As part of an effort to develop aqueous isotope harvesting techniques at radioactive beam facilities, 48V and a cocktail of primary- and secondary-beam ions created by the fragmentation reaction of a ...160 MeV/nucleon 58Ni beam were stopped in an aqueous target cell. After collection, 48V was separated from the mixture of beam ions using cation-exchange chromatography. The extraction efficiency from the aqueous solution was (47.0 ± 2.5)%, and the isolated 48V had a radiochemical purity of 95.8%. This proof-of-concept work shows that aqueous isotope harvesting could provide significant quantities of rare isotopes which are currently unavailable at conventional facilities.
•A selective method was developed to separate the multivalent element vanadium from a complex mixture of elements (Z ~ 1–28).•Vanadium-48 was produced via heavy-ion fragmentation of a 58Ni beam at the National Superconducting Cyclotron Laboratory.•The primary- and secondary-beam ions, including 48V, were stopped in an aqueous target cell and characterized.•By way of cation-exchange chromatography, the 48V recovery was 47.0 ± 2.5%.•The radionuclidic purity of the recovered 48V was 95.8 ± 2.5%.
An automated approach to the collection of 1H NMR (nuclear magnetic resonance) spectra using a benchtop NMR spectrometer and the subsequent analysis, processing, and elucidation of components present ...in seized drug samples are reported. An algorithm is developed to compare spectral data to a reference library of over 300 1H NMR spectra, ranking matches by a correlation-based score. A threshold for identification was set at 0.838, below which identification of the component present was deemed unreliable. Using this system, 432 samples were surveyed and validated against contemporaneously acquired GC–MS (gas chromatography–mass spectrometry) data. Following removal of samples which possessed no peaks in the GC–MS trace or in both the 1H NMR spectrum and GC–MS trace, the remaining 416 samples matched in 93% of cases. Thirteen of these samples were binary mixtures. A partial match (one component not identified) was obtained for 6% of samples surveyed whilst only 1% of samples did not match at all.
CD1c presents lipid-based antigens to CD1c-restricted T cells, which are thought to be a major component of the human T cell pool. However, the study of CD1c-restricted T cells is hampered by the ...presence of an abundantly expressed, non-T cell receptor (TCR) ligand for CD1c on blood cells, confounding analysis of TCR-mediated CD1c tetramer staining. Here, we identified the CD36 family (CD36, SR-B1, and LIMP-2) as ligands for CD1c, CD1b, and CD1d proteins and showed that CD36 is the receptor responsible for non-TCR-mediated CD1c tetramer staining of blood cells. Moreover, CD36 blockade clarified tetramer-based identification of CD1c-restricted T cells and improved identification of CD1b- and CD1d-restricted T cells. We used this technique to characterize CD1c-restricted T cells ex vivo and showed diverse phenotypic features, TCR repertoire, and antigen-specific subsets. Accordingly, this work will enable further studies into the biology of CD1 and human CD1-restricted T cells.
The purpose of this research was to determine the expected progression of adolescent female swimming performances using a longitudinal approach. The performances of 514 female swimmers (12⁻19 year ...olds) who participated in one or more FINA-regulated annual international schools' swimming championships over an eight-year period were analysed. Quadratic functions for each of the seven individual events (50, 100, 200 m freestyle, 100 m backstroke, breaststroke, butterfly, 200 m individual medley) were determined using mixed linear models. The predicted threshold of peak performance ranged from 16.8 ± 0.2 (200 m individual medley) to 20.6 ± 0.1 (100 m butterfly) years of age, preceded by gradual rates of improvement (mean rate of 1.6% per year). However, following cross validation, only three events (100 m backstroke, 200 m individual medley and 200 m freestyle) produced reliable models. Identifying the factors that contribute to the progression of female performance in this transitory period of life remains challenging, not least since the onset of puberty is likely to have occurred prior to reaching 12 years of age, the minimum competition age for this championship.
The effect of working on-call from home on the sympatho-adrenal medullary system activity is currently unknown. This study had two aims, Aim 1: examine salivary alpha amylase awakening response (AAR) ...and diurnal salivary alpha amylase (sAA) profile in fire and emergency service workers who operate on-call from home following a night on-call with a call (NIGHT-CALL), a night on-call without a call (NO-CALL) and an off-call night (OFF-CALL), and Aim 2: explore whether there was an anticipatory effect of working on-call from home (ON) compared to when there was an off-call (OFF) on the diurnal sAA profile. Participants wore activity monitors, completed sleep and work diaries and collected seven saliva samples a day for one week. AAR area under the curve with respect to ground (AUCG), AAR area under the curve with respect to increase (AUCI), AAR reactivity, diurnal sAA slope, diurnal sAA AUCG and mean 12-h sAA concentrations were calculated. Separate generalised estimating equation models were constructed for each variable of interest for each aim. For Aim 1, there were no differences between NIGHT-CALL or NO-CALL and OFF-CALL for any response variable. For Aim 2, there was no difference between any response variable of interest when ON the following night compared to when OFF the following night (n = 14). These findings suggest that there is no effect of working on-call from home on sAA, but should be interpreted with caution, as overnight data were not collected. Future research, using overnight heart rate monitoring, could help confirm these findings.
There is no consensus on the optimal timing and specific brain MRI sequences in the evaluation and management of severe pediatric traumatic brain injury (TBI), and information on current practices is ...lacking. The authors performed a survey of MRI practices among sites participating in a multicenter study of severe pediatric TBI to provide information for designing future clinical trials using MRI to assess brain injury after severe pediatric TBI.
Information on current imaging practices and resources was collected from 27 institutions participating in the Approaches and Decisions after Pediatric TBI Trial. Multiple-choice questions addressed the percentage of patients with TBI who have MRI studies, timing of MRI, MRI sequences used to investigate TBI, as well as the magnetic field strength of MR scanners used at the participating institutions and use of standardized MRI protocols for imaging after severe pediatric TBI.
Overall, the reported use of MRI in pediatric patients with severe TBI at participating sites was high, with 40% of sites indicating that they obtain MRI studies in > 95% of this patient population. Differences were observed in the frequency of MRI use between US and international sites, with the US sites obtaining MRI in a higher proportion of their pediatric patients with severe TBI (94% of US vs 44% of international sites reported MRI in at least 70% of patients with severe TBI). The reported timing and composition of MRI studies was highly variable across sites. Sixty percent of sites reported typically obtaining an MRI study within the first 7 days postinjury, with the remainder of responses distributed throughout the first 30-day postinjury period. Responses indicated that MRI sequences sensitive for diffuse axonal injury and ischemia are frequently obtained in patients with TBI, whereas perfusion imaging and spectroscopy techniques are less common.
Results from this survey suggest that despite the lack of consensus or guidelines, MRI is commonly obtained during the acute clinical setting after severe pediatric TBI. The variation in MRI practices highlights the need for additional studies to determine the utility, optimal timing, and composition of clinical MRI studies after TBI. The information in this survey describes current clinical MRI practices in children with severe TBI and identifies important challenges and objectives that should be considered when designing future studies.
Currently, there is a lack of appropriate skill assessments available for use in golf. The aim of this study was to examine the discriminative validity and the test-retest reliability of the newly ...developed "Nine-Ball Skills Test". Participants of two ability levels (elite, n = 14; high-level amateur, n = 16) each hit nine golf shots of differing combinations of trajectory (straight, fade, draw) and height (normal, high, low) at an individually determined target area. Each shot was scored on its percent error index from the target and whether it achieved the maximum height as required. Participants completed the test twice using a 5-iron club. The elite group scored significantly higher (P < 0.05) than the amateur group for both the first and second rounds of the test as well as the combined scores. The between-round test-retest reliability was deemed to be not acceptable, thus we propose that the test's protocol should include use of the two rounds as standard. Due to the importance of ball striking and flight control to performance in golf, the Nine-Ball Skills Test is appropriate for providing a measure of this skill component in elite and high-level amateur golfers.
