This study evaluates the potential of Boston Ivy (Parthenocissus tricuspidata) to reduce building surface temperature in a mid-latitude North American city center where vine use for this purpose is ...uncommon. Vegetation can regulate city summer temperatures by providing shade and evaporative cooling. While planting trees has been a focus for many urban municipalities, trees require space (above and below ground), access to water, costly planting and maintenance, and may only be desirable to some city residents. To explore viable vegetation alternatives with fewer growth constraints, we deployed temperature loggers on the exterior walls of buildings in the urban core of Toronto, Canada, a large mid-latitude city. Perennial vines shaded some walls, while others were bare. These devices systematically tracked exterior surface temperature fluctuations over six months, including the growing season, with full vine-leaf coverage. During peak solar access periods, average daily temperature differentials between vine-shaded and non-shaded building surfaces ranged from up to 6.5 °C on south-facing building exteriors to 7.0 °C on west-facing walls. Models were developed to estimate daily degree hour difference, a metric integrating the magnitude and duration of the temperature-moderating potential of vines. At ambient temperatures ≥ 23 °C, solar radiation intensity and ambient air temperature were positively correlated with vine effectiveness in mitigating the rise in built surface temperature; relative humidity was negatively associated. Installing vine cover on urban buildings in the form of green façades can complement tree planting as cities become hotter due to climate change, and space for growing trees diminishes with urban densification. Future research into the capacity of green façades to regulate outdoor temperature must establish uniform measurement protocols and undertake evaluations in diverse climatic scenarios.

The objective of this study was development of accelerated in vitro release method for peptide loaded PLGA microspheres using flow-through apparatus and assessment of the effect of ...dissolution parameters (pH, temperature, medium composition) on drug release rate and mechanism. Accelerated release conditions were set as pH 2 and 45°C, in phosphate buffer saline (PBS) 0.02M. When the pH was changed from 2 to 4, diffusion controlled phases (burst and lag) were not affected, while release rate during erosion phase decreased two-fold due to slower ester bonds hydrolyses. Decreasing temperature from 45°C to 40°C, release rate showed three-fold deceleration without significant change in release mechanism. Effect of medium composition on drug release was tested in PBS 0.01M (200 mOsm/kg) and PBS 0.01M with glucose (380 mOsm/kg). Buffer concentration significantly affected drug release rate and mechanism due to the change in osmotic pressure, while ionic strength did not have any effect on peptide release. Furthermore, dialysis sac and sample-and-separate techniques were used, in order to evaluate significance of dissolution technique choice on the release process. After fitting obtained data to different mathematical models, flow-through method was confirmed as the most appropriate for accelerated in vitro dissolution testing for a given formulation.
Many protein kinases themselves are regulated by reversible phosphorylation. Upon cell stimulation, specific kinases are transiently phosphorylated and activated. Several of these protein kinases are ...substrates for protein phosphatase 2A (PP2A), and PP2A appears to be the major kinase phosphatase in eukaryotic cells that downregulates activated protein kinases. This idea is substantiated by the observation that some viral proteins and naturally occurring toxins target PP2A and modulate its activity. There is increasing evidence that PP2A activity is regulated by extracellular signals and during the cell cycle. Thus, PP2A is likely to play an important role in determining the activation kinetics of protein kinase cascades.
Metformin might reduce insulin requirement and improve glycaemia in patients with type 1 diabetes, but whether it has cardiovascular benefits is unknown. We aimed to investigate whether metformin ...treatment (added to titrated insulin therapy) reduced atherosclerosis, as measured by progression of common carotid artery intima-media thickness (cIMT), in adults with type 1 diabetes at increased risk for cardiovascular disease.
REMOVAL was a double-blind, placebo-controlled trial undertaken at 23 hospital diabetes clinics in five countries (Australia, Canada, Denmark, the Netherlands, and the UK). Adults aged 40 years and older with type 1 diabetes of at least 5 years' duration and at least three of ten specific cardiovascular risk factors were randomly assigned (via an interactive voice response system) to oral metformin 1000 mg twice daily or placebo. Participants and site staff were masked to treatment allocation. The primary outcome was averaged mean far-wall cIMT, quantified annually for 3 years, analysed in a modified intention-to-treat population (all randomly assigned participants with post-randomisation data available for the outcome of interest at any given timepoint, irrespective of subsequent adherence or study participation), using repeated measures regression. , LDL cholesterol, estimated glomerular filtration rate (eGFR), incident microalbuminuria (not reported), incident retinopathy, bodyweight, insulin dose, and endothelial function, also analysed in all participants with post-randomisation data available for the outcome of interest at any given timepoint. Between Dec 14, 2011, and June 24, 2014, 493 participants entered a 3 month run-in to optimise risk factor and glycaemic control (single-blind placebo in the final month). Of 428 randomly assigned patients, 219 were allocated to metformin and 209 to placebo. Progression of mean cIMT was not significantly reduced with metformin (-0·005 mm per year, 95% CI -0·012 to 0·002; p=0·1664), although maximal cIMT (a prespecified tertiary outcome) was significantly reduced (-0·013 mm per year, -0·024 to -0·003; p=0·0093). HbA
(mean 8·1% SD 0·9 for metformin and 8·0% 0·8 for placebo at baseline) was reduced on average over 3 years by metformin (-0·13%, 95% CI -0·22 to -0·037; p=0·0060), but this was accounted for by a reduction at the 3-month timepoint (-0·24%, -0·34 to -0·13; p<0·0001) that was not sustained thereafter (p=0·0163 for visit-by-treatment interaction). Bodyweight (-1·17 kg, 95% CI -1·66 to -0·69; p<0·0001) and LDL cholesterol (-0·13 mmol/L, -0·24 to -0·03; p=0·0117) were reduced with metformin over 3 years of treatment, and eGFR was increased (4·0 mL/min per 1·73m
, 2·19 to 5·82; p<0·0001). Insulin requirement was not reduced on average over 3 years (-0·005 units per kg, 95% CI -0·022 to 0·012; p=0·545), but there was a significant visit-by-treatment interaction (p=0·0018). There was no effect on endothelial function as measured by reactive hyperaemia index, or on retinopathy. Discontinuation of treatment in 59 (27%) participants on metformin versus 26 (12%) on placebo (p=0·0002) was mainly due to an excess of gastrointestinal adverse effects, and there was no increase in hypoglycaemia with metformin. Five deaths occurred among patients allocated to metformin and two occurred among those allocated to placebo; none were judged by site principal investigators to be related to study medication.
These data do not support use of metformin to improve glycaemic control in adults with long-standing type 1 diabetes as suggested by current guidelines, but suggest that it might have a wider role in cardiovascular risk management.

This research integrates the Revised Universal Soil Loss Equation (RUSLE) with a Geographic Information System (GIS) to model erosion potential for soil conservation planning within the Sierra de ...Manantlán Biosphere Reserve (SMBR), Mexico. Mountainous topography and a tropical uni-modal precipitation regime characterize this region. These unique climatic and topographic characteristics required a modification of the standard RUSLE factors and their derivation. The resulting RUSLE–GIS model provides a robust soil conservation planning tool readily transferable and accessible to other land managers in similar environments. Future pressure to expand agriculture and grazing operations within the SMBR will unquestionably accentuate the already high rate of soil erosion and resultant sediment loading of watercourses occurring in this region. Until recently there did not exist a reliable or financially viable means to model and map soil erosion within large remote areas. An increase in the reliability and resolution of remote sensing techniques, modifications and advancements in watershed scale soil erosion modelling techniques, and advances in GIS, represent significantly improved tools that can be applied to both monitoring and modelling the effects of land use on soil erosion potential. Data used in this study to generate the RUSLE variables include a Landsat Thematic Mapper image (land cover), digitized topographic and soil maps, and tabular precipitation data. Soil erosion potential was modelled within Zenzontla, a sub-catchment of the Rı́o Ayuquı́la, located in the SMBR, and the results are presented as geo-referenced maps for each of the wet and dry precipitation seasons. These maps confirm that high and extreme areas of soil loss occur within the Zenzontla sub-catchment, and that erosion potential differs significantly between wet and dry seasons.

The Chinese Communist Party (CCP) policies in Xinjiang since 2017 have imprisoned some 300,000 and interned approximately a million more indigenous non-Han central Asians in educational ...transformation camps; razed or damaged thousands of mosques, shrines, and old neighborhoods; actively suppressed indigenous birth rates far below Han levels; illegalized core elements of Uyghur and Islamic culture; and pushed tens of thousands of non-Han adults into forced factory labor and children into state institutions. The revelation of these policies has contributed to the worst crisis for foreign policy in the People’s Republic of China (PRC) since 1989. Moreover, because the CCP requires declarations of loyal support from citizens and officials for its Xinjiang policies, this also comprises a Cultural Revolution–type domestic crisis in which no one can say no to a dictatorial leader despite the clearly destructive nature of that leader’s policies.

Polycystic ovary syndrome (PCOS) is a risk factor for Type 2 diabetes mellitus (T2DM) and cardiovascular disease (CVD), but these risks are poorly defined. This study ...aimed to evaluate the evidence for these risks and whether screening and risk reduction are feasible. Medline reviews and data quality analysis were used using standard tools. Results showed that (i) polycystic ovary syndrome is a risk factor forT2DM but the magnitude of risk is uncertain, (ii) fasting plasma glucose is an inadequate screening test forT2DM in this population and the oral glucose tolerance test is superior, (iii) the identification of women with PCOS for diabetes screening is constrained by current diagnostic criteria for PCOS; however, women with oligomenorrhoea and those with diagnosed PCOS and obesity or a family history of T2DM are at highest risk, (iv) risk factors for T2DM are improved by weight loss interventions and by metformin. However, no studies have determined whether T2DM incidence is reduced, (v) polycystic ovary syndrome is associated with cardiovascular disease (CVD) risk factors but data on CVD incidence are weak, (vi) risk factors for CVD are improved by the same interventions and statins and (vi) no studies have evaluated whether CVD incidence is reduced. While PCOS has important metabolic associations, and short‐term interventions reduce risk factors for T2DM and CVD, data on prevalence and incidence of T2DM and particularly CVD are poor. There is a need for a clear definition of PCOS, for diabetes screening protocols and for long‐term studies to determine whether risks can be reduced.

Our goal is to present a framework for collection and analysis of baseline data that can inform a management strategy that would serve to protect and enhance this significant natural asset. We found that Allan Garden’s tree population, while species rich (43), is dominated by maple (
Acer spp.) (48% of all park trees), making it reliant on very few species for the majority of its ecological and aesthetic benefits and raising disease and pest-related concerns. Age profiles (using size as a proxy) showed a dominance of older trees with an inadequate number of individuals in the young to early middle age cohort necessary for short- to medium-term replacement. Because leaf area represents the single-most important contributor to urban tree benefits modelling, we calculated it separately for every park tree, using hemispheric photography, to document current canopy condition. These empirical measurements were lower than estimates produced by STRATUM, especially when trees were in decline and lacked full canopies, highlighting the importance of individual tree condition in determining leaf area and hence overall forest benefits. Stewardship of natural spaces within cities demands access to accurate and timely resource-specific data. Our work provides an uncomplicated approach to the acquisition and interpretation of these data in the context of a forested urban park.

GLIS3 (GLI-similar 3) is a member of the GLI-similar zinc finger protein family encoding for a nuclear protein with 5 C2H2-type zinc finger domains. The protein is expressed early in ...embryogenesis and plays a critical role as both a repressor and activator of transcription. Human GLIS3 mutations are extremely rare.
Objective:
The purpose of this article was determine the phenotypic presentation of 12 patients with a variety of GLIS3 mutations.
Methods:
GLIS3 gene mutations were sought by PCR amplification and sequence analysis of exons 1 to 11. Clinical information was provided by the referring clinicians and subsequently using a questionnaire circulated to gain further information.
Results:
We report the first case of a patient with a compound heterozygous mutation in GLIS3 who did not present with congenital hypothyroidism. All patients presented with neonatal diabetes with a range of insulin sensitivities. Thyroid disease varied among patients. Hepatic and renal disease was common with liver dysfunction ranging from hepatitis to cirrhosis; cystic dysplasia was the most common renal manifestation. We describe new presenting features in patients with GLIS3 mutations, including craniosynostosis, hiatus hernia, atrial septal defect, splenic cyst, and choanal atresia and confirm further cases with sensorineural deafness and exocrine pancreatic insufficiency.
Conclusion:
We report new findings within the GLIS3 phenotype, further extending the spectrum of abnormalities associated with GLIS3 mutations and providing novel insights into the role of GLIS3 in human physiological development. All but 2 of the patients within our cohort are still alive, and we describe the first patient to live to adulthood with a GLIS3 mutation, suggesting that even patients with a severe GLIS3 phenotype may have a longer life expectancy than originally described.
