Soluble CD163 (sCD163) was recently identified as a strong risk marker for developing type 2 diabetes. We hypothesised that sCD163 independently associates with insulin resistance.
...Methods
This cross-sectional study includes 234 participants: 96 with type 2 diabetes, 34 with impaired glucose tolerance (IGT) and 104 with normal glucose tolerance (NGT), matched for sex and BMI. Glucose-lowering medication was paused for 1 week before plasma samples were obtained for determination of sCD163 and other inflammatory and metabolic variables. Insulin resistance was estimated by homeostasis model assessment of insulin resistance (HOMA-IR).
Results
Concentrations of sCD163 were 1.95 mg/l (0.63–6.97) in individuals with type 2 diabetes, 1.64 mg/l (0.58–4.19) in those with IGT, and 1.48 mg/l (0.48–4.11) (median range) in those with NGT (

< 0.0001). In univariate analyses, sCD163 correlated significantly with HOMA-IR (

= 0.44), insulin (
R
= 0.41), glucose (
R
= 0.30), triacylglycerol (
R
= 0.29) and HDL-cholesterol (
R
= −0.34) (all

< 0.0001). All but glucose remained significant when adjusting for age, sex, BMI and glycaemic group. In univariate regression analyses, HOMA-IR was associated with sCD163, C-reactive protein (CRP), TNF-α and IL-6 (all
p
≤ 0.0001). An increase of 50% in sCD163 resulted in an estimated increase in HOMA-IR of 36% (95% CI 26, 48;
p
< 0.0001). In multiple linear regression analyses, sCD163 (
p
= 0.001) and CRP (
p
= 0.01) remained independent predictors of HOMA-IR, whereas TNF-α and IL-6 did not.
Conclusions/interpretation
Macrophage-specific sCD163 was strongly associated with insulin resistance independently of TNF-α and other predictors. Moreover, sCD163 was associated with well-known variables of the metabolic syndrome.
Self-organization in many solution-processed, semiconducting conjugated polymers results in complex microstructures, in which ordered microcrystalline domains are embedded in an amorphous matrix. ...This has important consequences for electrical properties of these materials: charge transport is usually limited by the most difficult hopping processes and is therefore dominated by the disordered matrix, resulting in low charge-carrier mobilities ( 10-5 cm2 V-1 s-1). Here we use thin-film, field-effect transistor structures to probe the transport properties of the ordered microcrystalline domains in the conjugated polymer poly(3-hexylthiophene), P3HT. Self-organization in P3HT results in a lamella structure with two-dimensional conjugated sheets formed by interchain stacking. We find that, depending on processing conditions, the lamellae can adopt two different orientations-parallel and normal to the substrate-the mobilities of which differ by more than a factor of 100, and can reach values as high as 0.1 cm2 V-1 s-1 (refs 3, 4). Optical spectroscopy of the field-induced charge, combined with the mobility anisotropy, reveals the two-dimensional interchain character of the polaronic charge carriers, which exhibit lower relaxation energies than the corresponding radical cations on isolated one-dimensional chains. The possibility of achieving high mobilities via two-dimensional transport in self-organized conjugated lamellae is important for applications of polymer transistors in logic circuits and active-matrix displays.
To compare 2-year outcomes of total knee replacement (TKR) followed by non-surgical treatment to that of non-surgical treatment alone and outcomes of the same non-surgical treatment to that of ...written advice.
In two randomized trials, 200 (mean age 66) adults with moderate to severe knee osteoarthritis (OA), 100 eligible for TKR and 100 not eligible for TKR, were randomized to TKR followed by non-surgical treatment, non-surgical treatment alone, or written advice. Non-surgical treatment consisted of 12 weeks of supervised exercise, education, dietary advice, use of insoles, and pain medication. The primary outcome was the mean score of the Knee Injury and Osteoarthritis Outcome Score (KOOS) subscales, covering pain, symptoms, activities of daily living (ADL), and quality of life (QOL).
Patients randomized to TKR had greater improvements than patients randomized to non-surgical treatment alone (difference of 18.3 points (95% CI; 11.3 to 25.3)), who in turn improved more than patients randomized to written advice (difference of 7.0 points (95% CI; 0.4 to 13.5)). Among patients eligible for TKR, 16 (32%) from the non-surgical group underwent TKR during 2 years and among those initially ineligible, seven patients (14%) from the non-surgical group and ten (20%) from the written advice group underwent TKR.
TKR followed by non-surgical treatment is more effective on pain and function than non-surgical treatment alone, which in turn is more effective than written advice. Two out of three patients with moderate to severe knee OA eligible for TKR delayed surgery for at least 2 years following non-surgical treatment.

The aim of this study was to investigate the effects of individual physical characteristics on preweaning survival and growth of piglets born in a noncrate system. Data were collected from 3,402 ...neonatal piglets from 203 Landrace × Yorkshire sows housed in noncrate pens in a commercial Danish sow herd. Piglets were categorized into groups according to their survivability: surviving to weaning (SURV), stillborn (STILL), or dead between birth and weaning (DBW), which was subdivided into dead d 0 to 1 after farrowing (DEAD1) or dead d 2 to 26 after farrowing (DEAD26). Linear models were used to determine which physical characteristics affected survivability and growth of piglets. Results showed that characteristics related to the individual piglets had a greater degree of explanatory power in relation to survival than variables related to the sow. Survival of piglets increased if piglets were females (P < 0.001), had a greater body mass index (P < 0.001), and were born to sows of parity 3 or more (P = 0.017). Piglets with a greater birth weight were more likely to survive (P < 0.001), but birth weight was inferior to body mass index in explaining differences between SURV and DBW. Piglets that died 2 to 26 d after birth had a lower birth weight (P < 0.001), were born to sows of parity 1 or 2 (P = 0.014), and were born after a shorter gestation (P = 0.011) compared with SURV. Piglets that died on d 0 to 1 after birth had a lower body mass index (P < 0.001), displayed a greater degree of growth restriction (P = 0.004), and were born in large litters (P = 0.005). The gender of the piglets affected survivability at both d 0 to 1 (P < 0.001) and d 2 to 26 (P < 0.001). Piglets in DEAD1 differed from STILL by having a shorter crown to rump length (P < 0.001), a birth weight that deviated more from the mean weight of the litter (P = 0.001), and being more likely to be born before d 116 of gestation (P = 0.008). The only physical characteristic that was important for growth performance in the suckling period was birth weight (P < 0.001), yet using only birth weight as an indicator for survivability was too simplistic. The results of this study emphasize that individual characteristics of neonatal piglets could serve as indicators of survivability of piglets born in noncrate systems; however, the results suggest that the importance of characteristics differed in different periods of the preweaning period.
Background
Around 20% of patients with osteoarthritis (OA) have chronic post‐operative pain after total knee arthroplasty (TKA) and often undergo revision surgery with unfavourable pain outcome. This ...study compared sensitization in pain patients with knee OA and after revision TKA (re‐TKA).
Methods
Median pressure pain thresholds (PPTs) assessed from the most affected knee (localized sensitization) were used to subgroup 53 patients with OA pain and 20 patients with pain after re‐TKA: group 1: OA and high‐knee PPT; group 2: OA and low‐knee PPT; group 3: re‐TKA and high‐knee PPT; group 4: re‐TKA and low‐knee PPT. Clinical pain intensity was assessed using a visual analogue scale (VAS). Bilateral PPTs were measured from the lower leg and forearm (spreading sensitization). Furthermore, the pain intensities evoked by 10 repeated pressure pain stimuli (temporal summation) at the knee and lower leg were assessed on an electronic VAS.
Results
The mean clinical pain intensity was not significantly different between groups. The PPTs from both lower leg and forearm were significantly lower in group 4 compared to groups 1, 2, and 3 and in groups 2 and 3 compared to group 1 (p < 0.05). Temporal summations from the knee and lower leg were significantly facilitated in groups 3 and 4 compared to groups 1 and 2 (p < 0.05).
Conclusions
Despite similar pain intensities, facilitated temporal summation is worse in re‐TKA than in OA and patients with high local knee hyperalgesia show more prominent spreading sensitization. The study suggests that sensitization should be considered in knee OA especially before re‐TKA.
The cortical underconnectivity theory asserts that reduced long-range functional connectivity might contribute to a neural mechanism for autism. We examined resting-state blood oxygen level-dependent ...interhemispheric correlation in 53 males with high-functioning autism and 39 typically developing males from late childhood through early adulthood. By constructing spatial maps of correlation between homologous voxels in each hemisphere, we found significantly reduced interhemispheric correlation specific to regions with functional relevance to autism: sensorimotor cortex, anterior insula, fusiform gyrus, superior temporal gyrus, and superior parietal lobule. Observed interhemispheric connectivity differences were better explained by diagnosis of autism than by potentially confounding neuropsychological metrics of language, IQ, or handedness. Although both corpus callosal volume and gray matter interhemispheric connectivity were significantly reduced in autism, no direct relationship was observed between them, suggesting that structural and functional metrics measure different aspects of interhemispheric connectivity. In the control but not the autism sample, there was decreasing interhemispheric correlation with subject age. Greater differences in interhemispheric correlation were seen for more lateral regions in the brain. These findings suggest that long-range connectivity abnormalities in autism are spatially heterogeneous and that transcallosal connectivity is decreased most in regions with functions associated with behavioral abnormalities in autism. Autism subjects continue to show developmental differences in interhemispheric connectivity into early adulthood.
The objective of this study was to investigate piglet mortality in a commercial setting where sows were accommodated in a loose-housed system with an option to confine the sow for a few days around ...farrowing and during early lactation. The study was conducted in a Danish piggery where records were obtained from 2,139 farrowings. Sows were randomly allocated to 1 of 3 treatments: loose-loose (LL), loose-confined (LC), and confined-confined (CC). In LL, sows were loose housed from the time they entered the farrowing pens to weaning. In LC, sows were loose housed until farrowing was finished and then confined to d 4 after farrowing. In CC, sows were confined at d 114 of gestation to d 4 after farrowing. All sows were loose housed from d 5 to weaning. Total piglet mortality was analyzed at batch level to include piglets fostered by nurse sows and at sow level to analyze the effects of confinement during different time periods. Total piglet mortality was greater in LL (26.0%) and LC (25.4%) compared with CC (22.1%; < 0.001). The proportion of stillborn piglets was not different between treatments ( = 0.21) but a larger proportion was crushed in LL (10.7%) compared with LC (9.7%; = 0.03), which again was greater than CC (7.8%; < 0.001). Piglet mortality before equalization was lower in CC (3.7%) than in LL (7.5%) and LC (7.0%; < 0.001). Confinement reduced mortality from litter equalization to d 4 (7.6% for LL vs. 6.7% for LC; = 0.01) but more so in CC (5.6%) than in LC ( < 0.001). From d 4 to weaning, LL had lower mortality (5.6%) than LC (6.9%) and CC (6.6%; = 0.01). A larger proportion of sows in CC were classified as "low mortality" compared with LL and LC both before ( < 0.001) and after ( = 0.002) litter equalization. The results in this study emphasize that the period of time from the birth of the first piglet to litter equalization is important in relation to piglet mortality. The results also suggest that confinement for 4 d after farrowing can reduce mortality in this specific period, but only confinement from d 114 of gestation to d 4 after farrowing reduced total piglet mortality.
Abstract Aquaculture is a rapidly growing food production technology, but there are significant concerns related to its environmental impact and adverse social effects. We examine aquaculture ...outcomes in a three pillars of sustainability framework by analyzing data collected using the Aquaculture Performance Indicators. Using this approach, comparable data has been collected for 57 aquaculture systems worldwide on 88 metrics that measure social, economic, or environmental outcomes. We first examine the relationships among the three pillars of sustainability and then analyze performance in the three pillars by technology and species. The results show that economic, social, and environmental outcomes are, on average, mutually reinforced in global aquaculture systems. However, the analysis also shows significant variation in the degree of sustainability in different aquaculture systems, and weak performance of some production systems in some dimensions provides opportunity for innovative policy measures and investment to further align sustainability objectives.
