The dye compounds appeared to degrade readily in the equilibrium and exposure periods, suggesting a limited persistence of the parent compounds in the environment under test conditions. Although azo dye degradation products could not be reliably quantified, one was detected in DY7 sediment samples that elicited toxic effects to
Hexagenia
and
Tubifex
, providing evidence that DY7 degrades.
Hexagenia
survival and growth endpoints responded with similar sensitivity to the dyes, but DY7 was the most toxic, with a 21-day IC25 (concentration associated with 25% inhibition) for growth of 9.6 μg/g. Comparatively,
Tubifex
reproduction was the most sensitive endpoint for all dyes with 28-day IC25s for young production ranging from 1.3 to 11.8 μg/g. At sublethal concentrations, toxic effects to
Tubifex
differed between dyes: the solvent dye exerted an effect primarily on gametogenesis (cocoon production), while disperse dyes, most notably DY7, caused effects on embryogenesis (development of worm inside the cocoon). This study indicates that there could be potential hazard to oligochaetes based on the observed effect concentrations, but given the lack of environmental measurements, the risk of these compounds is unknown. Further research is required to determine if degradation products were formed in all dye samples and whether toxicity was caused by the parent molecules, which have limited persistence under test conditions, or by their degradation products. To avoid underestimating toxicity, this study stresses the need to use an infaunal deposit feeder such as the oligochaete
Tubifex
in sediment toxicity assessments where highly hydrophobic compounds are present.
Metformin is a glucose-lowering drug taken for diabetes. It is excreted by humans in urine and detected in municipal wastewater effluents and rivers. Fathead minnows ( Pimephales promelas) were ...exposed over a life cycle to measured concentrations of metformin: 3.0, 31, and 322 μg/L. No significant changes were observed in survival, maturation, growth, condition factor, or liver size. Relative ovary size of females exposed to 322 μg/L metformin was significantly larger than controls. There was no induction of vitellogenin in plasma of minnows, and gonad maturation was not statistically different from controls. The start of breeding was delayed by 9–10 d in the mid- and high metformin treatments (statistically significant only in the mid-concentration), but numbers and quality of eggs were not statistically different from controls. There were no effects of metformin on survival or growth of offspring. Exposure to metformin at environmentally relevant concentrations (i.e., 3.0 and 31 μg/L metformin) caused no adverse effects in fathead minnows exposed for a life cycle, with the exception of a delay in time to first breeding (that did not impact overall egg production). The results of the study are important to help understand whether metformin concentrations in rivers and lakes can harm fishes.
The objectives of this study were to evaluate the influence of natural oil sands on the early developmental stages of white sucker (Catostomus commersoni) and to determine whether biochemical ...responses in this species were similar to native fish caught in the Athabasca Oil Sands area. Early life stage (ELS) sediment toxicity tests were conducted using controls, reference sediments, natural oil sands, and industrially contaminated (wastewater pond) sediments collected from sites along the Athabasca River, Alberta (Canada). Eggs and larvae were observed for mortality, hatching, deformities, growth, and cytochrome P-4501A (CYP1A) activity using immunohistochemistry. E-Nat-, S-Nat-, and wastewater pond sediment-exposed groups showed significant premature hatching, reduced growth, and exposure-dependent increases in ELS mortality and larval malformations relative to controls. The most common larval deformities included edemas (pericardial, yolk sac, and subepidermal), hemorrhages, and spinal defects. Juveniles exposed to oil sands and wastewater pond sediments (96 h) demonstrated significantly increased 7-ethoxyresorufin-O-deethylase (EROD) activity (30- to 50-fold) as compared to controls. Reference sediment-exposed groups and water controls demonstrated reliable embryo and larval survival, minimal malformations, and negligible CYP1A staining. These observed signs of blue sac disease (ELS mortality, malformations, growth reductions, CYP1A activity induction) may produce deleterious reproductive effects in natural fish populations exposed to oil sands mixtures.

The invasive round goby has a recognised role in transferring contaminants through foodwebs, but little work has been done on contaminant impacts on round gobies themselves. Here we present the first ...case study of contaminant biomarkers and subpopulation structure variation in round gobies, in relation to habitat contamination, within a Canadian Area of Concern, Hamilton Harbour. Copper and cadmium were elevated in livers of fish from contaminated habitats. Although catch abundances were similar across sites, fish were smaller, a greater proportion of fish were female and more males were in reproductive condition in contaminated sites. Fish from contaminated areas showed more fin loss. Males from contaminated sites showed intersex gonads and genitalia. Ethoxyresorufin-
o-deethylase (EROD) activity was higher in fish collected near polycyclic aromatic hydrocarbon (PAH)-rich sediments. The results indicate that contaminants impact the characteristics of round goby populations, which could affect ecosystems beyond toxicant biomagnification. This study also confirms that round gobies can be abundant in polluted habitats, which may draw predators—facilitating mobilisation of contaminants in foodwebs.
Fathead minnow embryos and larvae are frequently used in toxicology, including short-term embryo-only tests which often use small volumes of test solution. The effect that such conditions may have on ...fathead minnow development has yet to be explicitly described. Here we compared rates of embryonic development in fathead minnow embryos reared under standard light and temperature conditions with a range of possible methods. All methods yielded excellent control survival. We demonstrated that fathead minnow embryos incubated in a range of small volumes in multi-well plates (500μL to 2mL per embryo) did not substantially vary in developmental rate, but flexed less frequently as embryos, hatched smaller, later and with larger yolk-sacs, and initiated feeding later than embryos reared in an excess of solution (20mL per embryo) with or without supplemental aeration. Faster hatch and growth were promoted with an orbital shaker, but growth benefits were not sustained into the larval stage. Developmental differences persisted in larvae reared to 20days post-fertilization when monitoring ceased, but growth differences did not magnify and in some measurements partially resolved. To our knowledge we are the first to report effects of incubation in multi-well plates in any fish taxa. As our data revealed that the eleutheroembryonic stage for fathead minnow may be prolonged in multi-well plates, this may allow the use of longer toxicity tests using fathead minnow embryos without conflicting with existing animal welfare legislation in many countries.
This article is a review of research efforts over the last decade on the sources and characteristics of substances in Canadian pulp mill effluents associated with two responses in fish: (1) induction ...of detoxification enzymes and (2) reproductive effects. The initial uncertainty regarding the role of chlorine bleaching and dioxins in these responses was resolved by the mid 1990s, when it was determined that effects were not correlated with effluent adsorbable organic halogen (AOX) levels and that releases of dioxins had decreased substantially. In the mid 1990s researchers were able to partially attribute enzyme activity induction in fish to wood components, while other studies showed individual wood extractives had the potential to affect fish reproduction. A lack of correlation between threshold reproductive responses and effluent concentrations indicated additional unidentified compounds and mechanisms were involved. In the late 1990s, source identification approaches in concert with the development of mechanistically linked in vitro and in vivo bioassays showed multiple compounds are affecting production and signaling of sex steroids in fish. These substances are bioavailable and accumulated rapidly, consistent with the body of evidence that has shown a sustained exposure is required to produce both elevated enzyme activity and depressions in sex steroid levels. The patterns of these substances in effluents and fish tissues are not correlated with production type or effluent treatment. Collectively, these findings show that bioactive substances originate from wood and are derived from lignin and/or terpenoids, they are liberated during pulp digestion, and in kraft mills they are present in black liquor and chemical recovery condensates. Additional bioactive substances are also present in bleachery effluents containing residual lignin. The lack of a definitive identification of the responsible compounds has prevented an evaluation of the effectiveness of industry-wide process changes. Continued research into the identities, origins, and environmental fate of these substances and the efficacy of effluent treatment is required to determine their significance and relationship to the existing impacts of effluents from pulp and paper mills in Canadian aquatic ecosystems.
