Curcumin is the primary polyphenol in turmeric's curcuminoid class. It has a wide range of therapeutic applications, such as anti-inflammatory, antioxidant, antidiabetic, hepatoprotective, ...antibacterial, and anticancer effects against various cancers, but has poor solubility and low bioavailability. Objective: To improve curcumin's bioavailability, plasma concentration, and cellular permeability processes. The nanocurcumin approach over curcumin has been proven appropriate for encapsulating or loading curcumin (nanocurcumin) to increase its therapeutic potential. Conclusion: Though incorporating curcumin into nanocurcumin form may be a viable method for overcoming its intrinsic limitations, and there are reasonable concerns regarding its toxicological safety once it enters biological pathways. This review article mainly highlights the therapeutic benefits of nanocurcumin over curcumin.
Alzheimer's disease (AD) is a chronic, age-related, and irreversible brain disorder that typically develops slowly and gets worse over time. The potent auspicious drug candidate for the treatment of ...AD is supposed to perform the simultaneous modulation of several targets linked to AD. The new therapeutic approach involves drug candidates that are designed to act on multiple targets and have various pharmacological properties. This trend has triggered the development of various multimodal drugs including TV-3326 (i.e. ladostigil) and M-30 (i.e. a new multitarget iron chelator). TV-3326 combines the neurorestorative/neuroprotective effects of the cholinesterase (ChE) inhibitory activity of rivastigmine with rasagiline (a selective monoamine oxidase-B inhibitor and novel antiparkinsonian agent) in a single molecule. M-30, the second derivative of rasagiline, was developed by combining the propargyl moiety of rasagiline into the skeleton of VK-28 (i.e. a novel brain permeable neuroprotective iron chelator). It has been revealed that both the compounds possess anti-AD effects and therefore, the clinical development is directed to the treatment of this type of neurodegenerative diseases (NDs). In this article, we have reviewed the neuroprotective molecular mechanisms and multimodal effects of TV-3326 and M-30.
Glial cells are the most abundant cells of the brain, outnumbering neurons. These multifunctional cells are crucial for maintaining brain homeostasis by providing trophic and nutritional support to ...neurons, sculpting synapses, and providing an immune defense. Glia are highly plastic and undergo both structural and functional alterations in response to changes in the brain microenvironment. Glial phenotypes are intimately regulated by underlying metabolic machinery, which dictates the effector functions of these cells. Altered brain energy metabolism and chronic neuroinflammation are common features of several neurodegenerative diseases. Microglia and astrocytes are the major glial cells fueling the ongoing neuroinflammatory process, exacerbating neurodegeneration. Distinct metabolic perturbations in microglia and astrocytes, including altered carbohydrate, lipid, and amino acid metabolism have been documented in neurodegenerative diseases. These disturbances aggravate the neurodegenerative process by potentiating the inflammatory activation of glial cells. This review covers the recent advances in the molecular aspects of glial metabolic changes in the pathophysiology of neurodegenerative diseases. Finally, we discuss studies exploiting glial metabolism as a potential therapeutic avenue in neurodegenerative diseases.
Abstract
The development of the advanced metering infrastructure (AMI) and the application of artificial intelligence (AI) enable electrical systems to actively engage in smart grid systems. Smart ...homes with energy storage systems (ESS) and renewable energy sources (RES)-known as home microgrids-have become a critical enabling technology for the smart grid. This article proposes a new model for the energy management system of a home microgrid integrated with a battery ESS (BESS). The proposed dynamic model integrates a deep learning (DL)-based predictive model, bidirectional long short-term memory (Bi-LSTM), with an optimization algorithm for optimal energy distribution and scheduling of a BESS-by determining the characteristics of distributed resources, BESS properties, and the user’s lifestyle. The aim is to minimize the per-day electricity cost charged by time-of-use (TOU) pricing while considering the day-basis peak demand penalty. The proposed system also considers the operational constraints of renewable resources, the BESS, and electrical appliances. The simulation results from realistic case studies demonstrate the validation and responsibility of the proposed system in reducing a household’s daily electricity cost.
Rapid industrialization, urbanization, global warming, and climate change are compromising surface water quality across the globe. Consequently, water conservation is essential for both environmental ...sustainability and human survival. This study assesses the water quality of the Jamuna River in Bangladesh at five distinct sites during wet and dry seasons. It employs six global water quality indices (WQIs) and contrasts the results with Bangladesh's Environmental Quality Standard (EQS) and the Department of Environment (DoE) criteria. The WQI models used are the Weighted Arithmetic WQI (WAWQI), British Columbia WQI (BCWQI), Canadian Council of Ministers of the Environment WQI (CWQI), Assigned WQI (AWQI), Malaysian WQI (MWQI), and Oregon WQI (OWQI). Fifteen physicochemical parameters were analyzed according to each WQI model's guidelines. The findings reveal that most parameters surpass the standard permissible values. The WQI model results indicate that the average water quality across the five sites falls into the lowest category. A comparison of the WQI models suggests potential correlations between WAWQI and AWQI, as well as between MWQI and OWQI. The straightforward presentation of the WQI models indicates that while the river water requires treatment for household and drinking use, it remains suitable for irrigation. The decline in water quality is likely attributable to human activities, urbanization, municipal waste disposal, and industrial effluents. Authorities must prioritize regular monitoring and assessment of water quality to address the identified challenges. Restoring the water to an acceptable standard will become increasingly difficult without proactive measures.
Cancer is a major burden of disease globally. Each year, tens of millions of people are diagnosed with cancer worldwide, and more than half of the patients eventually die from it. Significant ...advances have been noticed in cancer treatment, but the mortality and incidence rates of cancers are still high. Thus, there is a growing research interest in developing more effective and less toxic cancer treatment approaches. Curcumin (CUR), the major active component of turmeric (
L.), has gained great research interest as an antioxidant, anticancer, and anti-inflammatory agent. This natural compound shows its anticancer effect through several pathways including interfering with multiple cellular mechanisms and inhibiting/inducing the generation of multiple cytokines, enzymes, or growth factors including IκB kinase β (IκKβ), tumor necrosis factor-alpha (TNF-α), signal transducer, and activator of transcription 3 (STAT3), cyclooxygenase II (COX-2), protein kinase D1 (PKD1), nuclear factor-kappa B (NF-κB), epidermal growth factor, and mitogen-activated protein kinase (MAPK). Interestingly, the anticancer activity of CUR has been limited primarily due to its poor water solubility, which can lead to low chemical stability, low oral bioavailability, and low cellular uptake. Delivering drugs at a controlled rate, slow delivery, and targeted delivery are other very attractive methods and have been pursued vigorously. Multiple CUR nanoformulations have also been developed so far to ameliorate solubility and bioavailability of CUR and to provide protection to CUR against hydrolysis inactivation. In this review, we have summarized the anticancer activity of CUR against several cancers, for example, gastrointestinal, head and neck, brain, pancreatic, colorectal, breast, and prostate cancers. In addition, we have also focused on the findings obtained from multiple experimental and clinical studies regarding the anticancer effect of CUR in animal models, human subjects, and cancer cell lines.
Breast cancer is one of the most common types of cancer among women globally. It is caused by mutations in the estrogen/progesterone receptors and conventional treatment methods are commonly ...utilized. About 70-80 percent of individuals with the early-stage non-metastatic disease may be cured. Conventional treatment is far less than the optimal ratio, as demonstrated through the high mortality rate of women with this cancer. However, conventional treatment methods like surgery, radiotherapy, and chemotherapy are not as effective as expected and lead to concerns about low bioavailability, low cellular uptake, emerging resistance, and adverse toxicities. A nanomedicine-based approach is a promising alternative for breast cancer treatment. The present era is witnessing rapid advancements in nanomedicine as a platform for investigating novel therapeutic applications and modern intelligent healthcare management strategies. This paper focuses on nanomedicine-based therapeutic interventions that are becoming more widely accepted for improving treatment effectiveness and reducing undesired side effects in breast cancer patients. By evaluating the state-of-the-art tools and taking the challenges involved into consideration, various aspects of the proposed nano-enabled therapeutic approaches have been discussed in this review.

This paper presents a numerical investigation of the flexural behavior of timber beams externally strengthened with carbon-fiber-reinforced polymer (CFRP) sheets. At first, the accuracy of linear ...elastic and elastic-plastic models in predicting the behavior of bare timber beams was compared. Then, two modeling approaches (i.e., the perfect bond method and progressive damage technique using the cohesive zone model (CZM)) were considered to simulate the interfacial behavior between FRP and timber. The models were validated against published experimental data, and the most accurate numerical procedure was identified and subsequently used for a parametric study. The length of FRP sheets varied from 50% to 100% of the total length of the beam, while different FRP layers were considered. Moreover, the effects of two strengthening configurations (i.e., FRP attached in the tensile zone only and in both the tensile and compressive zones) on load-deflection response, flexural strength, and flexural rigidity were considered. The results showed that elastic-plastic models are more accurate than linear elastic models in predicting the flexural strength and failure patterns of bare timber beams. In addition, with increasing FRP length, the increase in flexural strength ranged from 10.3% to 52.9%, while no further increase in flexural strength could be achieved beyond an effective length of 80% of the total length of the beam. Attaching the FRP to both the tensile and compressive zone was more effective in enhancing the flexural properties of the timber beam than attaching the FRP to the tensile zone only.
