Monitoring wine-growing regions and maximising the value of production based on their region/local specificities requires accurate spatial and temporal monitoring. The increasing amount and ...variability of information from remote sensing data is a potential tool to assess this challenge for the grape and wine industry. This article provides a first insight into the capacity of a multiway analysis method applied to Sentinel-2 time series to assess the value of simultaneously considering spectral and temporal information to highlight site-specific canopy evolution in relation to environmental factors and management practices, which present a large diversity at this regional scale. Parallel Factor Analysis (PARAFAC) was used as an unsupervised technique to recover pure spectra and temporal signatures from multi-way spectral imagery of vineyards in the Languedoc-Roussillon region in the south of France. The model was developed using a time series of Sentinel-2 satellite imagery collected over 4978 vineyard blocks between May 2019 and August 2020. From the Sentinel-2 (spectral and temporal) signal, the PARAFAC analysis allowed the identification of spectral and temporal profiles in the form of pure components, which corresponded to vegetation and soil. The PARAFAC analysis also identified that two of the pure spectra were strongly related to characteristics and dynamics of vineyard cultivation at a regional scale. A conceptual framework was proposed in order to simultaneously consider both vegetation and soil profiles and to summarise the mass of data accordingly. This methodology allowed the computation of a concentration index that characterised how close a field was to a vegetation or a soil profile over the season. The concentration indices were validated for the vegetation and the soil over two growing seasons (2019 and 2020) with geostatistical analysis. A non-random distribution of the concentration index at the regional scale was assumed to highlight a strongly spatially organised phenomenon related to spatially organised environmental factors (soil, climate, training system, etc.). In a second step, spatial patterns of indices were subjected to the expertise of a panel of advisors of the wine industry in order to validate them in relation to vine-growing conditions. Results showed that the introduction of the PARAFAC method opened up the possibility to identify relevant spectro-temporal profiles for vine monitoring purposes.
Unexpected climatic conditions or extreme climatic events in vineyards are a worldwide problem that requires accurate spatial and temporal monitoring. Satellite-based remote sensing is an important ...source of data to assess this challenge in a climate-change context. This paper provides a first insight into the capacity of a multiway analysis method applied to Sentinel-2 time series to assess heatwave impacts in vineyards at a regional scale. Multi-way partial least squares (N-PLS) regression was used as a supervised technique to predict the intensity of damage caused to vineyards by the heatwave phenomenon that impacted the vineyards in the south of France in 2019. The model was developed based on available ground truth data of yield losses for 107 vineyard blocks in the Languedoc-Roussillon region and multispectral time-series predictor data for the period May to August 2019. The model showed a performance accuracy (R2) of 0.56 in the calibration set and of 0.66 in the validation set, with a standard error of cross-validation in the calibration set of 12.4% and a standard error of the prediction of yield losses in the validation set of 10.7. The model was applied at a regional scale on 4978 vineyard blocks to predict yield losses using spectral and temporal attributes. The prediction of the yield loss due to heat stress at a regional scale was related to the spatial pattern of maximum temperatures recorded during the extreme weather event. This relation was confirmed by a chi-square test (p < 5%). The introduction of N-PLS insights into the analysis enables the characterisation of heat stress responses in vineyards and the identification of spectro-temporal profiles relevant for understanding the effects of heatwaves on vine blocks at a regional scale.
A high-fidelity computational analysis carefully validated against concurrently obtained experimental results is employed to examine self-noise radiation of airfoils at transitional flow regimes, ...with a focus on elucidating the connection between the unsteady behavior of the laminar separation bubble (LSB) and the acoustic feedback-loop (AFL) resonant interactions observed in the airfoil boundary layers. The employed parametric study examines AFL sensitivity to the changes in the upstream flow conditions and the airfoil loading. Implicit Large-Eddy Simulations are performed for a NACA-0012 airfoil in selected transitional-flow regimes for which experimental measurements recorded characteristic multiple-tone acoustic spectra with a dual ladder-type frequency structure. The switch between the tone-producing and no-tone-producing regimes is traced to the LSB size and position as a function of the flow Reynolds number and the airfoil angle of attack, and further substantiated by the linear stability analysis. The results indicate a strong multi-tonal airfoil noise radiation associated with the AFL and attributed to the switch from the slowly-growing Tollmien–Schlichting to the fast-growing Kelvin–Helmholtz instabilities occurring in thin LSB regions when those are localized near the trailing-edge (TE) on either side of the airfoil. Such a process eventually results in the nonlinearly saturated flapping vortical modes (“rollers”) that scatter into acoustic waves at the TE.
Migrants represent an increasing proportion of people living with HIV in many developed countries. We aimed to describe the HIV care cascade and baseline genotypic resistance for newly diagnosed ...asylum seekers referred to the McGill University Health Centre (MUHC) in Montreal, Quebec, Canada.
We conducted a retrospective cohort study of patients linked to the MUHC from June 1, 2017 to October 31, 2018. We calculated the median time (days; interquartile range (IQR)) from: 1) entry into Canada to immigration medical examination (IME) (i.e. HIV screening); 2) IME to patient notification of diagnosis; 3) notification to linkage to HIV care (defined as a CD4 or viral load (VL) measure); 4) linkage to HIV care to combination antiretroviral therapy (cART) prescription; and 5) cART prescription to viral suppression (defined as a VL < 20 copies/mL). We reviewed baseline genotypes and interpreted mutations using the Stanford University HIV Drug Resistance Database. We calculated the proportion with full resistance to > 1 antiretroviral.
Overall, 43% (60/139) of asylum seekers were newly diagnosed in Canada. Among these, 62% were late presenters (CD4 < 350 cells/μl), 22% presented with advanced HIV (CD4 < 200 cells/μl), and 25% with high-level viremia (VL > 100,000 copies/ml). Median time from entry to IME: 27 days IQR:13;55; IME to notification: 28 days IQR:21;49; notification to linkage: 6 days IQR:2;19; linkage to cART prescription: 11 days IQR:6;17; and cART to viral suppression: 42 days IQR:31;88; 45% were linked to HIV care within 30 days. One-fifth (21%) had baseline resistance to at least one antiretroviral agent; the K103 N/S mutation was the most common mutation.
While the majority of newly diagnosed asylum seekers were late presenters, only 45% were linked to care within 30 days. Once linked, care and viral suppression were rapid. Delays in screening and linkage to care present increased risk for onward transmission, and in the context of 21% baseline resistance, consideration of point-of-care testing and immediate referral at IME screening should be made.

In this work, Raman hyperspectral images and multivariate curve resolution-alternating least squares (MCR-ALS) are used to study the distribution of actives and excipients within a pharmaceutical drug product. This article is mainly focused on the distribution of a low dose constituent. Different approaches are compared, using initially filtered or non-filtered data, or using a column-wise augmented dataset before starting the MCR-ALS iterative process including appended information on the low dose component. In the studied formulation, magnesium stearate is used as a lubricant to improve powder flowability. With a theoretical concentration of 0.5% (w/w) in the drug product, the spectral variance contained in the data is weak. By using a principal component analysis (PCA) filtered dataset as a first step of the MCR-ALS approach, the lubricant information is lost in the non-explained variance and its associated distribution in the tablet cannot be highlighted. A sufficient number of components to generate the PCA noise-filtered matrix has to be used in order to keep the lubricant variability within the data set analyzed or, otherwise, work with the raw non-filtered data. Different models are built using an increasing number of components to perform the PCA reduction. It is shown that the magnesium stearate information can be extracted from a PCA model using a minimum of 20 components. In the last part, a column-wise augmented matrix, including a reference spectrum of the lubricant, is used before starting MCR-ALS process. PCA reduction is performed on the augmented matrix, so the magnesium stearate contribution is included within the MCR-ALS calculations. By using an appropriate PCA reduction, with a sufficient number of components, or by using an augmented dataset including appended information on the low dose component, the distribution of the two actives, the two main excipients and the low dose lubricant are correctly recovered.
Data generated by analytical instruments, such as spectrometers, may contain unwanted variation due to measurement mode, sample state and other external physical, chemical and environmental factors. ...Preprocessing is required so that the property of interest can be predicted correctly. Different correction methods may remove specific types of artefacts while still leaving some effects behind. Using multiple preprocessing in a complementary way can remove the artefacts that would be left behind by using only one technique. This article summarizes the recent developments in new data preprocessing strategies and specifically reviews the emerging ensemble approaches to preprocessing fusion in chemometrics. A demonstration case is also presented. In summary, ensemble preprocessing allows the selection of several techniques and their combinations that, in a complementary way, lead to improved models. Ensemble approaches are not limited to spectral data but can be used in all cases where preprocessing is needed and identification of a single best option is not easily done.

We have previously reported that the female genital tract (FGT) of Beninese HIV highly-exposed seronegative (HESN) commercial sex workers (CSWs), presented elevated frequencies of a myeloid ...HLA-DR+CD14+CD11c+ population presenting "tolerogenic" monocyte derived dendritic cells (MoDC) features. In order to assess whether a differential profile of monocytes may be involved in the generation of these genital MoDCs, we have herein characterized the blood monocyte compartment of Beninese HESNs (HIV-uninfected ≥ 10 years CSWs) and relevant controls (HIV-uninfected 2.5-5 years CSWs herein termed "early HESNs"), HIV-infected CSWs, and low-risk HIV-uninfected women from the general population. Transcriptomic analyses by RNA-Seq of total sorted blood monocytes demonstrate that in comparison to the control groups, HESNs present increased expression levels of FCGR2C, FCAR, ITGAX, ITGAM, CR2, CD68, and CD163 genes, associated with effector functions. Moreover, we found increased expression levels of genes associated with protection/control against SHIV/HIV such as CCL3, CCL4, CCL5, BHLHE40, and TNFSF13, as well as with immune regulation such as IL-10, Ahr, CD83, and the orphan nuclear receptor (NR)4A1, NR4A2, and NR4A3. Through multicolor flow cytometry analyses, we noticed that the frequencies of intermediate and non-classical monocyte populations tended to be elevated in the blood of HESNs, and exhibited increased expression levels of effector CD16, CD11c, CD11b, as well as regulatory HLA-G, IL-10, and IFN-α markers when compared to HIV-uninfected women and/or HIV-infected CSWs. This profile is compatible with that previously reported in the FGT of HESNs, and likely confers an enormous advantage in their resistance to HIV infection.
Human leukocyte antigen (HLA)-G acts as negative regulator of the immune responses and its expression may enable tumor cells to escape immunosurveillance. The purpose of this study was to investigate ...the influence of HLA-G allelic variants and serum soluble HLA-G (sHLA-G) levels on risk of non-small-cell lung cancer (NSCLC). We analyzed 191 Caucasian adults with NSCLC and 191 healthy subjects recruited between January 2009 and March 2014 in Ariana (Tunisia). Serum sHLA-G levels were measured by immunoassay and HLA-G alleles were determined using a direct DNA sequencing procedures. The heterozygous genotypes of HLA-G 010101 and -G 010401 were associated with increased risks of both NSCLC and advanced disease stages. In contrast, the heterozygous genotypes of HLA-G 0105N and -G 0106 were associated with decreased risks of NSCC and clinical disease stage IV, respectively. Serum sHLA-G levels were significantly higher in patients with NSCLC and particularly in those with advanced disease stages compared to healthy subjects. The area under the receiver-operating characteristic (ROC) curves was 0.82 for controls vs patients. Given 100% specificity, the highest sensitivity achieved to detect NSCLC was 52.8% at a cutoff value of 24.9 U/ml. Patients with the sHLA-G above median level (≥ 50 U/ml) had a significantly shorter survival time. This study demonstrates that HLA-G allelic variants are independent risk factors for NSCLC. Serum sHLA-G levels in NSCLC patients could be useful biomarkers for the diagnostic and prognosis of NSCLC.
Sugar beet is the second biggest world contributor to sugar production and the only one grown in Europe. One of the main limitations for its competitiveness is the lack of effective tools for ...assessing sugar content in unprocessed sugar beet roots, especially in breeding programs. In this context, a dedicated near infrared (NIR) fiber-optic probe based approach is proposed. NIR technology is widely used for the estimation of sugar content in vegetable products, while optic fibers allow a wide choice of technical properties and configurations. The objective of this research was to study the best architecture through different technical choices for the estimation of sugar content in intact sugar beet roots. NIR spectral measurements were taken on unprocessed sugar beet samples using two types of geometries, single and multiple fiber-probes. Sugar content estimates were more accurate when using multiple fiber-probes (up to R
= 0.93) due to a lesser disruption of light specular reflection. In turn, on this configuration, the best estimations were observed for the smallest distances between emitting and collecting fibers, reducing the proportion of multiply scattered light in the spectra. Error of prediction (RPD) values of 3.95, 3.27 and 3.09 were obtained for distances between emitting and collecting fibers of 0.6, 1.2 and 1.8 µm respectively. These high RPD values highlight the good predictions capacities of the multi-fiber probes. Finally, this study contributes to a better understanding of the effects of the technical properties of optical fiber-probes on the quality of spectral models. In addition, and beyond this specificity related to sugar beet, these findings could be extended to other turbid media for quantitative optical spectroscopy and eventually to validate considered fiber-optic probe design obtained in this experimental study.
