely, the majority of Australian threatened species are affected by climate change, harvesting, agriculture and invasive species. Only a small proportion of crayfish are found within the boundaries of protected areas, suggesting that alternative means of long-term protection will be required. Our study highlights many of the significant challenges yet to come for freshwater biodiversity unless conservation planning shifts from a reactive to proactive approach.

of body-surface area in the preceding year to receive allopurinol (100 to 300 mg daily) or placebo. The primary outcome was the change in eGFR from randomization to week 104, calculated with the Chronic Kidney Disease Epidemiology Collaboration creatinine equation.
Enrollment was stopped because of slow recruitment after 369 of 620 intended patients were randomly assigned to receive allopurinol (185 patients) or placebo (184 patients). Three patients per group withdrew immediately after randomization. The remaining 363 patients (mean eGFR, 31.7 ml per minute per 1.73 m

Between 1985 and 2014, the number of US doctoral graduates in Anthropology increased from about 350 to 530 graduates per year. This rise in doctorates entering the work force along with an overall ...decrease in the numbers of tenure-track academic positions has resulted in highly competitive academic job market. We estimate that approximately79% of US anthropology doctorates do not obtain tenure-track positions at BA/BS, MA/MS, and PhD institutions in the US. Here, we examine where US anthropology faculty obtained their degrees and where they ultimately end up teaching as tenure-track faculty. Using data derived from the 2014-2015 AnthroGuide and anthropology departmental web pages, we identify and rank PhD programs in terms of numbers of graduates who have obtained tenure-track academic jobs; examine long-term and ongoing trends in the programs producing doctorates for the discipline as a whole, as well as for the subfields of archaeology, bioanthropology, and sociocultural anthropology; and discuss gender inequity in academic anthropology within the US.
Continuum robotics has rapidly become a rich and diverse area of research, with many designs and applications demonstrated. Despite this diversity in form and purpose, there exists remarkable ...similarity in the fundamental simplified kinematic models that have been applied to continuum robots. However, this can easily be obscured, especially to a newcomer to the field, by the different applications, coordinate frame choices, and analytical formalisms employed. In this paper we review several modeling approaches in a common frame and notational convention, illustrating that for piecewise constant curvature, they produce identical results. This discussion elucidates what has been articulated in different ways by a number of researchers in the past several years, namely that constant-curvature kinematics can be considered as consisting of two separate submappings: one that is general and applies to all continuum robots, and another that is robot-specific. These mappings are then developed both for the single-section and for the multi-section case. Similarly, we discuss the decomposition of differential kinematics (the robot’s Jacobian) into robot-specific and robot-independent portions. The paper concludes with a perspective on several of the themes of current research that are shaping the future of continuum robotics.
Excess dormant origins bound by the minichromosome maintenance (MCM) replicative helicase complex play a critical role in preventing replication stress, chromosome instability and tumorigenesis. In ...response to DNA damage, replicating cells must coordinate DNA repair and dormant origin firing to ensure complete and timely replication of the genome; how cells regulate this process remains elusive. Herein, we identify a member of the Fanconi Anemia (FA) DNA repair pathway, FANCI, as a key effector of dormant origin firing in response to replication stress. Cells lacking FANCI have reduced number of origins, increased inter-origin distances and slowed proliferation rates. Intriguingly, ATR-mediated FANCI phosphorylation inhibits dormant origin firing while promoting replication fork restart/DNA repair. Using super-resolution microscopy, we show that FANCI co-localizes with MCM-bound chromatin in response to replication stress. These data reveal a unique role for FANCI as a modulator of dormant origin firing and links timely genome replication to DNA repair.
Central nervous system-expressed long non-coding RNAs (lncRNAs) are often located in the genome close to protein coding genes involved in transcriptional control. Such lncRNA-protein coding gene ...pairs are frequently temporally and spatially co-expressed in the nervous system and are predicted to act together to regulate neuronal development and function. Although some of these lncRNAs also bind and modulate the activity of the encoded transcription factors, the regulatory mechanisms controlling co-expression of neighbouring lncRNA-protein coding genes remain unclear. Here, we used high resolution NG Capture-C to map the cis-regulatory interaction landscape of the key neuro-developmental Paupar-Pax6 lncRNA-mRNA locus. The results define chromatin architecture changes associated with high Paupar-Pax6 expression in neurons and identify both promoter selective as well as shared cis-regulatory-promoter interactions involved in regulating Paupar-Pax6 co-expression. We discovered that the TCF7L2 transcription factor, a regulator of chromatin architecture and major effector of the Wnt signalling pathway, binds to a subset of these candidate cis-regulatory elements to coordinate Paupar and Pax6 co-expression. We describe distinct roles for Paupar in Pax6 expression control and show that the Paupar DNA locus contains a TCF7L2 bound transcriptional silencer whilst the Paupar transcript can act as an activator of Pax6. Our work provides important insights into the chromatin interactions, signalling pathways and transcription factors controlling co-expression of adjacent lncRNAs and protein coding genes in the brain.
Abstract
This study presents a numerical analysis of the topology of a set of cosmologically interesting 3D Gaussian random fields in terms of their Betti numbers β0, β1, and β2. We show that Betti ...numbers entail a considerably richer characterization of the topology of the primordial density field. Of particular interest is that the Betti numbers specify which topological features – islands, cavities, or tunnels – define the spatial structure of the field. A principal characteristic of Gaussian fields is that the three Betti numbers dominate the topology at different density ranges. At extreme density levels, the topology is dominated by a single class of features. At low levels this is a Swiss-cheeselike topology dominated by isolated cavities, and, at high levels, a predominantly Meatball-like topology composed of isolated objects. At moderate density levels, two Betti numbers define a more Sponge-like topology. At mean density, the description of topology even needs three Betti numbers, quantifying a field consisting of several disconnected complexes, not of one connected and percolating overdensity. A second important aspect of Betti number statistics is that they are sensitive to the power spectrum. They reveal a monotonic trend, in which at a moderate density range, a lower spectral index corresponds to a considerably higher (relative) population of cavities and islands. We also assess the level of complementary information that the Betti numbers represent, in addition to conventional measures such as Minkowski functionals. To this end, we include an extensive description of the Gaussian Kinematic Formula, which represents a major theoretical underpinning for this discussion.
AIM: The species–area relationship (SAR) is widely used in conservation science to predict the number of species likely to go extinct as a result of habitat loss. Often, studies employing the SAR use ...total species richness as the dependent variable. However, this overlooks the fact that habitat specialists and generalists differ in their susceptibility to habitat loss. We undertook a synthetic review of 23 habitat island datasets for birds to determine the impact of habitat generalists on the SAR. LOCATION: Global. METHODS: We sourced 19 habitat island datasets from the literature and combined these data with four of our own empirically gathered datasets. For each dataset, we classified all bird species as either forest habitat specialists or generalists. We then fitted the power SAR model (log–log and nonlinear forms) to the specialists, generalists and all species for each dataset and compared the resulting model parameters. We compared differences in the rate of change in richness with area between specialists and generalists using the first derivative of a multimodel SAR. RESULTS: We found that the slope of the power model was steeper for habitat specialists in the majority of datasets, and this difference was significant in 15 and 16 of the 23 datasets, for the nonlinear and log–log forms of the power model, respectively. Comparison of the multimodel SAR curve derivatives revealed further differences in the rate of change in species richness with area between subsets. MAIN CONCLUSIONS: The z values of both forms of the power model of the specialists' SARs were generally larger, often considerably so, than the values used in most SAR studies predicting extinctions from habitat loss. Thus, studies that have used z values derived from SAR studies using total richness may be underestimating the impact of habitat loss on specialist species, which are likely to be those of greatest conservation concern.
Anaesthesiologists and non-anaesthesiologist sedationists have a limited set of available i.v. hypnotics, further reduced by the withdrawal of thiopental in the USA and its near disappearance in ...Europe. Meanwhile, demand for sedation increases and new clinical groups are using what traditionally are anaesthesiologists' drugs. Improved understanding of the determinants of perioperative morbidity and mortality has spotlighted hypotension as a potent cause of patient harm, and practice must be adjusted to respect this. High-dose propofol sedation may be harmful, and a critical reappraisal of drug choices and doses is needed. The development of remimazolam, initially for procedural sedation, allows reconsideration of benzodiazepines as the hypnotic component of a general anaesthetic even if their characterisation as i.v. anaesthetics is questionable. Early data suggest that a combination of remimazolam and remifentanil can induce and maintain anaesthesia. Further work is needed to define use cases for this technique and to determine the impact on patient outcomes.
Continuum robots, which are composed of multiple concentric, precurved elastic tubes, can provide dexterity at diameters equivalent to standard surgical needles. Recent mechanics-based models of ...these "active cannulas" are able to accurately describe the curve of the robot in free space, given the preformed tube curves and the linear and angular positions of the tube bases. However, in practical applications, where the active cannula must interact with its environment or apply controlled forces, a model that accounts for deformation under external loading is required. In this paper, we apply geometrically exact rod theory to produce a forward kinematic model that accurately describes large deflections due to a general collection of externally applied point and/or distributed wrench loads. This model accommodates arbitrarily many tubes, with each having a general preshaped curve. It also describes the independent torsional deformation of the individual tubes. Experimental results are provided for both point and distributed loads. Average tip error under load was 2.91 mm (1.5% - 3% of total robot length), which is similar to the accuracy of existing free-space models.
