This paper discusses current state-of-the-art features of the “plant evaluation model”, a framework which, starting from the representation of vegetation and its effects in microclimate models, ...defines a number of indices which can be employed for the evaluation of outdoor microclimate in terms of thermal environment and comfort in the urban environment. The key point of taking into account the impact of vegetation on microclimate is to implement appropriate parameterizations of such impact in a microclimate model. The paper, based on a review of literature studies, thus illustrates the basic principle and technical path of the impact assessment model of vegetation on microclimate and introduces related software. The aim is to provide the scientific community with a summary of (i) the current definition of vegetation in models employed for the evaluation of the impact of vegetation on urban outdoor microclimate, (ii) main models and evaluation indices and (iii) main input, output, vegetation-related processes implemented, strengths and weaknesses of those models, with suggested measures for output improvement. This review is not exhaustive but may help the user to select the proper model, which takes into account the effects of vegetation on outdoor urban microclimate, depending on the specific objective.

Bioclimatic design provides solutions to mitigating the urban heat island (UHI) effect and improving urban quality of life. Waterfront settings provide a unique opportunity for UHI mitigation, as cool winds (i.e. sea breezes) provide good cooling conditions. We used ENVI-met numerical simulations to investigate the synergistic mitigation effects of redeveloped urban forms, ventilation corridors, and extensive greenery on local microclimate and outdoor thermal comfort at an urban waterfront in Victoria Harbour, Hong Kong. The thermal performance along the waterfront and within an inner urbanised area was evaluated in three scenarios: existing configuration (Case A), redeveloped building form (Case B), and Case B with a ventilation corridor and extensive greenery added. In the study area's subtropical climate, the additional strategies used led to a synergistic improvement in microclimate and thermal comfort in both settings. The appropriate use of combined urban forms and ventilation corridors along waterfronts can thus lead to a more acceptable pedestrian-level wind environment and enhance the ventilation potential within inner urbanised areas. The cooling intensity of greenery (turf and green facades) along such waterfronts can also be extended for further benefits. Overall, the synergistic mitigation effects of redeveloped urban forms, ventilation corridors, and extensive greenery in proximity to water bodies demonstrated here provide science-based guidance for the use of bioclimatic design along urban waterfronts in order to achieve improved microclimates and thermal comfort at neighbourhood scales.

This study aimed at evaluating DOP efficiency at inhibiting 1-Methyl-2-nitro-1-nitrosoguanidine ...(MNNG) induced precancerous lesions of gastric cancer (PLGC) in rats through the Wnt/b-catenin pathway and analyzing the variations of serum endogenous metabolites. PLGC was established in male Sprague-Dawley (SD) rats by administering 150 μg/mL MNNG in drinking water for 7 months and giving 0.1 mL of 10% NaCl once weekly during the initial 20 weeks. Treatment with DOP inhibited the progress of PLGC through decreasing the expression of β-catenin by immunohistochemical analysis. The futher study indicated DOP downregulated gene expression of Wnt2β, Gsk3β, PCNA, CyclinD1, and β-catenin, as well as protein expression of Wnt2β, PCNA, and β-catenin. On the other hand, there were nine endogenous metabolites identified after the DOP treatment. Among these, the most significant one is betaine because of its strong antioxidant activity, leading to an anti-tumor effect. DOP can inhibit MNNG-induced PLGC models via regulating Wnt/β-catenin pathway and by changing endogenous metabolites.
Leptin (LEP) is reported to play important roles in controlling energy balance in vertebrates, including birds. However, it remains an open question whether an authentic “LEP gene” exists and ...functions in birds. Here, we identified and characterized a LEP gene (zebra finch LEP zbLEP) encoding a 172-amino acid precursor in zebra finches. Despite zbLEP showing limited amino acid sequence identity (26%–29%) to human and mouse LEPs, synteny analysis proved that zbLEP is orthologous to mammalian LEP. Using a pAH32 luciferase reporter system and Western blot analysis, we demonstrated that the recombinant zbLEP protein could potently activate finch and chicken LEP receptors (zbLEPR; cLEPR) expressed in human embryonic kidney 293 cells and enhance signal transducer and activator of transcription 3 phosphorylation, further indicating that zbLEP is a functional ligand for avian LEPRs. Interestingly, quantitative real-time RT-PCR revealed that zbLEP mRNA is expressed nearly exclusively in the pituitary and various brain regions but undetectable in adipose tissue and liver, whereas zbLEPR mRNA is widely expressed in adult finch tissues examined with abundant expression noted in pituitary, implying that unlike mammalian LEP, finch LEP may not act as an adipocyte-derived signal to control energy balance. As in finches, a LEP highly homologous to zbLEP was also identified in budgerigar genome. Strikingly, finch and budgerigar LEPs show little homology with chicken LEP (cLEP) previously reported, suggesting that the so-called cLEP is incorrect. Collectively, our data provide convincing evidence for the existence of an authentic functional LEP in avian species and suggest an important role of brain- and pituitary-derived LEP played in vertebrates.
Avian infectious bronchitis virus (IBV) is a gamma coronavirus that severely affects the poultry industry worldwide. Long non-coding RNAs (lncRNAs), a subset of non-coding RNAs with a length of more ...than 200 nucleotides, have been recently recognized as pivotal factors in the pathogenesis of viral infections. However, little is known about the function of lncRNAs in host cultured cells in response to IBV infection.
We used next-generation high throughput sequencing to reveal the expression profiles of mRNAs and lncRNAs in IBV-infected HD11 cells. Compared with the uninfected cells, we identified 153 differentially expressed (DE) mRNAs (106 up-regulated mRNAs, 47 down-regulated mRNAs) and 181 DE lncRNAs (59 up-regulated lncRNAs, 122 down-regulated lncRNAs) in IBV-infected HD11 cells. Moreover, gene ontology (GO) and pathway enrichment analyses indicated that DE mRNAs and lncRNAs were mainly involved in cellular innate immunity, amino acid metabolism, and nucleic acid metabolism. In addition, 2640 novel chicken lncRNAs were identified, and a competing endogenous RNA (ceRNAs) network centered on gga-miR-30d and miR-146a-5p was established.

Slurry based lithium-ion flow battery is a promising technology to improve the energy density of redox flow batteries for various applications. However, the high viscosity and flow resistance of ...slurry increase the pumping loss and limit the volume ratio of active materials, which hinders its further improvement in energy density. Here we propose a concept of single-component slurry based lithium-ion flow battery by utilizing carbon felt as the 3D current collector. The single-component slurry, consisting of lithium intercalation particles and electrolyte, can significantly decrease the slurry viscosity due to elimination of adding discrete carbon particles, enabling an increase in the volume ratio of active materials to further improve volumetric capacity and energy density of the lithium-ion flow battery. The demonstrated low-viscosity lithium iron phosphate slurry based battery achieves an energy density of 230 Wh L−1 and coulombic efficiency >95% over 100 cycles in a coin cell and good stability in flow-cell tests for both intermittent and continuous flow modes. The concept may offer a new opportunity for slurry based flow battery to minimize the viscosity and maximize the volumetric capacity.

also poses a serious threat to public health. This study aims to characterize antibiotic resistance, virulence genes, and biofilm formation of enterococci from yaks. From April 2018 to July 2019, we collected 395 fecal samples of yaks in Aba Tibetan Autonomous Prefecture, China. Enterococci isolated from the samples were identified and classified according to the 16S rDNA sequence. The antibiotic resistance of each isolate was detected according to the Kirby-Bauer disk diffusion method, and antibiotic resistance genes were detected by polymerase chain reaction (PCR) and sequencing. Enterococcal biofilms were assessed using standard procedures. Different virulence genes were detected by PCR and sequencing. In total, 381 enterococci strains were recovered, with
Enterococcus faecalis
(41.99%) and
Enterococcus faecium
(37.80%) being the predominant species. Many isolates were multidrug- resistant (60.37%) and showed a high resistance rate to rifampicin (64.30%) and tetracycline (61.54%). We also detected various antimicrobial resistance (AMR) genes in the tested strains. The
E. faecalis
strains had higher frequency of biofilm formation and virulence genes than other enterococcal species. This is the first report that shows yaks are repositories for drug-resistant enterococci with virulent determinants and biofilms that may spread into humans and to environment. This study also provides useful data suggesting that enterococci may pose a potential health risk to yaks. Therefore, active surveillance of AMR and pathogenesis in enterococci from yaks is urgently warranted.
Salt bridges are important factors in maintaining the stability of proteins, and their contribution to protein folding has received much attention. Although the interaction energies, or stabilizing ...contributions, of individual salt bridges have been measured in various proteins, a systematic assessment of various types of salt bridges in a relatively uniform environment is still a valuable analysis. Here, we used a collagen heterotrimer as a host-guest platform to construct 48 heterotrimers with the same charge pattern. A variety of salt bridges were formed between the oppositely charged residues Lys, Arg, Asp, and Glu. The melting temperature (Tm) of the heterotrimers was measured with circular dichroism. The atomic structures of 10 salt bridges were shown in three x-ray crystals of heterotrimer. Molecular dynamics simulation based on the crystal structures indicated that strong, intermediate, and weak salt bridges have distinctive N-O distances. A linear regression model was used to predict the stability of heterotrimers with high accuracy (R2 = 0.93). We developed an online database to help readers understand how a salt bridge stabilizes collagen. This work will help us better understand the stabilizing mechanism of salt bridges in collagen folding and provide a new strategy to design collagen heterotrimers.

Atomistic simulations and mutagenesis based on the solved structure are used to explore the contributions of specific interactions to energetics. A predictive model of collagen stability and specificity is developed for engineering novel collagen structures.
Avian infectious bronchitis (IB), caused by avian infectious bronchitis virus (IBV), is an acute and highly contagious disease that is extremely harmful to the poultry industry throughout the world. ...The cross-using of different attenuated live vaccine strains has led to the occurrence of diverse IBV serotypes. In this study, we isolated an IBV strain from a chicken farm in southwest China and designated it CK/CH/SCMY/160315. Construction of a phylogenetic tree based on full S1 gene sequence analysis suggested that CK/CH/SCMY/160315 bears similarity to GI-28, and further comparison of S1 amino acid residues revealed that CK/CH/SCMY/160315 showed mutations and deletions in many key positions between LDT3-A and other GI-28 reference strains. Importantly, CK/CH/SCMY/160315 was identified as a novel recombinant virus derived from live attenuated vaccine strains H120 (GI-1), 4/91 (GI-13), LDT3-A (GI-28), and the field strain LJL/08-1 (GI-19), identifying at least 5 recombination sites in both structural and accessory genes. Pathogenicity analysis indicated that CK/CH/SCMY/160315 caused listlessness, sneezing, huddling, head shaking, and increased antibody levels in the inoculated chickens. To further describe pathogenicity of this novel strain, we assessed viral load in different tissues and conducted hematoxylin and eosin (HE) staining of the trachea, lungs and kidneys. Our results provide evidence for the continuing evolution of IBV field strains via genetic recombination and mutation, leading to outbreaks in the vaccinated chicken populations in China.
