Circular dichroism (CD) spectroscopy is a highly used method for the examination and characterization of proteins, including, amongst other features, their secondary and tertiary structures, thermal ...stability, comparisons of wildtype and mutant proteins, and monitoring the binding of small molecules, folding/unfolding pathways, and formation of macromolecular complexes. This article describes CDtoolX, a new, user‐friendly, free‐to‐download‐and‐use software program that enables processing, displaying, archiving, calibrating, comparisons, and analyses of CD and synchrotron radiation circular dichroism spectroscopic data.
The COVID-19 pandemic, the accompanying lockdown measures, and their possible long-term effects have made mental health a pressing public health concern. Acts that focus on benefiting others-known as ...prosocial behaviors-offer one promising intervention that is both flexible and low cost. However, neither the range of emotional states prosocial acts impact nor the size of those effects is currently clear, both of which directly influence its attractiveness as a treatment option. Using a large online sample from Canada and the United States, we will examine the effect of a three-week prosocial intervention on two indicators of emotional well-being (happiness and the belief that one's life is valuable) and mental health (anxiety and depression). Respondents will be randomly assigned to perform prosocial, self-focused, or neutral behaviors each week. Two weeks after the intervention, a final survey will assess whether the intervention has a lasting effect on mental health and emotional well-being. Our results will illuminate whether prosocial interventions are a viable approach to addressing mental health needs during the current COVID-19 pandemic, as well for those who face emotional challenges during normal times.
The social scientific analysis of social class is attracting renewed interest given the accentuation of economic and social inequalities throughout the world. The most widely validated measure of ...social class, the Nuffield class schema, developed in the 1970s, was codified in the UK's National Statistics Socio-Economic Classification (NS-SEC) and places people in one of seven main classes according to their occupation and employment status. This principally distinguishes between people working in routine or semi-routine occupations employed on a 'labour contract' on the one hand, and those working in professional or managerial occupations employed on a 'service contract' on the other. However, this occupationally based class schema does not effectively capture the role of social and cultural processes in generating class divisions. We analyse the largest survey of social class ever conducted in the UK, the BBC's 2011 Great British Class Survey, with 161,400 web respondents, as well as a nationally representative sample survey, which includes unusually detailed questions asked on social, cultural and economic capital. Using latent class analysis on these variables, we derive seven classes. We demonstrate the existence of an 'elite', whose wealth separates them from an established middle class, as well as a class of technical experts and a class of 'new affluent' workers. We also show that at the lower levels of the class structure, alongside an ageing traditional working class, there is a 'precariat' characterised by very low levels of capital, and a group of emergent service workers. We think that this new seven class model recognises both social polarisation in British society and class fragmentation in its middle layers, and will attract enormous interest from a wide social scientific community in offering an up-to-date multi-dimensional model of social class.
Circular dichroism (CD) spectroscopy is a valuable method for defining canonical secondary structure contents of proteins based on empirically‐defined spectroscopic signatures derived from proteins ...with known three‐dimensional structures. Many proteins identified as being “Intrinsically Disordered Proteins” have a significant amount of their structure that is neither sheet, helix, nor turn; this type of structure is often classified by CD as “other”, “random coil”, “unordered”, or “disordered”. However the “other” category can also include polyproline II (PPII)‐type structures, whose spectral properties have not been well‐distinguished from those of unordered structures. In this study, synchrotron radiation circular dichroism spectroscopy was used to investigate the spectral properties of collagen and polyproline, which both contain PPII‐type structures. Their native spectra were compared as representatives of PPII structures. In addition, their spectra before and after treatment with various conditions to produce unfolded or denatured structures were also compared, with the aim of defining the differences between CD spectra of PPII and disordered structures. We conclude that the spectral features of collagen are more appropriate than those of polyproline for use as the representative spectrum for PPII structures present in typical amino acid‐containing proteins, and that the single most characteristic spectroscopic feature distinguishing a PPII structure from a disordered structure is the presence of a positive peak around 220nm in the former but not in the latter. These spectra are now available for inclusion in new reference data sets used for CD analyses of the secondary structures of soluble proteins.
This paper examines the impact of false data injection (FDI) cyber-attacks on distribution system state estimation (DSSE) and the resulting distribution locational marginal price (DLMP) in power ...markets. Two robust high-breakdown regression estimators, namely S- and MM- estimators, are implemented to provide resistance against FDI attacks targeting measurements and grid topology, creating leverage points. The introduced estimators are compared to the weighted least squares (WLS) with a bad data detection and rejection module (BDD) and the robust Huber M-estimator. The proposed estimators are shown to be effective and compare favorably to both existing Huber M- and the WLS with BDD in the presence of topology FDI attacks. Both the S- and MM-estimators provide good performance in the case of clean and corrupted measurements. Their performance is comparable in this case to the Huber M- and the WLS, followed by a BDD module. The simulation considered a modified distribution IEEE 13 and 34-bus systems where the impact of FDI attack scenarios is shown on the state and the DLMP pricing in the presence of distributed generation.

To assess the effect of prosocial activity on emotional well-being (happiness, belief that one's life is valuable) and mental health (anxiety, depression). 1,234 respondents from the United States and Canada were recruited from Amazon's Mechanical Turk and randomly assigned (by computer software) to perform prosocial (N = 411), self-focused (N = 423), or neutral (N = 400) behaviors three times a week for three weeks. A follow-up assessment was given two weeks after the intervention. Participants were blind to alternative conditions. Analyses were based on 1052 participants (N.sub.prosocial = 347, N.sub.self = 365, N.sub.neutral = 340). Those in the prosocial condition did not differ on any outcome from those in the self-focused or neutral acts conditions during the intervention or at follow-up, nor did prosocial effects differ for those who had been negatively affected socially or economically by the pandemic (all p's > 0.05). Exploratory analyses that more tightly controlled for study compliance found that prosocial acts reduced anxiety relative to neutral acts control (beta = -0.12 95% CI: -0.22 to -0.02) and increased the belief that one's life is valuable (beta = 0.11 95% CI: 0.03 to 0.19). These effects persisted throughout the intervention and at follow-up. Prosocial acts may provide small, lasting benefits to emotional well-being and mental health. Future work should replicate these results using tighter, pre-registered controls on study compliance.
Dual-process models of culture and action posit that fast, automatic cognitive processes largely drive human action, with conscious processes playing a much smaller role than was previously supposed. ...These models have done much to advance our understanding of behavior, but they focus on generic processes rather than specific cultural content. As useful as this has been, it tells us little about which forms of culture matter for action. Drawing on a cross-disciplinary set of theory and evidence, I argue that values are tied to many forms of behavior, across both contexts and cultures, and they operate in ways consistent with dual-process models. I illustrate the plausibility of these claims using data from the second wave of the European Social Survey, as well as real-time decision data from a large, online survey. I show that values predict self-reported behaviors in a variety of substantive domains and across 25 nations, and they operate using automatic cognitive processes. These findings suggest that values merit renewed theoretical and empirical attention.
Moral culture can mean many things, but two major elements are a concern with moral goods and moral prohibitions. Moral psychologists have developed instruments for assessing both of these and such ...measures can be directly imported by sociologists. Work by Schwartz and his colleagues on values offers a well-established way of measuring moral goods, while researchers using Haidt's Moral Foundations Theory have developed validated measures of moral prohibitions. Both values and moral foundations are distributed across the social landscape in systematic, sociologically interesting ways. Although typically measured using questionnaires, we show that values and moral foundations also can be used to analyze interview, archival, or "big data." Combining psychological and sociological tools and frameworks promises to clarify relations among existing sociological treatments of moral culture and to connect such treatments to a thriving conversation in moral psychology.
Intrinsically disordered proteins (IDPs) are comprised of significant numbers of residues that form neither helix, sheet, nor any other canonical type of secondary structure. They play important ...roles in a broad range of biological processes, such as molecular recognition and signalling, largely due to their chameleon-like ability to change structure from unordered when free in solution to ordered when bound to partner molecules. Circular dichroism (CD) spectroscopy is a widely-used method for characterising protein secondary structures, but analyses of IDPs using CD spectroscopy have suffered because the methods and reference datasets used for the empirical determination of secondary structures do not contain adequate representations of unordered structures. This work describes the creation, validation and testing of a standalone Windows-based application, DichroIDP, and a new reference dataset, IDP175, which is suitable for analyses of proteins containing significant amounts of disordered structure. DichroIDP enables secondary structure determinations of IDPs and proteins containing intrinsically disordered regions.
