The rice blast fungus Magnaporthe oryzae elaborates a specialized cell called an appressorium, which is used to breach the tough outer cuticle of a rice leaf, enabling the fungus entry to ...host plant cells. The appressorium generates enormous turgor by accumulating glycerol to very high concentrations within the cell. Glycerol accumulation and melanization of the appressorium cell wall collectively drive turgor‐mediated penetration of the rice leaf. In this review, we discuss the potential metabolic sources of glycerol in the rice blast fungus and how appressorium turgor is focused as physical force at the base of the infection cell, leading to the formation of a rigid penetration peg. We review recent studies of M. oryzae and other relevant appressorium‐forming fungi which shed light on how glycerol is synthesized and how appressorium turgor is regulated. Finally, we provide some questions to guide avenues of future research that will be important in fully understanding the role of glycerol in rice blast disease.
To cause nee blast disease, the fungus Magnaporthe oryzae develops a pressurized dome-shaped cell called an appressorium, which physically ruptures the leaf cuticle to gain entry to plant tissue. ...Here, we report that a toroidal F-actin network assembles in the appressorium by means of four septin guanosine triphosphatases, which polymerize into a dynamic, hetero-oligomeric ring. Septins scaffold F-actin, via the ezrin-radixin-moesin protein Tea1, and phosphatidylinositide interactions at the appressorium plasma membrane. The septin ring assembles in a Cdc42-and Chm1-dependent manner and forms a diffusion barrier to localize the inverse-bin-amphiphysin-RVS-domain protein Rvs167 and the Wiskott-Aldrich syndrome protein Las17 at the point of penetration. Septins thereby provide the cortical rigidity and membrane curvature necessary for protrusion of a rigid penetration peg to breach the leaf surface.

The electropolymerisation of aniline is a well-studied and often used technology. While the mechanism has been investigated in a variety of media these have all concentrated on understanding the process on a molecular level. Anomalies in the electropolymerisation of aniline in four deep eutectic solvents, DESs, using urea, ethylene glycol, glycerol and oxalic acids as hydrogen bond donors with choline chloride led to an investigation of the aniline phase behaviour. It was only with oxalic acid that polymerisation was achieved and adjusting the pH of the other DESs using sulphuric acid did not enable polymer formation suggesting that pH was not the only factor enabling polymer growth. When 10 wt% water was added, polymers could be grown in all the DESs despite negligible change in solution pH. Dynamic light scattering showed that polymer only formed in systems where aniline formed an emulsion. Scanning electron microscopy, SEM and atomic force microscopy, AFM showed that the polyaniline films were formed of an agglomeration of small particles of the same dimensions as the dispersed monomer phase in solution. This suggests that the droplets of the monomer arrive at the electrode surface where they polymerise. This provides the first evidence that polyaniline grows by an emulsion polymerisation mechanism even in aqueous solutions.

The oceans represent a significant global source of atmospheric aerosols. Sea spray aerosol (SSA) particles comprise sea salts and organic species in varying proportions. In addition to size, the ...overall composition of SSA particles determines how effectively they can form cloud droplets and ice crystals. Thus, understanding the factors controlling SSA composition is critical to predicting aerosol impacts on clouds and climate. It is often assumed that submicrometer SSAs are mainly formed by film drops produced from bursting bubble-cap films, which become enriched with hydrophobic organic species contained within the sea surface microlayer. In contrast, jet drops formed from the base of bursting bubbles are postulated to mainly produce larger supermicrometer particles from bulk seawater, which comprises largely salts and water-soluble organic species. However, here we demonstrate that jet drops produce up to 43% of total submicrometer SSA number concentrations, and that the fraction of SSA produced by jet drops can be modulated by marine biological activity. We show that the chemical composition, organic volume fraction, and ice nucleating ability of submicrometer particles from jet drops differ from those formed from film drops. Thus, the chemical composition of a substantial fraction of submicrometer particles will not be controlled by the composition of the sea surface microlayer, a major assumption in previous studies. This finding has significant ramifications for understanding the factors controlling the mixing state of submicrometer SSA particles and must be taken into consideration when predicting SSA impacts on clouds and climate.
Plants use pattern recognition receptors to defend themselves from microbial pathogens. These receptors recognize pathogen-associated molecular patterns (PAMPs) and activate signaling pathways that ...lead to immunity. In rice (Oryza sativa), the chitin elicitor binding protein (CEBiP) recognizes chitin oligosaccharides released from the cell walls of fungal pathogens. Here, we show that the rice blast fungus Magnaporthe oryzae overcomes this first line of plant defense by secreting an effector protein, Secreted LysM Protein1 (Slp1), during invasion of new rice cells. We demonstrate that Slp1 accumulates at the interface between the fungal cell wall and the rice plasma membrane, can bind to chitin, and is able to suppress chitin-induced plant immune responses, including generation of reactive oxygen species and plant defense gene expression. Furthermore, we show that Slp1 competes with CEBiP for binding of chitin oligosaccharides. Slp1 is required by M. oryzae for full virulence and exerts a significant effect on tissue invasion and disease lesion expansion. By contrast, gene silencing of CEBiP in rice allows M. oryzae to cause rice blast disease in the absence of Slp1. We propose that Slp1 sequesters chitin oligosaccharides to prevent PAMP-triggered immunity in rice, thereby facilitating rapid spread of the fungus within host tissue.

We constrain the Hubble constant H0 using Fast Radio Burst (FRB) observations from the Australian Square Kilometre Array Pathfinder (ASKAP) and Murriyang (Parkes) radio telescopes. We use ...the redshift-dispersion measure (‘Macquart’) relationship, accounting for the intrinsic luminosity function, cosmological gas distribution, population evolution, host galaxy contributions to the dispersion measure (DMhost), and observational biases due to burst duration and telescope beamshape. Using an updated sample of 16 ASKAP FRBs detected by the Commensal Real-time ASKAP Fast Transients (CRAFT) Survey and localized to their host galaxies, and 60 unlocalized FRBs from Parkes and ASKAP, our best-fitting value of H0 is calculated to be $73_{-8}^{+12}$ km s−1 Mpc−1. Uncertainties in FRB energetics and DMhost produce larger uncertainties in the inferred value of H0 compared to previous FRB-based estimates. Using a prior on H0 covering the 67–74 km s−1 Mpc−1 range, we estimate a median ${\rm DM}_{\rm host}= 186_{-48}^{+59}\,{\rm pc \, cm^{-3}}$, exceeding previous estimates. We confirm that the FRB population evolves with redshift similarly to the star-formation rate. We use a Schechter luminosity function to constrain the maximum FRB energy to be log10Emax$=41.26_{-0.22}^{+0.27}$ erg assuming a characteristic FRB emission bandwidth of 1 GHz at 1.3 GHz, and the cumulative luminosity index to be $\gamma =-0.95_{-0.15}^{+0.18}$. We demonstrate with a sample of 100 mock FRBs that H0 can be measured with an uncertainty of ±2.5 km s−1 Mpc−1, demonstrating the potential for clarifying the Hubble tension with an upgraded ASKAP FRB search system. Last, we explore a range of sample and selection biases that affect FRB analyses.
Animal experiments remain essential to understand the fundamental mechanisms underpinning malignancy and to discover improved methods to prevent, diagnose and treat cancer. Excellent standards of ...animal care are fully consistent with the conduct of high quality cancer research. Here we provide updated guidelines on the welfare and use of animals in cancer research. All experiments should incorporate the 3Rs: replacement, reduction and refinement. Focusing on animal welfare, we present recommendations on all aspects of cancer research, including: study design, statistics and pilot studies; choice of tumour models (e.g., genetically engineered, orthotopic and metastatic); therapy (including drugs and radiation); imaging (covering techniques, anaesthesia and restraint); humane endpoints (including tumour burden and site); and publication of best practice.
