Abstract Background and purpose For the first time, delivered dose to the rectum has been calculated and accumulated throughout the course of prostate radiotherapy using megavoltage computed ...tomography (MVCT) image guidance scans. Dosimetric parameters were linked with toxicity to test the hypothesis that delivered dose is a stronger predictor of toxicity than planned dose. Material and methods Dose–surface maps (DSMs) of the rectal wall were automatically generated from daily MVCT scans for 109 patients within the VoxTox research programme. Accumulated-DSMs, representing total delivered dose, and planned-DSMs, from planning CT data, were parametrised using Equivalent Uniform Dose (EUD) and ‘DSM dose-width’, the lateral dimension of an ellipse fitted to a discrete isodose cluster. Associations with 6 toxicity endpoints were assessed using receiver operator characteristic curve analysis. Results For rectal bleeding, the area under the curve (AUC) was greater for accumulated dose than planned dose for DSM dose-widths up to 70 Gy. Accumulated 65 Gy DSM dose-width produced the strongest spatial correlation (AUC 0.664), while accumulated EUD generated the largest AUC overall (0.682). For proctitis, accumulated EUD was the only reportable predictor (AUC 0.673). Accumulated EUD was systematically lower than planned EUD. Conclusions Dosimetric parameters extracted from accumulated DSMs have demonstrated stronger correlations with rectal bleeding and proctitis, than planned DSMs.

The Ribosomal Database Project (RDP-II), previously described by Maidak et al. Nucleic Acids Res. (2000), 28, 173-174, continued during the past year to add new rRNA sequences to the aligned data and ...to improve the analysis commands. Release 8.0 (June 1, 2000) consisted of 16 277 aligned prokaryotic small subunit (SSU) rRNA sequences while the number of eukaryotic and mitochondrial SSU rRNA sequences in aligned form remained at 2055 and 1503, respectively. The number of prokaryotic SSU rRNA sequences more than doubled from the previous release 14 months earlier, and approximately 75% are longer than 899 bp. An RDP-II mirror site in Japan is now available (http://wdcm.nig.ac.jp/RDP/html/index.h tml). RDP-II provides aligned and annotated rRNA sequences, derived phylogenetic trees and taxonomic hierarchies, and analysis services through its WWW server (http://rdp.cme.msu.edu/). Analysis services include rRNA probe checking, approximate phylogenetic placement of user sequences, screening user sequences for possible chimeric rRNA sequences, automated alignment, production of similarity matrices and services to plan and analyze terminal restriction fragment polymorphism experiments. No adeno-associated virus (AAV) capsid has been described in the literature to exhibit a primary oligodendrocyte tropism when a constitutive promoter drives gene expression, which is a significant ...barrier for efficient in vivo oligodendrocyte gene transfer. The vast majority of AAV vectors, such as AAV1, 2, 5, 6, 8 or 9, exhibit a dominant neuronal tropism in the central nervous system. However, a novel AAV capsid (Olig001) generated using capsid shuffling and directed evolution was recovered after rat intravenous delivery and subsequent capsid clone rescue, which exhibited a >95% tropism for striatal oligodendrocytes after rat intracranial infusion where a constitutive promoter drove gene expression. Olig001 contains a chimeric mixture of AAV1, 2, 6, 8 and 9, but unlike these parental serotypes after intravenous administration Olig001 has very low affinity for peripheral organs, especially the liver. Furthermore, in mixed glial cell cultures, Olig001 exhibits a 9-fold greater binding when compared with AAV8. This novel oligodendrocyte-preferring AAV vector exhibits characteristics that are a marked departure from previously described AAV serotypes.
ABSTRACT
Infrared dark clouds (IRDCs) are potential hosts of the elusive early phases of high mass star formation (HMSF). Here, we conduct an in-depth analysis of the fragmentation properties of a ...sample of 10 IRDCs, which have been highlighted as some of the best candidates to study HMSF within the Milky Way. To do so, we have obtained a set of large mosaics covering these IRDCs with Atacama Large Millimeter/submillimeter Array (ALMA) at Band 3 (or 3 mm). These observations have a high angular resolution (∼3 arcsec; ∼0.05 pc), and high continuum and spectral line sensitivity (∼0.15 mJy beam−1 and ∼0.2 K per 0.1 km s−1 channel at the N2H+ (1 − 0) transition). From the dust continuum emission, we identify 96 cores ranging from low to high mass (M = 3.4−50.9 M⊙) that are gravitationally bound (αvir = 0.3−1.3) and which would require magnetic field strengths of B = 0.3−1.0 mG to be in virial equilibrium. We combine these results with a homogenized catalogue of literature cores to recover the hierarchical structure within these clouds over four orders of magnitude in spatial scale (0.01–10 pc). Using supplementary observations at an even higher angular resolution, we find that the smallest fragments (<0.02 pc) within this hierarchy do not currently have the mass and/or the density required to form high-mass stars. None the less, the new ALMA observations presented in this paper have facilitated the identification of 19 (6 quiescent and 13 star-forming) cores that retain >16 M⊙ without further fragmentation. These high-mass cores contain trans-sonic non-thermal motions, are kinematically sub-virial, and require moderate magnetic field strengths for support against collapse. The identification of these potential sites of HMSF represents a key step in allowing us to test the predictions from high-mass star and cluster formation theories.
Summary
Background
Periostin is highly expressed in eosinophilic oesophagitis (EoE), but has not been extensively studied as a non‐invasive biomarker.
Aim
To assess whether serum periostin ...distinguished EoE from controls at baseline, had utility for monitoring treatment response, or was associated with IL‐13 levels.
Methods
This was a sub‐analysis of a prospective cohort study of adults undergoing out‐patient upper endoscopy. Incident cases of EoE were diagnosed per consensus guidelines. Controls were subjects with either GERD or dysphagia without EoE. EoE patients were treated with swallowed/topical steroids and had repeat endoscopy/biopsy. Serum periostin levels for cases and controls were compared at baseline, and pre/post‐treatment levels were compared for cases. Serum IL‐13 and tissue expression of periostin were also assessed.
Results
A total of 61 incident EoE cases and 87 controls were analysed. Despite a marked increase in tissue periostin expression in cases, the median baseline serum periostin level was only slightly higher in cases than controls (22.1 ng/mL vs. 20.7; P = 0.04); there was no change in post‐treatment levels. There was also no difference in serum periostin for cases by histologic response or atopic status. There was a strong trend towards higher serum IL‐13 levels in cases in the highest periostin quartile (57.1 pg/mL vs. 2.6; P = 0.07).
Conclusions
Serum periostin levels were similar in cases and controls, and there were no changes post‐treatment. Given elevated IL‐13 levels in the EoE patients with the highest periostin levels, future studies could explore periostin as a biomarker in EoE, perhaps in the setting of anti‐IL‐13 therapy.
We describe observations from the Fennec supersite at Bordj Badji Mokhtar (BBM) made during the June 2011 Fennec Intensive Observation Period. These are the first detailed in situ observations of ...meteorology and dust from the central Sahara, close to the center of the Saharan heat low and the summertime dust maximum. Historically, a shortage of such Saharan observations has created problems for evaluating processes, models, and remote sensing. There was a monsoon influence at BBM before 8 June and after 12 June, with dry Harmattan winds in between. A split boundary layer, generated by ventilation from the Atlantic, persisted during the drier phase. Extensive cold pools (haboobs) and microburst‐type events were regularly observed. Moisture reached BBM at night from the monsoon and the embedded haboobs. As well as the regularly occurring nocturnal low‐level jet (LLJ), a Saharan upper boundary layer (650 hPa) jet was observed, where winds feel drag from dry convection in the afternoon. This jet is linked to the diurnal cycles of moisture and cloud. Most dust was observed in the cloudier monsoon‐affected periods, and covarying dust and cloud amounts explain most of the variations in shortwave radiation that control the surface sensible flux. Dustiness is related to a standard parameterization of uplift using 10 m winds (“uplift potential”), and this is used to estimate uplift. Around 50% of uplift is nocturnal. Around 30% is from the LLJ, and 50% is from haboobs, which are mainly nocturnal. This demonstrates, for the first time from observations, the key role of haboobs, which are problematic for models.
