In the early summer of 2012, sudden mass mortality among songbirds, particularly in greenfinches (
, syn:
) was observed in Austria, which was caused by the protozoan parasite
. This pathogen induced ...fibrinonecrotic ingluvitis and/or esophagitis, leading to impairment of food intake and ultimately death due to starvation. The pathogen was successfully detected within the lesions by polymerase chain reaction (PCR) and chromogenic in situ hybridization. The epizootic resulted in a significant decline in the Austrian greenfinch population. Continuing passive surveillance in the subsequent years (2013-2020) revealed that the condition occurred each year and was present in the entire country. Genetic characterization of the pathogen showed the presence of an identical strain irrespective of geographical location, bird species, and year.
Human activities often cause changes and homogenization in landscape structure. To investigate the impact of changing cultivation systems on structural and functional aspects of a tropical ...agroforestry system, we developed satellite based land cover maps of the La Gamba area in southwestern Costa Rica and refined them by mapping fine-scale linear landscape elements. Performing a landscape pattern analysis, we compared eight sections of the study area by landscape metrics. Furthermore we performed a Morphological spatial pattern analysis (MSPA) for hypothetical non-forest, forest generalist and forest specialist species, and compared the current situation to a possible future scenario with double the area covered by oil palm plantations after a virtual conversion of other agricultural patches. The heterogeneous rural sections clearly differed from the homogeneous forests and especially pasture-dominated rural sections included many diverse, small and elongated patches, many linear landscape elements but few big plantations. According to the scenario with double the area covered by oil palm plantations, non-forest species lost large parts of their habitat, while forest species mainly lost corridors. The protection of natural landscape elements that support wildlife movement between forest areas is of major importance, particularly as the globally increasing cultivation of oil palm is significantly altering many tropical land mosaics, including the countryside of La Gamba. We propose the establishment of eight least cost path corridor routes in the study area to make the agricultural area pervious for wildlife.
Rivers and streams lined by narrow forest strips are common in the lowland countryside of south-western Costa Rica. We studied the importance of these gallery forests for understorey birds, ...especially forest species. Using mist-nets, we captured 1110 birds belonging to 90 species between June and September 2007 at 16 sites spread equally over four habitat types: forest interior, forest margin, gallery forest connected to closed forest, and isolated gallery forest. Though isolated gallery forests had the greatest number of expected species in total, they supported the lowest number of forest-specialist species, lower even than connected gallery forests. Granivorous birds showed an increase from forest to isolated gallery forest, all other feeding guilds however showed no change. The studied habitats differed from each other in their faunal composition. Assemblages of three species groups categorised by their habitat preferences (forest specialists, forest generalists, non-forest species) showed nested distribution patterns across the four habitats. There was no significant difference in the proportion of birds with brood patches or of recaptures across the studied habitat types, leading to the assumption that birds not only use gallery forests for movement and foraging but also for reproduction. Though of limited conservation value for most forest understorey birds, at least for a small fraction of forest species gallery forests constitute an important secondary habitat. More significantly, they can form corridors or stepping stones that allow movements within the matrix of human-dominated habitats, and represent an important landscape component benefiting total bird species richness in the Costa Rican countryside.
Declines in European bird populations are reported for decades but the direct effect of major anthropogenic pressures on such declines remains unquantified. Causal relationships between pressures and ...bird population responses are difficult to identify as pressures interact at different spatial scales and responses vary among species. Here, we uncover direct relationships between population time-series of 170 common bird species, monitored at more than 20,000 sites in 28 European countries, over 37 y, and four widespread anthropogenic pressures: agricultural intensification, change in forest cover, urbanisation and temperature change over the last decades. We quantify the influence of each pressure on population time-series and its importance relative to other pressures, and we identify traits of most affected species. We find that agricultural intensification, in particular pesticides and fertiliser use, is the main pressure for most bird population declines, especially for invertebrate feeders. Responses to changes in forest cover, urbanisation and temperature are more species-specific. Specifically, forest cover is associated with a positive effect and growing urbanisation with a negative effect on population dynamics, while temperature change has an effect on the dynamics of a large number of bird populations, the magnitude and direction of which depend on species' thermal preferences. Our results not only confirm the pervasive and strong effects of anthropogenic pressures on common breeding birds, but quantify the relative strength of these effects stressing the urgent need for transformative changes in the way of inhabiting the world in European countries, if bird populations shall have a chance of recovering.
Identifying climate refugia is key to effective biodiversity conservation under a changing climate, especially for mountain‐specialist species adapted to cold conditions and highly threatened by ...climate warming. We combined species distribution models (SDMs) with climate forecasts to identify climate refugia for high‐elevation bird species (Lagopus muta, Anthus spinoletta, Prunella collaris, Montifringilla nivalis) in the European Alps, where the ecological effects of climate changes are particularly evident and predicted to intensify. We considered future (2041–2070) conditions (SSP585 scenario, four climate models) and identified three types of refugia: (1) in‐situ refugia potentially suitable under both current and future climate conditions, ex‐situ refugia suitable (2) only in the future according to all future conditions, or (3) under at least three out of four future conditions. SDMs were based on a very large, high‐resolution occurrence dataset (2901–12,601 independent records for each species) collected by citizen scientists. SDMs were fitted using different algorithms, balancing statistical accuracy, ecological realism and predictive/extrapolation ability. We selected the most reliable ones based on consistency between training and testing data and extrapolation over distant areas. Future predictions revealed that all species (with the partial exception of A. spinoletta) will undergo a range contraction towards higher elevations, losing 17%–59% of their current range (larger losses in L. muta). We identified ~15,000 km2 of the Alpine region as in‐situ refugia for at least three species, of which 44% are currently designated as protected areas (PAs; 18%–66% among countries). Our findings highlight the usefulness of spatially accurate data collected by citizen scientists, and the importance of model testing by extrapolating over independent areas. Climate refugia, which are only partly included within the current PAs system, should be priority sites for the conservation of Alpine high‐elevation species and habitats, where habitat degradation/alteration by human activities should be prevented to ensure future suitability for alpine species.

Broadly neutralizing HIV antibodies are much sought after (a) to guide vaccine design, both as templates and as indicators of the authenticity of vaccine candidates, (b) to assist in structural ...studies, and (c) to serve as potential therapeutics. However, the number of targets on the viral envelope spike for such antibodies has been limited. Here, we describe a set of human monoclonal antibodies that define what is, to the best of our knowledge, a previously undefined target on HIV Env. The antibodies recognize a glycan-dependent epitope on the prefusion conformation of gp41 and unambiguously distinguish cleaved from uncleaved Env trimers, an important property given increasing evidence that cleavage is required for vaccine candidates that seek to mimic the functional HIV envelope spike. The availability of this set of antibodies expands the number of vaccine targets on HIV and provides reagents to characterize the native envelope spike.

Acrylamide in food products-chiefly in commercially available potato chips, potato fries, cereals, and bread-was determined by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Samples were ...homogenized with water/dichloromethane, centrifuged, and filtered through a 5 kDa filter. The filtrate was cleaned up on mixed mode, anion and cation exchange (Oasis MAX and MCX) and carbon (Envirocarb) cartridges. Analysis was done by isotope dilution (D(3)- or (13)C(3)acrylamide) electrospray LC-MS/MS using a 2 x 150 mm (or 2 x 100 mm) Thermo HyperCarb column eluted with 1 mM ammonium formate in 15% (or 10% for the 2 x 100 mm column) methanol. Thirty samples of foods were analyzed. Concentrations of acrylamide varied from 14 ng/g (bread) to 3700 ng/g (potato chips). Acrylamide was formed during model reactions involving heating of mixtures of amino acids and glucose in ratios similar to those found in potatoes. In model reactions between amino acids and glucose, asparagine was found to be the main precursor of acrylamide. Thus, in the reaction between nitrogen-15 (amido)-labeled asparagine and glucose, corresponding (15)N-labeled acrylamide was formed. The yield of the model reaction is approximately 0.1%.
