In view of the fact that the factors affecting the subsidence deformation of subway engineering are complex and the subsidence monitoring data are random, this paper constructs an optimized wavelet ...neural network model with markov chain improvement to improve the accuracy of the predicted data. Firstly, the wavelet neural network is optimized by adding momentum and adjusting learning rate adaptively. Finally, based on markov chain theory, the residual between the predicted value and the actual monitoring value of the optimized wavelet neural network is improved. Taking the settlement data of a settlement monitoring point of xi'an metro line 14 as the research object, the prediction accuracy of the model was compared and analyzed by C++ program. The results of the program show that the additional momentum and the adaptive learning rate are effective for the optimization of the wavelet neural network model, and the prediction accuracy of the optimized wavelet neural network model improved by the markov chain is higher than that of the single model, which is suitable for the prediction of the subway engineering settlement.
In recent years, online learning has become more and more popular. However, because of information overload, learners often find it difficult to retrieve suitable learning resources. Although many ...scholars have proposed excellent online learning resource recommendation algorithms, the accuracy of personalized recommendation results still needs to be improved. This study proposes an improved hybrid ontology-based approach for online learning resource recommendations, combining collaborative filtering algorithm and sequential pattern mining (SPM) techniques. Ontology can be used effectively for knowledge representation to avoid cold start and data sparsity problems. And the history of learners’ sequential access patterns helps in providing recommendations that are more consistent with the law of learning activities. Experimental results reveal that our improved hybrid approach for learning resource recommendations yields better performance and recommendation quality than other related algorithms. Compared with previous research outcomes, our collaborative filtering engine, with ontology domain knowledge, makes full use of the historical learning paths of similar learners. The ontology construction in this study has a more reliable theoretical basis and the selection of features is more representative. In addition, improvement of the SPM process further improves the efficiency of our recommended algorithm.
Biotransformation and biodegradation of estrogenic compounds by bacteria and even fungi have been reported widely, but the role of microalgae in the elimination of estrogens from municipal wastewater ...treatment plants and their interaction with other microorganisms in wastewater are not clear. This study reported the feasibility of repeatedly removing a mixture of 17β-estradiol (E2) and 17α-ethinylestradiol (EE2), each was 100 μg L
, from primary settled municipal sewage by Selenastrum capricornutum (SC), a ubiquitous microalga, in four exposure cycles, each lasted 7 days, and how they interacted with the microbial consortium in sewage. Mixed estrogen in sewage stimulated the growth of SC, and the indigenous microorganisms in sewage also affected the microalgal growth. The indigenous microorganisms, particularly bacteria, could easily remove E2 (with 99.5% removal), so the role of SC was insignificant. On the contrary, EE2 was difficult to remove by indigenous microorganisms but the removal was significantly enhanced by SC, with almost all spiked EE2 being removed, even at the end of the fourth cycle (with 99.0% removal). These results indicated that SC, together with the indigenous microorganisms in wastewater, could be repeatedly used for simultaneous removal of E2 and EE2 from municipal sewage.

The development of alternative isothermal amplification assays including multiple cross displacement amplification (MCDA) may address speed and portability limitations of real-time PCR (rt-PCR) ...methods for SARS-CoV-2 detection. We developed a novel SARS-CoV-2 MCDA assay and compared its speed and sensitivity to loop-mediated isothermal amplification (LAMP) and rt-PCR. Two MCDA assays targeting SARS-CoV-2 N gene and ORF1ab were designed. The fastest time to detection and sensitivity of MCDA was compared to LAMP and rt-PCR using DNA standards and transcribed RNA. For the N gene, MCDA was faster than LAMP and rt-PCR by 10 and 20 min, respectively with fastest time to detection at 5.2 min. rt-PCR had the highest sensitivity with the limit of detection at 10 copies/µl compared with MCDA (100 copies/µl) and LAMP (500 copies/µl). For ORF1ab, MCDA and LAMP had similar speed with fastest time to detection at 9.7 and 8.4 min, respectively. LAMP was more sensitive for ORF1ab detection with 50 copies/µl compared to MCDA (500 copies/µl). In conclusion, different nucleic acid amplification methods provide different advantages. MCDA is the fastest nucleic acid amplification method for SARS-CoV-2 while rt-PCR is the most sensitive. These advantages should be considered when determining the most suitable nucleic acid amplification methods for different applications.
Osteosarcoma (OS) is a highly aggressive malignant bone tumor that mainly occurs in adolescents. At present, chemotherapy is the most commonly used method in clinical practice to treat OS. However, ...due to drug resistance, toxicity and long-term side effects, chemotherapy can’t always provide sufficient benefits for OS patients, especially those with metastasis and recurrence. Natural products have long been an excellent source of anti-tumor drug development. In the current study, we evaluated the anti-OS activity of Echinatin (Ecn), a natural active component from the roots and rhizomes of licorice, and explored the possible mechanism. We found that Ecn inhibited the proliferation of human OS cells and blocked cell cycle at S phase. In addition, Ecn suppressed the migration and invasion, while induced the apoptosis of human OS cells. However, Ecn had less cytotoxicity against normal cells. Moreover, Ecn inhibited the xenograft tumor growth of OS cells in vivo. Mechanistically, Ecn inactivated Wnt/β-catenin signaling pathway while activated p38 signaling pathway. β-catenin over-expression and the p38 inhibitor SB203580 both attenuated the inhibitory effect of Ecn on OS cells. Notably, we demonstrated that Ecn exhibited synergistic inhibitory effect with cisplatin (DDP) on OS cells in vitro and in vivo. Therefore, our results suggest that Ecn may exert anti-OS effects at least partly through regulating Wnt/β-catenin and p38 signaling pathways. Most meaningfully, the results obtained suggest a potential strategy to improve the DDP-induced tumor-killing effect on OS cells by combining with Ecn.

A highly sensitive and selective molecularly imprinted (MIP) sensor combined with magnetic molecularly imprinted solid phase extraction (MMISPE) was developed for the determination of dibutyl ...phthalate (DBP) in complex matrixes. The magnetic molecularly imprinted polymer (MMIP) was synthesized as solid phase extraction (SPE) sorbet to extract DBP from complex matrixes and as sensing element to improve the selectivity of the imprinted sensor. The morphologies of MMIP and MIP-sensor were characterized by using scanning electron microscope (SEM) and transmission electron microscopy (TEM). The electrochemical performances of MIP-sensor were investigated by cyclic voltammetry (CV) and differential pulse voltammetry (DPV). The conditions of preconcentration, elution and electrochemical determination were studied in detail. Under the optimized experimental conditions, the response currents of the MIP-sensor exhibited a linear relationship towards DBP concentrations ranging from 1.0×10−8g/L to 1.0×10−3g/L. The limit of detection of the MMIP-sensor coupled with the MMISPE was calculated as 0.052ng/L. The MMIP-sensor coupled with the MMISPE was applied to detect DBP in complex samples successfully.
•A sensitive magnetic imprinted sensor combined with magnetic imprinted extraction was developed.•Molecularly imprinted polymer was synthesized as SPE sorbent and as sensing element.•Trace DBP can be determined by the MMIP sensor combined with MMISPE with LOD of 5.2×10−11g/L.
Manganese (Mn), an essential element for plants, can be toxic when present in excess. Stylo (Stylosanthes) is a pioneer tropical legume with great potential for Mn tolerance, but its Mn tolerance ...mechanisms remain poorly understood.
In this study, variations in Mn tolerance were observed among nine stylo genotypes. Stylo genotype 'RY5' exhibited the highest Mn tolerance compared to the other tested genotypes, whereas 'TF2001' was a Mn-sensitive genotype. The mechanisms underlying the response of stylo to Mn toxicity were further investigated using these two genotypes with contrasting Mn tolerance. Results showed that stylo genotype RY5 exhibited Mn tolerance superior to that of genotype TF2001, showing lower reductions in leaf chlorophyll concentration, chlorophyll fluorescence parameters, photosynthetic indexes and plant dry weight under Mn toxicity. A label-free quantitative proteomic analysis was conducted to investigate the protein profiles in the leaves and roots of RY5 in response to Mn toxicity. A total of 356 differentially expressed proteins (DEPs) were identified, including 206 proteins from leaves and 150 proteins from roots, which consisted of 71 upregulated, 62 downregulated, 127 strongly induced and 96 completely suppressed proteins. These DEPs were mainly involved in defense response, photosynthesis, carbon fixation, metabolism, cell wall modulation and signaling. The qRT-PCR analysis verified that 10 out of 12 corresponding gene transcription patterns correlated with their encoding proteins after Mn exposure. Finally, a schematic was constructed to reveal insights into the molecular processes in the leaves and roots of stylo in response to Mn toxicity.
These findings suggest that stylo plants may cope with Mn toxicity by enhancing their defense response and phenylpropanoid pathways, adjusting photosynthesis and metabolic processes, and modulating protein synthesis and turnover. This study provides a platform for the future study of Mn tolerance mechanisms in stylo and may lead to a better understanding of the potential mechanisms underlying tropical legume adaptation to Mn toxicity.
Abstract
Obstructive sleep apnea syndrome (OSAS), a state of sleep disorder, is characterized by repetitive apnea, chronic hypoxia, oxygen desaturation, and hypercapnia. Previous studies have ...revealed that intermittent hypoxia (IH) conditions in OSAS patients elicited neuron injury (especially in the hippocampus and cortex), leading to cognitive dysfunction, a significant and extraordinary complication of OSAS patients. The repeated courses of airway collapse and obstruction in OSAS patients resulted in apnea and arousal during sleep, leading to IH and excessive daytime sleepiness (EDS) and subsequently contributing to the development of inflammation. IH-mediated inflammation could further trigger various types of cognitive dysfunction. Many researchers have found that, besides continuous positive airway pressure (CPAP) treatment and surgery, anti-inflammatory substances might alleviate IH-induced neurocognitive dysfunction. Clarifying the role of inflammation in IH-mediated cognitive impairment is crucial for potentially valuable therapies and future research in the related domain. The objective of this article was to critically review the relationship between inflammation and cognitive deficits in OSAS.
