Phosphatidylinositol 3-kinase (PI3K) promotes cancer cell survival, migration, growth and proliferation by generating phosphatidylinositol 3,4,5-trisphosphate (PIP3) in the inner leaflet of the ...plasma membrane. PIP3 recruits pleckstrin homology domain-containing proteins to the membrane to activate oncogenic signaling cascades. Anticancer therapeutics targeting the PI3K/AKT/mTOR (mammalian target of rapamycin) pathway are in clinical development. In a mass spectrometric screen to identify PIP3-regulated proteins in breast cancer cells, levels of the Rac activator PIP3-dependent Rac exchange factor-1 (P-REX1) increased in response to PI3K inhibition, and decreased upon loss of the PI3K antagonist phosphatase and tensin homolog (PTEN). P-REX1 mRNA and protein levels were positively correlated with ER expression, and inversely correlated with PI3K pathway activation in breast tumors as assessed by gene expression and phosphoproteomic analyses. P-REX1 increased activation of Rac1, PI3K/AKT and MEK/ERK signaling in a PTEN-independent manner, and promoted cell and tumor viability. Loss of P-REX1 or inhibition of Rac suppressed PI3K/AKT and MEK/ERK, and decreased viability. P-REX1 also promoted insulin-like growth factor-1 receptor activation, suggesting that P-REX1 provides positive feedback to activators upstream of PI3K. In support of a model where PIP3-driven P-REX1 promotes both PI3K/AKT and MEK/ERK signaling, high levels of P-REX1 mRNA (but not phospho-AKT or a transcriptomic signature of PI3K activation) were predictive of sensitivity to PI3K inhibitors among breast cancer cell lines. P-REX1 expression was highest in estrogen receptor-positive breast tumors compared with many other cancer subtypes, suggesting that neutralizing the P-REX1/Rac axis may provide a novel therapeutic approach to selectively abrogate oncogenic signaling in breast cancer cells.
We study the stellar, brightest cluster galaxy (BCG) and intracluster medium (ICM) masses of 14 South Pole Telescope (SPT) selected galaxy clusters with median redshift z = 0.9 and mass M
...500 = 6 × 1014 M⊙. We estimate stellar masses for each cluster and BCG using six photometric bands, the ICM mass using X-ray observations and the virial masses using the SPT Sunyaev–Zel'dovich effect signature. At z = 0.9, the BCG mass

constitutes 0.12 ± 0.01 per cent of the halo mass for a 6 × 1014 M⊙ cluster, and this fraction falls as

The ecology of Bornean rainforests is driven by El Niño-induced droughts that trigger synchronous fruiting among trees and bursts of faunal reproduction that sustain vertebrate populations. However, ...many of these species- and carbon-rich ecosystems have been destroyed by logging and conversion, which increasingly threaten protected areas. Our satellite, Geographic Information System, and field-based analyses show that from 1985 to 2001, Kalimantan's protected lowland forests declined by more than 56% (>29,000 square kilometers). Even uninhabited frontier parks are logged to supply international markets. "Protected" forests have become increasingly isolated and deforested and their buffer zones degraded. Preserving the ecological integrity of Kalimantan's rainforests requires immediate transnational management.

We derive the fundamental parameters (temperature and luminosity) of 107 619 Hipparcos stars and place these stars on a true Hertzsprung–Russell diagram. This is achieved by comparing ...bt‐settl model atmospheres to spectral energy distributions (SEDs) created from Hipparcos, Tycho, Sloan Digital Sky Survey, DENIS, Two Micron All Sky Survey, MSX, AKARI, IRAS and Wide‐field Infrared Survey Explorer data. We also identify and quantify from these SEDs any infrared excesses attributable to circumstellar matter. We compare our results to known types of objects, focusing on the giant branch stars. Giant star dust production (as traced by infrared excess) is found to start in earnest around 680 L⊙.
Sister chromatids are tethered together by the cohesin complex from the time they are made until their separation at anaphase. The ability of cohesin to tether sister chromatids together depends on ...acetylation of its Smc3 subunit by members of the Eco1 family of cohesin acetyltransferases. Vertebrates express two orthologs of Eco1, called Esco1 and Esco2, both of which are capable of modifying Smc3, but their relative contributions to sister chromatid cohesion are unknown. We therefore set out to determine the precise contributions of Esco1 and Esco2 to cohesion in vertebrate cells. Herewe show that cohesion establishment is critically dependent upon Esco2. Although most Smc3 acetylation is Esco1 dependent, inactivation of the ESCO1 gene has little effect on mitotic cohesion. The unique ability of Esco2 to promote cohesion is mediated by sequences in the N terminus of the protein. We propose that Esco1-dependent modification of Smc3 regulates almost exclusively the noncohesive activities of cohesin, such as DNA repair, transcriptional control, chromosome loop formation, and/or stabilization. Collectively, our data indicate that Esco1 and Esco2 contribute to distinct and separable activities of cohesin in vertebrate cells.
We present a catalog of galaxy clusters selected via their Sunyaev-Zel'dovich (SZ) effect signature from 2500 deg super(2) of South Pole Telescope (SPT) data. This work represents the complete sample ...of clusters detected at high significance in the 2500 deg super(2) SPT-SZ survey, which was completed in 2011. A total of 677 (409) cluster candidates are identified above a signal-to-noise threshold of xi = 4.5 (5.0). Ground- and space-based optical and near-infrared (NIR) imaging confirms overdensities of similarly colored galaxies in the direction of 516 (or 76%) of the xi > 4.5 candidates and 387 (or 95%) of the xi > 5 candidates; the measured purity is consistent with expectations from simulations. Of these confirmed clusters, 415 were first identified in SPT data, including 251 new discoveries reported in this work. We estimate photometric redshifts for all candidates with identified optical and/or NIR counterparts; we additionally report redshifts derived from spectroscopic observations for 141 of these systems. The mass threshold of the catalog is roughly independent of redshift above z ~ 0.25 leading to a sample of massive clusters that extends to high redshift. The median mass of the sample is M sub(500c)(rho sub(crit)) ~3.5 x 10 super(14) M sub(middot in circle) h super(-1) sub(70), the median redshift is z sub(med) = 0.55, and the highest-redshift systems are at z > 1.4. The combination of large redshift extent, clean selection, and high typical mass makes this cluster sample of particular interest for cosmological analyses and studies of cluster formation and evolution.
ABSTRACT
We search for the signature of cosmological shocks in stacked gas pressure profiles of galaxy clusters using data from the South Pole Telescope (SPT). Specifically, we stack the latest ...Compton-y maps from the 2500 deg2 SPT-SZ survey on the locations of clusters identified in that same data set. The sample contains 516 clusters with mean mass $\langle M_{\rm 200m}\rangle = 10^{14.9} \, {\rm M}_\odot$ and redshift 〈z〉 = 0.55. We analyse in parallel a set of zoom-in hydrodynamical simulations from the three hundred project. The SPT-SZ data show two features: (i) a pressure deficit at R/R200m = 1.08 ± 0.09, measured at 3.1σ significance and not observed in the simulations, and; (ii) a sharp decrease in pressure at R/R200m = 4.58 ± 1.24 at 2.0σ significance. The pressure deficit is qualitatively consistent with a shock-induced thermal non-equilibrium between electrons and ions, and the second feature is consistent with accretion shocks seen in previous studies. We split the cluster sample by redshift and mass, and find both features exist in all cases. There are also no significant differences in features along and across the cluster major axis, whose orientation roughly points towards filamentary structure. As a consistency test, we also analyse clusters from the Planck and Atacama Cosmology Telescope Polarimeter surveys and find quantitatively similar features in the pressure profiles. Finally, we compare the accretion shock radius ($R_{\rm sh,\, acc}$) with existing measurements of the splashback radius (Rsp) for SPT-SZ and constrain the lower limit of the ratio, $R_{\rm sh,\, acc}/R_{\rm sp}\gt 2.16 \pm 0.59$.
ABSTRACT X-ray cavities are key tracers of mechanical (or radio mode) heating arising from the active galactic nuclei (AGNs) in brightest cluster galaxies (BCGs). We report on a survey for X-ray ...cavities in 83 massive, high-redshift ( ) clusters of galaxies selected by their Sunyaev-Zel'dovich signature in the South Pole Telescope data. Based on Chandra X-ray images, we find a total of six clusters having symmetric pairs of surface brightness depressions consistent with the picture of radio jets inflating X-ray cavities in the intracluster medium (ICM). The majority of these detections are of relatively low significance and require deeper follow-up data in order to be confirmed. Further, this search will miss small (<10 kpc) X-ray cavities that are unresolved by Chandra at high ( ) redshift. Despite these limitations, our results suggest that the power generated by AGN feedback in BCGs has remained unchanged for over half of the age of the universe ( Gyr at ). On average, the detected X-ray cavities have powers of , enthalpies of , and radii of ∼17 kpc. Integrating over 7 Gyr, we find that the supermassive black holes in BCGs may have accreted 108 to several of material to power these outflows. This level of accretion indicates that significant supermassive black hole growth may occur not only at early times, in the quasar era, but at late times as well. We also find that X-ray cavities at high redshift may inject an excess heat of 0.1-1.0 keV per particle into the hot ICM above and beyond the energy needed to offset cooling. Although this result needs to be confirmed, we note that the magnitude of excess heating is similar to the energy needed to preheat clusters, break self-similarity, and explain the excess entropy in hot atmospheres.
The Amazon River floodplain is subject to large seasonal variations in water level and flood extent, due to the large size and low relief of the basin, and the large amount of precipitation in the ...region. Synthetic Aperture Radar (SAR) data can be used to map flooded area in these wetlands, given its ability to provide continuous information without being heavily affected by cloud cover. As part of JAXA's Kyoto & Carbon Initiative, extensive wide-swath, multi-temporal SAR coverage of the Amazon basin has been obtained using the ScanSAR mode of ALOS PALSAR. This study presents a method for monitoring flood extent variation using ALOS ScanSAR images, tested at the Curuai Lake floodplain, in the lower Amazon River, Brazil. Twelve ScanSAR scenes were acquired between 2006 and 2010, including seven during the 2007 hydrological year. Water level records, field photographs, optical images (Landsat-5/TM and MODIS/Terra and Aqua) and topographic data were used as auxiliary information. A data mining algorithm allowed the implementation of a hierarchical, object-based classification algorithm, able to map land cover types and flooding status in the study area for all available dates. Land cover based on the entire time series (classification levels 1 and 2) had overall accuracies of 90% and 83%, respectively. Level 3 classifications (one map per image date) were validated only for the lowest and highest water stages, with overall accuracies of 76% and 78%, respectively. Total flood extent (Level 4) was mapped with 84% and 94% accuracies, for the low and high water stages, respectively. Regression models were fitted between mapped flooded area and water levels at the Curuai gauge to predict flood extent. A polynomial model had R2=0.95 (p<0.05) and an overall root mean square error (RMSE) of 241km2, while a logistic model had R2=0.98 (p<0.05) and RMSE=127km2.
► Flood extent was monitored from 2007 to 2010 for a lake in the Lower Amazon Floodplain. ► PALSAR ScanSAR, TM and MODIS images were combined using object-based image analysis. ► Final mapping accuracy was ~76% for land cover and ~90% for flood status. ► Logistic models predicted flooded area better than polynomial or simple regressions. ► The present method could be used to monitor flood extent for the entire Amazon.
We estimate the total dust input from the cool evolved stars in the Small Magellanic Cloud, using the 8 mu m excess emission as a proxy for the dust-production rate (DPR). We find that asymptotic ...giant branch (AGB) and red supergiant (RSG) stars produce (8.6-9.5) x 10 super(-7) M sub(middot in circle) yr super(-1) of dust, depending on the fraction of far-infrared sources that belong to the evolved star population (with 10%-50% uncertainty in individual DPRs). RSGs contribute the least (<4%), while carbon-rich AGB stars (especially the so-called extreme AGB stars) account for 87%-89% of the total dust input from cool evolved stars. We also estimate the dust input from hot stars and supernovae (SNe), and find that if SNe produce 10 super(-3) M sub(middot in circle) of dust each, then the total SN dust input and AGB input are roughly equivalent. We consider several scenarios of SN dust production and destruction and find that the interstellar medium (ISM) dust can be accounted for solely by stellar sources if all SNe produce dust in the quantities seen around the dustiest examples and if most SNe explode in dense regions where much of the ISM dust is shielded from the shocks. We find that AGB stars contribute only 2.1% of the ISM dust. Without a net positive contribution from SNe to the dust budget, this suggests that dust must grow in the ISM or be formed by another unknown mechanism.
