DEC-205 (CD205) belongs to the macrophage mannose receptor family of C-type lectin endocytic receptors and behaves as an antigen uptake/processing receptor for dendritic cells (DC). To investigate ...DEC-205 tissue distribution in human leukocytes, we generated a series of anti-human DEC-205 monoclonal antibodies (MMRI-5, 6 and 7), which recognized epitopes within the C-type lectin-like domains 1 and 2, and the MMRI-7 immunoprecipitated a single ∼200 kDa band, identified as DEC-205 by mass spectrometry. MMRI-7 and another DEC-205 mAb (MG38), which recognized the epitope within the DEC-205 cysteine-rich and fibronectin type II domain, were used to examine DEC-205 expression by human leukocytes. Unlike mouse DEC-205, which is reported to have predominant expression on DC, human DEC-205 was detected by flow cytometry at relatively high levels on myeloid blood DC and monocytes, at moderate levels on B lymphocytes and at low levels on NK cells, plasmacytoid blood DC and T lymphocytes. MMRI-7 F(ab′)2 also labeled monocytes, B lymphocytes and NK cells similarly excluding reactivity due to non-specific binding of the mAb to FcγR. Tonsil mononuclear cells showed a similar distribution of DEC-205 staining on the leukocytes. DEC-205-specific semiquantitative immunoprecipitation/western blot and quantitative reverse transcriptase-PCR analysis established that these leukocyte populations expressed DEC-205 protein and the cognate mRNA. Thus, human DEC-205 is expressed on more leukocyte populations than that were previously assumed based on mouse DEC-205 tissue localization studies. The broader DEC-205 tissue expression in man is relevant to clinical DC targeting strategies and DEC-205 functional studies.

Here, we identify priority policy developments that would support conservation in the UK in the light of ...technological developments, changes in knowledge or environmental change. 2. A team of seven representatives from governmental organizations, 17 from non-governmental organizations and six academics provided an assessment of the priority issues. The representatives consulted widely and identified a long-list of 117 issues. 3. Following voting and discussion during a 2-day meeting, these were reduced to a final list of 25 issues and their potential policy options and research needs were identified. Many of the policies related to recent changes in approaches to conservation, such as increased interest in ecosystem services, adaptation to climate change and landscape ecology. 4. We anticipate that this paper will be useful for policy makers, nature conservation delivery agencies, the research community and conservation policy advocates. 5. Although many of the options have global significance, we suggest that other countries consider an equivalent exercise. We recommend that such an exercise be carried out in the UK at regular intervals, say every 5 years, to explore how biodiversity conservation can best be supported by linked policy development and research in a changing world. 6. Synthesis and applications. Opportunities for policy development were prioritized and for each of the top 25 we identified the current context, policy options and research questions. These largely addressed new issues relating to developing topics such as ecosystem services, landscape planning and nanotechnology. We envisage that this will largely be used by researchers wishing to make a contribution to potential policy debates.

Robert Bourassa, a pariah after losing power to the separatist Parti Québécois in the 1976 election, emerged a decade later from political exile to lead his party back to power. As he said: "I ...succeeded my successor." Claude Ryan, formerly the respected publisher of Le Devoir, had led the Quebec Liberal Party and the federalist coalition to a decisive victory in the 1980 referendum on Quebec sovereignty, but the uneasy alliance of Ryan and Pierre Trudeau did not survive the prime minister's unilateral patriation of the Canadian constitution. This contributed to Ryan's defeat in the 1981 Quebec election and to Bourassa's restoration.

AIM: To discuss the advantages of ultra-high field (7T) for 1H and 13C magnetic resonance spectroscopy (MRS) studies of metabolism.made at both 3 and 7T using 1H MRS. Measurements of glycogen and ...lipids in muscle were measured using 13C and 1H MRS respectively. RESULTS: In the brain, increased signal-to-noise ratio (SNR) and dispersion allows spectral separation of the amino-acids glutamate, glutamine and γ-aminobutyric acid (GABA), without the need for sophisticated editing sequences. Improved quantification of these me-tabolites is demonstrated at 7T relative to 3T. SNR was 36% higher, and measurement repeatability (% coefficients of variation) was 4%, 10% and 10% at 7T, vs 8%, 29% and 21% at 3T for glutamate, glutamine and GABA respectively. Measurements at 7T were used to compare metabolite levels in the anterior cingulate cortex (ACC) and insula. Creatine and glutamate levels were found to be significantly higher in the insula compared to the ACC (P < 0.05). In muscle, the increased SNR and spectral resolution at 7T enables interleaved studies of glycogen (13C) and intra-myocellular lipid (IMCL) and extra-myocellular lipid (EMCL) (1H) following exercise and refeeding. Glycogen levels were sig-nificantly decreased following exercise (-28% at 50% VO2 max; -58% at 75% VO2 max). Interestingly, levels of glycogen in the hamstrings followed those in the quadriceps, despite reduce exercise loading. No changes in IMCL and EMCL were found in the study. CONCLUSION: The demonstrated improvements in brain and muscle MRS measurements at 7T will increase the potential for use in investigating human metabolism and changes due to pathologies.
