Nanosafety is paramount considering the risks associated with manufactured nanomaterials (MNMs) whose implications could outweigh their advantages for environmental applications. Although ...nanotechnology-based solutions to implement pollution control, remediation and prevention are incremental with clear benefits for public health and Earth’ natural ecosystems, nanoremediation is having a setback due to the risks associated with the safety of MNMs for humans and the environment. MNMs are diverse, work differently and bionano-interactions occurring upon environmental exposure will guide their fate and hazardous outcomes. Here we propose a new ecologically-based design strategy (eco-design) having its roots in green nanoscience and LCA that will ground on an Ecological Risk Assessment approach, which introduces the evaluation of MNMs' ecotoxicity along with their performances and efficacies at the design stage. As such, the proposed eco-design strategy will allow recognition and design-out since the very beginning of material synthesis, those hazardous peculiar features that can be hazardous to living beings and the natural environment. A more ecologically sound eco-design strategy in which nanosafety is conceptually included in MNMs design will sustain safer nanotechnologies including those for the environment as remediation by leveraging any risks for humans and natural ecosystems.

Nanosponges (NSs) are a new age branched cyclodextrin (CD) polymeric systems exhibiting tremendous potential in pharmaceutical, agro science, and biomedical applications. Over the past decade, ...different varieties of NS based on the type of CD and the crosslinker have been developed tailored for specific applications. NS technology has been instrumental in achieving solubilization, stabilization, sustained release, enhancement of activity, permeability enhancement, protein delivery, ocular delivery, stimuli sensitive drug release, enhancement of bioavailability, etc. There is a major explosion of research in the area of NS‐aided cancer therapeutics. A wide of anticancer molecules both from a pharmacological and physicochemical perspective have been developed as NS formulations by several groups including ours. Our objective in this review is to capture a systematic and comprehensive snapshot of the state‐of‐the‐art of NS‐aided cancer therapeutics reported so far. This review will provide an ideal platform for both the formulation scientists working on new polymeric/drug development and cancer biologists/scientists to understand the current nanotechnologies in CD‐based NS‐aided cancer therapeutics. The scope of the review is limited to small molecules and CD‐based NS. The review covers in detail the problems associated with anticancer small molecules, and the solution provided by CD‐based NS specifically for camptothecin, curcumin, paclitaxel, tamoxifen, resveratrol, quercetin, oxygen‐NS, temozolomide, doxorubicin, and 5‐Fluorouracil. Cyclodextrin-based nanosponges, which are proposed as a new nanosized delivery system, are innovative cross-linked cyclodextrin polymers nanostructured within a three-dimensional network. This type ...of cyclodextrin polymer can form porous insoluble nanoparticles with a crystalline or amorphous structure and spherical shape or swelling properties. The polarity and dimension of the polymer mesh can be easily tuned by varying the type of cross-linker and degree of cross-linking. Nanosponge functionalisation for site-specific targeting can be achieved by conjugating various ligands on their surface. They are a safe and biodegradable material with negligible toxicity on cell cultures and are well-tolerated after injection in mice. Cyclodextrin-based nanosponges can form complexes with different types of lipophilic or hydrophilic molecules. The release of the entrapped molecules can be varied by modifying the structure to achieve prolonged release kinetics or a faster release. The nanosponges could be used to improve the aqueous solubility of poorly water-soluble molecules, protect degradable substances, obtain sustained delivery systems or design innovative drug carriers for nanomedicine.
Pedestrian detection and tracking have become an important field in the computer vision research area. This growing interest, started in the last decades, might be explained by the multitude of ...potential applications that could use the results of this research field, e.g. robotics, entertainment, surveillance, care for the elderly and disabled, and content-based indexing. In this survey paper, vision-based pedestrian detection systems are analysed based on their field of application, acquisition technology, computer vision techniques and classification strategies. Three main application fields have been individuated and discussed: video surveillance, human-machine interaction and analysis. Due to the large variety of acquisition technologies, this paper discusses both the differences between 2D and 3D vision systems, and indoor and outdoor systems. The authors reserved a dedicated section for the analysis of the Deep Learning methodologies, including the Convolutional Neural Networks in pedestrian detection and tracking, considering their recent exploding adoption for such a kind systems. Finally, focusing on the classification point of view, different Machine Learning techniques have been analysed, basing the discussion on the classification performances on different benchmark datasets. The reported results highlight the importance of testing pedestrian detection systems on different datasets to evaluate the robustness of the computed groups of features used as input to classifiers.
Babchi (
) oil is an important essential oil used in several traditional medicines to cure various disorders. This phytotherapeutic agent possesses a number of pharmacological activities including ...antibacterial, antifungal, antioxidant, anti-inflammatory, immunomodulatory, and antitumor factors. However, volatile nature, poor stability, and solubility of babchi oil (BO) restrict its pharmaceutical applications. Therefore, the aim of the present work was to encapsulate this oil in β-cyclodextrin nanosponges (NS) in order to overcome the above limitations. To fabricate nanosponges, β-cyclodextrin was cross-linked with diphenyl carbonate in different molar ratios viz. 1:2, 1:4, 1:6, 1:8, and 1:10. The blank nanosponges were loaded with BO using the freeze-drying method. The particle size of the BO loaded nanosponges was found to lie between 200 and 500 nm with low polydispersity index. Furthermore, the zeta potential, the Fourier transform infrared spectroscopy, X-ray diffraction, thermal analysis, and electron microscopy were carried out for characterization of BO nanosponges. Results obtained from spectral analysis ascertained the formation of inclusion complexes. Additionally, solubilisation efficiency of BO was checked in distilled water and found enhanced by 4.95 times with optimized β-cyclodextrin nanosponges. The cytotoxicity study was carried out by the MTT assay using HaCaT cell lines. A significant improvement in photo-stability of essential oil was also observed by inclusion innanosponges. Camptothecin (CAM), a plant alkaloid and a potent antitumor agent, has a limited therapeutic utility because of its poor aqueous solubility, lactone ring instability and serious side effects. ...Cyclodextrin-based nanosponges (NS) are a novel class of cross-linked derivatives of cyclodextrins. They have been used to increase the solubility of poorly soluble actives, to protect the labile groups and control the release. This study aimed at formulating complexes of CAM with three types of β-cyclodextrin NS obtained with different cross-linking ratio (viz. 1:2, 1:4 and 1:8 on molar basis with the cross-linker) to protect the lactone ring from hydrolysis and to prolong the release kinetics of CAM. h with a 80% w/w of intact lactone ring when compared to only around 20% w/w of plain CAM. The cytotoxicity studies on HT-29 cells showed that the CAM formulations were more cytotoxic than plain CAM after 24
h of incubation.
The cross-linking density influences the physicochemical properties of cyclodextrin-based nanosponges (CD-NSs). Although the effect of the cross-linker type and content on the NSs performance has ...been investigated, a detailed study of the cross-linking density has never been performed. In this contribution, nine ester-bridged NSs based on β-cyclodextrin (β-CD) and different quantities of pyromellitic dianhydride (PMDA), used as a cross-linking agent in stoichiometric proportions of 2, 3, 4, 5, 6, 7, 8, 9, and 10 moles of PMDA for each mole of CD, were synthesized and characterized in terms of swelling and rheological properties. The results, from the swelling experiments, exploiting Flory-Rehner theory, and rheology, strongly showed a cross-linker content-dependent behavior. The study of cross-linking density allowed to shed light on the efficiency of the synthesis reaction methods. Overall, our study demonstrates that by varying the amount of cross-linking agent, the cross-linked structure of the NSs matrix can be controlled effectively. As PMDA βCD-NSs have emerged over the years as a highly versatile class of materials with potential applications in various fields, this study represents the first step towards a full understanding of the correlation between their structure and properties, which is a key requirement to effectively tune their synthesis reaction in view of any specific future application or industrial scale-up.
The ability to employ waste products, such as vegetable scraps, as raw materials for the synthesis of new promising adsorbing materials is at the base of the circular economy and end of waste ...concepts. Dextrin-based nanosponges (D_NS), both cyclodextrin (CD) and maltodextrin (MD), have shown remarkable adsorption abilities in the removal of toxic compounds from water and wastewater, thus representing a bio-based low-cost solution which is establishing itself in the market. Nevertheless, their environmental safety for either aquatic or terrestrial organisms has been overlooked, raising concern in terms of potential hazards to natural ecosystems. Here, the environmental safety (ecosafety) of six newly synthesized batches of D_NS was determined along with their full characterization by means of dynamic light scattering (DLS), thermogravimetric analysis (TGA), Fourier transformed infrared spectroscopy with attenuated total reflection (FTIR-ATR) and transmission electron microscopy (SEM). Ecotoxicity evaluation was performed using a battery of model organisms and ecotoxicity assays, such as the microalgae growth inhibition test using the freshwater Raphidocelis subcapitata and the marine diatom Dunaliella tertiolecta, regeneration assay using the freshwater cnidarian Hydra vulgaris and immobilization assay with the marine brine shrimp Artemia franciscana. Impact on seedling germination of a terrestrial plant of commercial interest, Cucurbita pepo was also investigated. Ecotoxicity data showed mild to low toxicity of the six batches, up to 1 mg/mL, in the following order: R. subcapitata >H. vulgaris >D. tertiolecta >A. franciscana > C. pepo. The only exception was represented by one batch (NS-Q+_BDE_(GLU2) which resulted highly toxic for both freshwater species, R. subcapitata and H. vulgaris. Those criticalities were solved with the synthesis of a fresh new batch and were hence attributed to the single synthesis and not to the specific D_NS formulation. No effect on germination of pumpkin but rather more a stimulative effect was observed. To our knowledge this is the first evaluation of the environmental safety of D_ NS. As such we emphasize that current formulations and exposure levels in the range of mg/mL do not harm aquatic and terrestrial species thus representing an ecosafe solution also for environmental applications.

There is a global perception that psychotropic utilization in children and adolescents is increasing, especially with the onset of COVID-19 pandemic. Available literature data on paediatric ...psychotropic medication prescriptions in Italy are limited to one or few regions and not updated. The aim of this study was to provide updated data on psychotropic prescriptions referred to the whole Italian paediatric population, as overall and by subgroups of medications and to evaluate if the COVID-19 pandemic during 2020 had an impact on prescription rates.
A descriptive study on psychotropic drug utilization in children and adolescents (< 18 years) resident in all Italian regions during 2020 was performed. Patients registered in the Pharmaceutical Prescriptions database with at least one prescription/dispensing of a psychotropic medication (antipsychotics-N05A), (antidepressants-N06A) and (psychostimulants-N06BA) during the study period were considered. The indicators used were the prescription rate (number of prescriptions per 1000 children) and prevalence of use (proportion of the paediatric population with at least one prescription in the relevant year).
During the 2020 the prevalence of psychotropic drug use in the paediatric population was 0.3%, increased of 7.8% if compared to 2019. The same trend was observed for the prescription rate, which recorded an average of 28.2 per 1000 children with an increase of 11.6% if compared to previous year, representing the 0.6% of the overall drug use in this age group. The data showed a growing trend prescription by age, reaching the peak in adolescents aged 12-17 years old, with a prescription rate of 65 per 1000 children and a prevalence of 0.71%. Considering the subgroups of psychotropic medications, the highest prevalence of use was found for antipsychotic drugs, received by the 0.19% of the paediatric population during 2020.
Psychotropic drug utilization in children and adolescents has grown during 2020 in Italy and worldwide, raising alarms from health care clinicians and patient advocates about the increase of burden of mental diseases in paediatric population during the COVID-19 pandemic. A more systematic monitoring of the use of psychotropic medications should be implemented in all countries for collecting relevant information about children and adolescents taking psychotropic drugs, in order to address the present and the future of the mental health of the paediatric population.
