Introduction L’implication du transporteur des sucres GLUT2 dans la sécrétion d’insuline par les cellules beta pancréatiques est bien caractérisée chez les rongeurs mais reste controversée chez ...l’Homme. Le syndrome de Fanconi-Bickel (FBS), caractérisé par des désordres de l’homéostasie glucidique sans diabète, est associé à des mutations inactivantes des fonctions de hGLUT2. Certains jeunes patients FBS peuvent souffrir d’hyperglycémie et d’hypoinsulinémie en réponse à une charge de glucose; cependant ces symptômes disparaissent avec l’âge. Réciproquement dans le syndrome d’hyperinsulinisme congénital (CHI), des mutations activantes de hGLUT2 pourraient être à l’origine de l’hyperinsulinémie et de l’hypoglycémie sévère des nouveau-nés. Le but de cette étude est d’évaluer les conséquences fonctionnelles sur le transport de sucre et la sécrétion d’insuline de mutations de hGLUT2 identifiées chez des patients FBS et CHI. Matériels et méthodes Neuf mutations faux-sens de hGLUT2 (4 identifiées chez des patients FBS, 3 chez des patients CHI et 2 dans la population générale) ont été introduites dans des vecteurs d’expression de hGLUT2 par mutagénèse dirigée. L’expression protéique de hGLUT2 a été analysée par immunohistochimie et western blot. Les paramètres cinétiques de transport ont été mesurés par le captage de 2-deoxy-D-glucose radiomarqué dans des ovocytes de xénope exprimant les variants hGLUT2. La sécrétion d’insuline en réponse au glucose a été mesurée dans les cellules insulino-sécrétrice MIN6 infectées avec les variants CHI de hGLUT2. Résultats Nous montrons que hGLUT2 est exprimé dans les cellules insulino-positives du pancréas de patients CHI. De plus, deux mutations identifiées chez les patients CHI provoquent une sécrétion d’insuline exacerbée même en absence de glucose dans le milieu de culture des cellules. En revanche, les mutations associées au FBS, inhibent l’expression protéique ou les capacités de transport de hGLUT2. Conclusion Dans le pancréas de nouveau-né humain, hGLUT2 jouerait un rôle essentiel dans le contrôle de la sécrétion d’insuline. Nous proposons donc de séquencer hGLUT2 chez les patients CHI.
A search for the standard model Higgs boson decaying to W+W- in pp collisions at sqrt(s) = 7 TeV is reported. The data are collected at the LHC with the CMS detector, and correspond to an integrated ...luminosity of 4.6 inverse femtobarns. The W+W- candidates are selected in events with two charged leptons and large missing transverse energy. No significant excess of events above the standard model background expectations is observed, and upper limits on the Higgs boson production relative to the standard model Higgs expectation are derived. The standard model Higgs boson is excluded in the mass range 129-270 GeV at 95% confidence level.
A search for the standard model Higgs boson decaying into two Z bosons with subsequent decay into a final state containing two quark jets and two leptons, H to ZZ(*) to q q-bar l-l+ is presented. ...Results are based on data corresponding to an integrated luminosity of 4.6 inverse femtobarns of proton-proton collisions at sqrt(s)=7 TeV, collected with the CMS detector at the LHC. In order to discriminate between signal and background events, kinematic and topological quantities, including the angular spin correlations of the decay products, are employed. Events are further classified according to the probability of the jets to originate from quarks of light or heavy flavor or from gluons. No evidence for the Higgs boson is found, and upper limits on its production cross section are determined for a Higgs boson of mass between 130 and 600 GeV.
The production of b jets in association with a Z/gamma* boson is studied using proton-proton collisions delivered by the LHC at a centre-of-mass energy of 7 TeV and recorded by the CMS detector. The ...inclusive cross section for Z/gamma* + b-jet production is measured in a sample corresponding to an integrated luminosity of 2.2 inverse femtobarns. The Z/gamma* + b-jet cross section with Z/gamma* to ll (where ll = ee or mu mu) for events with the invariant mass 60 < M(ll) < 120 GeV, at least one b jet at the hadron level with pT > 25 GeV and abs(eta) < 2.1, and a separation between the leptons and the jets of Delta R > 0.5 is found to be 5.84 +/- 0.08 (stat.) +/- 0.72 (syst.) +(0.25)/-(0.44) (theory) pb. The kinematic properties of the events are also studied and found to be in agreement with the predictions made by the MadGraph event generator with the parton shower and the hadronisation performed by PYTHIA.
A measurement of the exclusive two-photon production of muon pairs in proton-proton collisions at sqrt(s)= 7 TeV, pp to p mu^+ mu^- p, is reported using data corresponding to an integrated luminosity ...of 40 inverse picobarns. For muon pairs with invariant mass greater than 11.5 GeV, transverse momentum pT(mu) > 4 GeV and pseudorapidity |eta(mu)| < 2.1, a fit to the dimuon pt(mu^+ mu^-) distribution results in a measured cross section of sigma(pp to p mu^+ mu^- p) = 3.38 +0.58 -0.55 (stat.) +/- 0.16 (syst.) +/- 0.14 (lumi.) pb, consistent with the theoretical prediction evaluated with the event generator Lpair. The ratio to the predicted cross section is 0.83 +0.14-0.13 (stat.) +/- 0.04 (syst.) +/- 0.03 (lumi.). The characteristic distributions of the muon pairs produced via photon-photon fusion, such as the muon acoplanarity, the muon pair invariant mass and transverse momentum agree with those from the theory.
A search is presented for a massive particle, generically referred to as a Z', decaying into a t t-bar pair. The search focuses on Z' resonances that are sufficiently massive to produce highly ...Lorentz-boosted top quarks, which yield collimated decay products that are partially or fully merged into single jets. The analysis uses new methods to analyze jet substructure, providing suppression of the non-top multijet backgrounds. The analysis is based on a data sample of proton-proton collisions at a center-of-mass energy of 7 TeV, corresponding to an integrated luminosity of 5 inverse femtobarns. Upper limits in the range of 1 pb are set on the product of the production cross section and branching fraction for a topcolor Z' modeled for several widths, as well as for a Randall--Sundrum Kaluza--Klein gluon. In addition, any enhancement to t t-bar production for invariant masses above 1 TeV in the standard model is constrained.

No significant excess is observed above the background expectation, and upper limits are set on the Higgs boson production cross section. The presence of the standard model Higgs boson with a mass in the 270-440 GeV range is excluded at 95% confidence level.
Measurements of jet characteristics from inclusive jet production in proton-proton collisions at a centre-of-mass energy of 7 TeV are presented. The data sample was collected with the CMS detector at ...the LHC during 2010 and corresponds to an integrated luminosity of 36 inverse picobarns. The mean charged hadron multiplicity, the differential and integral jet shape distributions, and two independent moments of the shape distributions are measured as functions of the jet transverse momentum for jets reconstructed with the anti-kT algorithm. The measured observables are corrected to the particle level and compared with predictions from various QCD Monte Carlo generators.
A search is reported for the standard model Higgs boson in the decay mode H to ZZ to tau plus lepton pairs, where the leptons are either electrons or muons, in proton-proton collisions at sqrt(s)=7 ...TeV, corresponding to an integrated luminosity of 4.7 inverse femtobarn collected with the CMS detector at the LHC. No evidence is found for a significant deviation from the background expectation. An upper limit four to twelve times larger than the predicted value is set at 95% confidence level for the product of the standard model Higgs boson production cross section and decay branching fraction in the mass range 190 < m(H) < 600 GeV.
