New knowledge about the gut microbiota and its interaction with the host's metabolic regulation has emerged during the last few decades. Several factors may affect the composition of the gut ...microbiota, including dietary fiber. Dietary fiber is not hydrolyzed by human digestive enzymes, but it is acted upon by gut microbes, and metabolites like short-chain fatty acids are produced. The short-chain fatty acids may be absorbed into the circulation and affect metabolic regulation in the host or be a substrate for other microbes. Some studies have shown improved insulin sensitivity, weight regulation, and reduced inflammation with increases in gut-derived short-chain fatty acids, all of which may reduce the risk of developing metabolic diseases. To what extent a dietary intervention with fiber may affect the human gut microbiota and hence metabolic regulation, is however, currently not well described. The aim of the present review is to summarize recent research on human randomized, controlled intervention studies investigating the effect of dietary fiber on gut microbiota and metabolic regulation. Metabolic regulation is discussed with respect to markers relating to glycemic regulation and lipid metabolism. Taken together, the papers on which the current review is based, suggest that dietary fiber has the potential to change the gut microbiota and alter metabolic regulation. However, due to the heterogeneity of the studies, a firm conclusion describing the causal relationship between gut microbiota and metabolic regulation remains elusive.
Gut microbiota have recently been suggested to play a part in low-grade systemic inflammation, which is considered a key risk factor for cardiometabolic disorders. Diet is known to affect gut ...microbiota; however, the effects of diet and dietary components on gut microbiota and inflammation are not fully understood. In the present review, we summarize recent research on human dietary intervention studies, investigating the effects of healthy diets or dietary components on gut microbiota and systemic inflammation. We included 18 studies that reported how different dietary components altered gut microbiota composition, short-chain fatty acid levels, and/or inflammatory markers. However, the heterogeneity among the intervention studies makes it difficult to conclude whether diets or dietary components affect gut microbiota homeostasis and inflammation. More appropriately designed studies are needed to better understand the effects of diet on the gut microbiota, systemic inflammation, and risk of cardiometabolic disorders.
CAD/CAM milling systems provide a rapid and individual method for the manufacturing of zirconia dental restorations. However, the disadvantages of these systems include limited accuracy, possible ...introduction of microscopic cracks, and a waste of material due to the principle of the ‘subtractive process’. The hypothesis of this study was that these issues can be overcome by a novel generative manufacturing technique, direct inkjet printing. A tailored zirconia-based ceramic suspension with 27 vol% solid content was synthesized. The suspension was printed on a conventional, but modified, drop-on-demand inkjet printer. A cleaning unit and a drying device allowed for the build-up of dense components of the size of a posterior crown. A characteristic strength of 763 MPa and a mean fracture toughness of 6.7 MPam0.5 were determined on 3D-printed and subsequently sintered specimens. The novel technique has great potential to produce, cost-efficiently, all-ceramic dental restorations at high accuracy and with a minimum of materials consumption.
The impact of dietary fat on the risk of cardiovascular disease (CVD) has been extensively studied in recent decades. Solid evidence indicates that replacing saturated fatty acids (SFAs) with ...polyunsaturated fatty acids (PUFAs) decreases blood cholesterol levels and prevents CVD and CVD mortality. Studies indicate that fat quality also may affect insulin sensitivity and hence, the risk of type 2 diabetes (T2D). A high intake of SFAs has shown to increase the risk of T2D in prospective studies, while a high intake of PUFAs reduces the risk. Whether PUFAs from marine or vegetable sources affect glycemic regulation differently in T2D remains to be elucidated. The aim of the present review was therefore to summarize research on human randomized, controlled intervention studies investigating the effect of dietary PUFAs on glycemic regulation in T2D. About half of the studies investigating the effect of fish, fish oils, vegetable oils, or nuts found changes related to glycemic control in people with T2D, while the other half found no effects. Even though some of the studies used SFA as controls, the majority of the included studies compared PUFAs of different quality. Considering that both marine and vegetable oils are high in PUFAs and hence both oils may affect glycemic regulation, the lack of effect in several of the included studies may be explained by the use of an inappropriate control group. It is therefore not possible to draw a firm conclusion, and more studies are needed.

The 15-Country Collaborative Study of Cancer Risk among Radiation Workers in the Nuclear Industry: Estimates of Radiation-Related Cancer Risks. Radiat. Res. 167, 396– 416 (2007). A 15-Country collaborative cohort study was conducted to provide direct estimates of cancer risk following protracted low doses of ionizing radiation. Analyses included 407,391 nuclear industry workers monitored individually for external radiation and 5.2 million person-years of follow-up. A significant association was seen between radiation dose and all-cause mortality excess relative risk (ERR) 0.42 per Sv, 90% CI 0.07, 0.79; 18,993 deaths. This was mainly attributable to a dose-related increase in all cancer mortality (ERR/Sv 0.97, 90% CI 0.28, 1.77; 5233 deaths). Among 31 specific types of malignancies studied, a significant association was found for lung cancer (ERR/Sv 1.86, 90% CI 0.49, 3.63; 1457 deaths) and a borderline significant (P = 0.06) association for multiple myeloma (ERR/Sv 6.15, 90% CI <0, 20.6; 83 deaths) and ill-defined and secondary cancers (ERR/Sv 1.96, 90% CI −0.26, 5.90; 328 deaths). Stratification on duration of employment had a large effect on the ERR/Sv, reflecting a strong healthy worker survivor effect in these cohorts. This is the largest analytical epidemiological study of the effects of low-dose protracted exposures to ionizing radiation to date. Further studies will be important to better assess the role of tobacco and other occupational exposures in our risk estimates.
Background Ionizing radiation at very high (radio-therapeutic) dose levels can cause diseases other than cancer, particularly heart diseases. There is increasing evidence that doses of the order of a ...few sievert (Sv) may also increase the risk of non-cancer diseases. It is not known, however, whether such effects also occur following the lower doses and dose rates of public health concern. Methods We used data from an international (15-country) nuclear workers cohort study to evaluate whether mortality from diseases other than cancer is related to low doses of external ionizing radiation. Analyses included 275 312 workers with adequate information on socioeconomic status, over 4 million person-years of follow-up and an average cumulative radiation dose of 20.7 mSv; 11 255 workers had died of non-cancer diseases. Results The excess relative risk (ERR) per Sv was 0.24 95% CI (confidence intervals) −0.23, 0.78 for mortality from all non-cancer diseases and 0.09 (95% CI −0.43, 0.70) for circulatory diseases. Higher risk estimates were observed for mortality from respiratory and digestive diseases, but confidence intervals included zero. Increased risks were observed among the younger workers (attained age <50 years, identified post hoc) for all groupings of non-cancer causes of death, including external causes. It is unclear therefore whether these findings reflect real effects of radiation, random variation or residual confounding. Conclusions The most informative low-dose radiation study to date provides little evidence for a relationship between mortality from non-malignant diseases and radiation dose. However, we cannot rule out risks per unit dose of the same order of magnitude as found in studies at higher doses.
Confocal Raman microscopic (CRM) imaging has evolved to become a key tool for spatially resolved, compositional analysis and imaging, down to the μm-scale, and nowadays one may choose between ...numerous commercial instruments. That notwithstanding, situations may arise which exclude the use of a commercial instrument, e.g., if the analysis involves toxic or radioactive samples/environments; one may not wish to render an expensive instrument unusable for other uses, due to contamination. Therefore, custom-designed CRM instrumentation-being adaptable to hazardous conditions and providing operational flexibility-may be beneficial. Here, we describe a CRM setup, which is constructed nearly in its entirety from off-the-shelf optomechanical and optical components. The original aim was to develop a CRM suitable for the investigation of samples exposed to tritium. For increased flexibility, the CRM system incorporates optical fiber coupling to both the Raman excitation laser and the spectrometer. Lateral raster scans and axial profiling of samples are facilitated by the use of a motorized xyz-translation assembly. Besides the description of the construction and alignment of the CRM system, we also provide (i) the experimental evaluation of system performance (such as, e.g., spatial resolution) and (ii) examples of Raman raster maps and axial profiles of selected thin-film samples (such as, e.g., graphene sheets).
