1. Calibrating indices of animal abundance to true densities is critical in wildlife studies especially when direct density estimations are precluded by high costs, lack of required data or model ...parameters, elusiveness and rarity of target species. For studies deploying camera traps, the use of photographic rate (photographs per sampling time) as an index of abundance potentially applies to the majority of terrestrial mammals where individual recognition, and hence capture—recapture analysis, are unfeasible. The very few studies addressing this method have either been limited by lack of independence between trapping rates and density estimations, or because they combined different species, thus introducing potential bias in camera trap detection rates. This study uses a single model species from several sites to analyse calibration of trapping rates to independently derived estimations of density. The study also makes the first field test of the method by Rowcliffe et al. (2008) for density derivation from camera trapping rates based on modelling animal-camera contacts. 2. We deployed camera traps along line transects at six sites in the Udzungwa Mountains of Tanzania and correlated trapping rates of Harvey's duiker Cephalophus harveyi with densities estimated from counts made along the same transects. 3. We found a strong, linear relationship (R² = 0·90) between trapping rate and density. Sampling precision analysis indicates that camera trapping rates reach satisfactory precision when trapping effort amounts to 250-300 camera days. Density estimates using Rowcliffe et al.'s (2008) gas model conversion are higher than from transect censuses; we discuss the possible reasons and stress the need for more field tests. 4. Synthesis and applications. Subject to rigorous and periodic calibration, and standardization of sampling procedures in time and over different sites, camera trapping rate is shown to be, in this study, a valid index of density in the target species. Comparative data indicate that this may also apply to forest ungulates in general. The method has great potential for standardizing monitoring programmes and reducing the costs of wildlife surveys, especially in remote areas.

This record low leakage current density for InAsSb based devices demonstrates the suitability of nanowires for the integration of III–V semiconductors with silicon technology.
The application of single-molecule fluorescence techniques to complex biological systems places demands on the performance of single fluorophores. We present an enzymatic oxygen scavenging system for ...improved dye stability in single-molecule experiments. We compared the previously described protocatechuic acid/protocatechuate-3,4-dioxygenase system to the currently employed glucose oxidase/catalase system. Under standardized conditions, we observed lower dissolved oxygen concentrations with the protocatechuic acid/protocatechuate-3,4-dioxygenase system. Furthermore, we observed increased initial lifetimes of single Cy3, Cy5, and Alexa488 fluorophores. We further tested the effects of chemical additives in this system. We found that biological reducing agents increase both the frequency and duration of blinking events of Cy5, an effect that scales with reducing potential. We observed increased stability of Cy3 and Alexa488 in the presence of the antioxidants ascorbic acid and
n-propyl gallate. This new O
2-scavenging system should have wide application for single-molecule fluorescence experiments.
The initiation of translation establishes the reading frame for protein synthesis and is a key point of regulation. Initiation involves factor-driven assembly at a start codon of a messenger RNA of ...an elongation-competent 70S ribosomal particle (in bacteria) from separated 30S and 50S subunits and initiator transfer RNA. Here we establish in Escherichia coli, using direct single-molecule tracking, the timing of initiator tRNA, initiation factor 2 (IF2; encoded by infB) and 50S subunit joining during initiation. Our results show multiple pathways to initiation, with orders of arrival of tRNA and IF2 dependent on factor concentration and composition. IF2 accelerates 50S subunit joining and stabilizes the assembled 70S complex. Transition to elongation is gated by the departure of IF2 after GTP hydrolysis, allowing efficient arrival of elongator tRNAs to the second codon presented in the aminoacyl-tRNA binding site (A site). These experiments highlight the power of single-molecule approaches to delineate mechanisms in complex multicomponent systems.
Accurately simulating the spatiotemporal distribution of mountain snow water equivalent improves estimates of available meltwater and benefits the water resource management community. In this paper ...we present the first integration of lidar‐derived distributed snow depth data into a physics‐based snow model using direct insertion. Over four winter seasons (2013–2016) the National Aeronautics and Space Administration/Jet Propulsion Laboratory (NASA/JPL) Airborne Snow Observatory (ASO) performed near‐weekly lidar surveys throughout the snowmelt season to measure snow depth at high resolution over the Tuolumne River Basin above Hetch Hetchy Reservoir in the Sierra Nevada Mountains of California. The modeling component of the ASO program implements the iSnobal model to estimate snow density for converting measured depths to snow water equivalent and to provide temporally complete snow cover mass and thermal states between flights. Over the four years considered in this study, snow depths from 36 individual lidar flights were directly inserted into the model to provide updates of snow depth and distribution. Considering all updates to the model, the correlation between ASO depths and modeled depths with and without previous updates was on average r2 = 0.899 (root‐mean‐square error = 12.5 cm) and r2 = 0.162 (root‐mean‐square error = 41.5 cm), respectively. The precise definition of the snow depth distribution integrated with the iSnobal model demonstrates how the ASO program represents a new paradigm for the measurement and modeling of mountain snowpacks and reveals the potential benefits for managing water in the region.

In regions that depend primarily on snow to support life, water availability is becoming an increasingly important topic. National Aeronautics and Space Administration (NASA)'s Airborne Snow Observatory (ASO) is a new platform for estimating the amount of water stored in mountain snowpacks. Since 2013, the ASO has combined detailed measurements of snow depth from an aircraft with snowpack density estimates from a physics‐based snow model to provide predictions of total snow water equivalent stored in the Tuolumne River Basin in the California Sierra Nevada. This work describes the process of updating the snow model using the measured ASO snow depths through a direct insertion process. When the distribution of all the snow in the basin is known more accurately, the model results are improved.

Attention deficit hyperactivity disorder (ADHD) is a common and persistent condition characterized by developmentally atypical and impairing inattention, hyperactivity, and impulsiveness. We ...identified de novo and rare copy number variations (CNVs) in 248 unrelated ADHD patients using million-feature genotyping arrays. We found de novo CNVs in 3 of 173 (1.7%) ADHD patients for whom we had DNA from both parents. These CNVs affected brain-expressed genes: DCLK2, SORCS1, SORCS3, and MACROD2. We also detected rare inherited CNVs in 19 of 248 (7.7%) ADHD probands, which were absent in 2357 controls and which either overlapped previously implicated ADHD loci (for example, DRD5 and 15q13 microduplication) or identified new candidate susceptibility genes (ASTN2, CPLX2, ZBBX, and PTPRN2). Among these de novo and rare inherited CNVs, there were also examples of genes (ASTN2, GABRG1, and CNTN5) previously implicated by rare CNVs in other neurodevelopmental conditions including autism spectrum disorder (ASD). To further explore the overlap of risks in ADHD and ASD, we used the same microarrays to test for rare CNVs in an independent, newly collected cohort of 349 unrelated individuals with a primary diagnosis of ASD. Deletions of the neuronal ASTN2 and the ASTN2-intronic TRIM32 genes yielded the strongest association with ADHD and ASD, but numerous other shared candidate genes (such as CHCHD3, MACROD2, and the 16p11.2 region) were also revealed. Our results provide support for a role for rare CNVs in ADHD risk and reinforce evidence for the existence of common underlying susceptibility genes for ADHD, ASD, and other neuropsychiatric disorders.
Defaunation is causing declines of large-seeded animal-dispersed trees in tropical forests worldwide, but whether and how these declines will affect carbon storage across this biome is unclear. Here ...we show, using a pan-tropical data set, that simulated declines of large-seeded animal-dispersed trees have contrasting effects on aboveground carbon stocks across Earth's tropical forests. In our simulations, African, American and South Asian forests, which have high proportions of animal-dispersed species, consistently show carbon losses (2-12%), but Southeast Asian and Australian forests, where there are more abiotically dispersed species, show little to no carbon losses or marginal gains (±1%). These patterns result primarily from changes in wood volume, and are underlain by consistent relationships in our empirical data (∼2,100 species), wherein, large-seeded animal-dispersed species are larger as adults than small-seeded animal-dispersed species, but are smaller than abiotically dispersed species. Thus, floristic differences and distinct dispersal mode-seed size-adult size combinations can drive contrasting regional responses to defaunation.
Optical spectroscopic sensing is a technique that is commonly employed for the identification and compositional analysis of a wide variety of substances, from biological samples to greenhouse gases. ...High-resolution spectrometers are well established, however, attempts to miniaturise the designs can suffer from adverse effects due to the miniaturisation, for both Fourier transform based interferometric designs, as well as dispersive designs. In this work, a linear array of resonant cavity-enhanced photodiodes is realised with spatially chirped resonance wavelength, offering chip-scale free-space hyperspectral sensing. Resonant cavity-enhanced photodiodes sense over a narrow spectral band, which can be tuned by the thicknesses of the heterostructure. Through this work, multiple narrow spectral bands can be sensed by resonant cavity-enhanced photodiodes on a single chip by grading the thicknesses across the wafer. Photocurrent measurements from a fabricated array determine the wavelength of incident light with an accuracy of ± 2 nm.
Glaucoma, a disease characterized by progressive optic nerve degeneration, can be prevented through timely diagnosis and treatment. We characterize optic nerve photographs of 67,040 UK Biobank ...participants and use a multitrait genetic model to identify risk loci for glaucoma. A glaucoma polygenic risk score (PRS) enables effective risk stratification in unselected glaucoma cases and modifies penetrance of the MYOC variant encoding p.Gln368Ter, the most common glaucoma-associated myocilin variant. In the unselected glaucoma population, individuals in the top PRS decile reach an absolute risk for glaucoma 10 years earlier than the bottom decile and are at 15-fold increased risk of developing advanced glaucoma (top 10% versus remaining 90%, odds ratio = 4.20). The PRS predicts glaucoma progression in prospectively monitored, early manifest glaucoma cases (P = 0.004) and surgical intervention in advanced disease (P = 3.6 × 10
). This glaucoma PRS will facilitate the development of a personalized approach for earlier treatment of high-risk individuals, with less intensive monitoring and treatment being possible for lower-risk groups.
There has never been a more pressing and opportune time for science and practice to collaborate towards restoration of the world's forests. Multiple uncertainties remain for achieving successful, ...long-term forest landscape restoration (FLR). In this article, we use expert knowledge and literature review to identify knowledge gaps that need closing to advance restoration practice, as an introduction to a landmark theme issue on FLR and the UN Decade on Ecosystem Restoration. Aligned with an Adaptive Management Cycle for FLR, we identify 15 essential science advances required to facilitate FLR success for nature and people. They highlight that the greatest science challenges lie in the conceptualization, planning and assessment stages of restoration, which require an evidence base for why, where and how to restore, at realistic scales. FLR and underlying sciences are complex, requiring spatially explicit approaches across disciplines and sectors, considering multiple objectives, drivers and trade-offs critical for decision-making and financing. The developing tropics are a priority region, where scientists must work with stakeholders across the Adaptive Management Cycle. Clearly communicated scientific evidence for action at the outset of restoration planning will enable donors, decision makers and implementers to develop informed objectives, realistic targets and processes for accountability. This article paves the way for 19 further articles in this theme issue, with author contributions from across the world.
