Understanding the causes and consequences of wildfires in forests of the western United States requires integrated information about fire, climate changes, and human activity on multiple temporal ...scales. We use sedimentary charcoal accumulation rates to construct long-term variations in fire during the past 3,000 y in the American West and compare this record to independent fire-history data from historical records and fire scars. There has been a slight decline in burning over the past 3,000 y, with the lowest levels attained during the 20th century and during the Little Ice Age (LIA, ca. 1400–1700 CE Common Era). Prominent peaks in forest fires occurred during the Medieval Climate Anomaly (ca. 950–1250 CE) and during the 1800s. Analysis of climate reconstructions beginning from 500 CE and population data show that temperature and drought predict changes in biomass burning up to the late 1800s CE. Since the late 1800s , human activities and the ecological effects of recent high fire activity caused a large, abrupt decline in burning similar to the LIA fire decline. Consequently, there is now a forest "fire deficit" in the western United States attributable to the combined effects of human activities, ecological, and climate changes. Large fires in the late 20th and 21st century fires have begun to address the fire deficit, but it is continuing to grow.
Fire histories of mid-elevation mixed-conifer forests are uncommon in the eastern Cascades, limiting our understanding of long-term fire dynamics in these environments. The purpose of this study was ...to reconstruct the fire and vegetation history for a moist mid-elevation mixed-conifer site, and to determine whether Holocene fire activity in this watershed was intermediate to fire regimes observed at higher and lower elevations in the eastern Cascades. Fire activity and vegetation change was reconstructed using macroscopic charcoal and pollen analysis of sediment core from Long Lake. This site is located ~45 km west of Yakima, WA, and exists in a grand fir-dominated, mixed-conifer forest. Results show low fire activity from ca. 9870 to 6000 cal yr BP, after which time fire increased and remained frequent until ca. 500 cal yr BP. A woodland environment existed at the site in the early Holocene, with the modern coniferous forest establishing ca. 6000–5500 cal yr BP. A mixed-severity fire regime has existed at the site for the past ~6000 years, with both higher- and lower-severity fire episodes occurring on average every ~80–100 years. However, only one fire episode occurred in the Long Lake watershed during the past 500 years, and none within the past ~150 years. Based on a comparison with other eastern Cascade sites, Holocene fire regimes at Long Lake, particularly during the late Holocene, appear to be intermediate between those observed at higher- and lower elevation sites, both in terms of fire severity and frequency.
Managing and understanding fisheries dynamics are becoming more complex as new and seemingly more complicated environmental factors are identified. Often management requires resources beyond that of ...any one entity and calls for collaboration among partners with differing priorities and backgrounds to account for the complexity of factors influencing fisheries. We present a collaborative case study from the Susquehanna River basin, Pennsylvania, where Smallmouth Bass Micropterus dolomieu have faced population declines, mortality events, and notable signs of disease in recent years. Collaboration was required to study many facets of the fishery and the environment simultaneously to better understand risk factors and underlying relationships influencing Smallmouth Bass health. The outcomes from this interdisciplinary collaboration allowed for identification of contributing risk factors, led to the development of products and analytical techniques that were mutually beneficial to all partners involved, and provided knowledge that was integrated into fish health and fisheries management.
In order to fully appreciate the role that fire, both natural and anthropogenic, had in shaping pre-Euro-American settlement landscapes in the Pacific Northwest (PNW), it is necessary to develop a ...more robust method of evaluating paleofire reconstructions. Here we demonstrate an approach that includes the identification of charcoal morphotypes (i.e. visually distinct charcoal particles), and incorporates both paleoecological and archaeological data sets, to more specifically determine both the nature of past fire regimes (i.e. fuel type and fire severity) and the likely ignition source of those fires. We demonstrate the usefulness of this approach by reconstructing the late Holocene fire and vegetation histories of Lake Oswego (Clackamas County), Oregon, and Fish Lake (Okanogan County), Washington, using macroscopic charcoal and pollen analysis of sediment cores. The histories were compared with climatic records from the PNW as well as archaeological, ethnographic, and historical records from the Lower Columbia River Valley and Southern Columbia Plateau cultural regions. Our results indicate that while centennial-to-millennial-scale climate change had limited influence on the fire regimes at the study sites during the past ∼3800 years, the use of fire by Native Americans for a variety of reasons, particularly after ca. 1200 calendar years before present (AD 750), had a far greater impact. Charcoal morphotype ratios also indicate that fires in the two watersheds were fundamentally different in their severity and impact, and led to major shifts in the forests and woodlands surrounding Lake Oswego, but helped maintain the ponderosa pine-dominated forest at Fish Lake. The elimination of fire from the two study sites during the past 100–300 years is likely the combined result of Euro-American contact and the arrival of disease in the PNW, as well as 20th-century fire suppression and grazing effects on fuel continuity, which has implications for future forest management and restoration efforts in the PNW.
Leucine-rich repeat kinase 2 (LRRK2) is a large, multidomain protein which contains a kinase domain and GTPase domain among other regions. Individuals possessing gain of function mutations in the ...kinase domain such as the most prevalent G2019S mutation have been associated with an increased risk for the development of Parkinson's disease (PD). Given this genetic validation for inhibition of LRRK2 kinase activity as a potential means of affecting disease progression, our team set out to develop LRRK2 inhibitors to test this hypothesis. A high throughput screen of our compound collection afforded a number of promising indazole leads which were truncated in order to identify a minimum pharmacophore. Further optimization of these indazoles led to the development of MLi-2 (1): a potent, highly selective, orally available, brain-penetrant inhibitor of LRRK2.
The robust specification of organ development depends on coordinated cell-cell communication. This process requires signal integration among multiple pathways, relying on second messengers such as ...calcium ions. Calcium signaling encodes a significant portion of the cellular state by regulating transcription factors, enzymes, and cytoskeletal proteins. However, the relationships between the inputs specifying cell and organ development, calcium signaling dynamics, and final organ morphology are poorly understood. Here, we have designed a quantitative image-analysis pipeline for decoding organ-level calcium signaling. With this pipeline, we extracted spatiotemporal features of calcium signaling dynamics during the development of the Drosophila larval wing disc, a genetic model for organogenesis. We identified specific classes of wing phenotypes that resulted from calcium signaling pathway perturbations, including defects in gross morphology, vein differentiation, and overall size. We found four qualitative classes of calcium signaling activity. These classes can be ordered based on agonist stimulation strength Gαq-mediated signaling. In vivo calcium signaling dynamics depend on both receptor tyrosine kinase/phospholipase C γ and G protein-coupled receptor/phospholipase C β activities. We found that spatially patterned calcium dynamics correlate with known differential growth rates between anterior and posterior compartments. Integrated calcium signaling activity decreases with increasing tissue size, and it responds to morphogenetic perturbations that impact organ growth. Together, these findings define how calcium signaling dynamics integrate upstream inputs to mediate multiple response outputs in developing epithelial organs.
The Smallmouth Bass Micropterus dolomieu fishery in the Susquehanna River basin, Pennsylvania, is one of the most socioeconomically important fisheries in the region and has recently undergone ...considerable changes. These changes started in 2005, when disease was documented in young‐of‐the‐year (age‐0) Smallmouth Bass. Shortly thereafter, declines in abundance of both juveniles and adults were observed. These declines in abundance coincided with disease infections in age‐0, intersex in adults, and concerns regarding contaminant exposure. Natural mortality rates, particularly for age‐0, increased during this period (2005–2011), and there were concerns for the overall health of this world‐class fishery. However, in recent years (2012–2017), there have been decreases in both mortality rates and external observations of disease and increases in abundance across multiple size‐classes. Recent changes are encouraging for the future of the Smallmouth Bass fishery in the Susquehanna River. Yet, in light of the ever changing environmental, social, and anthropogenic influences on aquatic ecosystems, there remain concerns for Smallmouth Bass health and management. Because of this, ongoing research efforts are needed to monitor population and health changes and to conduct integrative research that considers complex relationships between organisms and their environments.
A myxozoan parasite, Myxobolus inornatus, is one disease agent identified in young of the year (YOY) smallmouth bass in the Susquehanna River Basin, Pennsylvania. We investigated spatial and temporal ...variability in M. Inornatus prevalence across the Susquehanna River Basin and at several out‐of‐basin sites. We examined potential land use drivers of M. Inornatus prevalence including agricultural and developed land use. In 1,267 YOY smallmouth bass collected from 32 sites during 2013–2016, M. Inornatus was documented in 43.6% of samples. Among‐site variability in parasite prevalence was greater than among‐year variability. The effect of agricultural land use on M. Inornatus prevalence had a high probability of being positively correlated at multiple spatial scales (probability of positive effect > 0.80). The effect of developed land use on M. Inornatus prevalence had a relatively high probability of being negatively correlated at multiple spatial scales (probability of negative effect > 0.70). Our results suggest that land use practices could be related to M. Inornatus infection of smallmouth bass. Further study will be necessary to determine whether disease dynamics are a consequence of effects on the host, alterations of instream habitat mediating invertebrate host dynamics and/or survival and dispersal of the parasite infective stage.
Understanding sedimentation patterns in small coastal watersheds due to landscape perturbations is critical for connecting hillslope and fluvial processes, in addition to managing aquatic habitats ...for anadromous fish and other aquatic species in the Oregon Coast Range (OCR). Changes in sedimentation patterns spanning the last 250 years are preserved in two landslide‐dammed lakes in small watersheds (< 10 km2) underlain by the Tyee Formation in the central OCR. Dendrochronology of drowned Douglas‐fir stumps in both lakes provided precise timing of the damming and formation of the lakes, with Klickitat Lake forming in winter ad 1751/52 and Wasson Lake in winter ad 1819/20. Perturbations from wildfires, logging and road development, and previously underappreciated snow events affect sedimentation rates in the lakes to different degrees, and are identified in the sediment record using cesium‐137 (137Cs), high‐resolution charcoal stratigraphy, local fire records, and aerial photography. Each lake has variable sedimentation accumulation rates (0.05–4.4 cm yr−1) and mass accumulation rates (0.02–1.42 g cm−2 yr−1). Sedimentation rates remained low from the landslide‐damming events until the mid‐19th century, when they increased following stand‐replacing wildfires. Aside from a sediment remobilization triggered by human modification of the landslide dam at Klickitat Lake around 1960, the largest peaks in mass accumulation rates in the mid‐20th century at both lakes in the early 1950s precede major road construction and logging activity in the watersheds. Subsequent sedimentation rates are lower, but variable, and possible effects of logging and road development might be exacerbated by abnormal precipitation and heavy snow events. A comparison of previous studies of landslide‐dammed lakes in larger watershed of the OCR are consistent with our findings of increased sedimentation in the mid‐20th century, as well as higher sedimentation rates in the debris‐flow dominated southern Tyee Formation than in the lower‐relief northern Tyee Formation.
Sedimentation patterns of small landslide‐dammed lakes in the Oregon Coast Range are quantified using dendrochronology of drowned trees, cesium‐137 (137Cs), and high‐resolution charcoal stratigraphy, to reveal sustained changes in sedimentation rates following 19th‐century wildfires, close‐proximity anthropogenic disturbances, and abnormal weather events.
High-resolution macroscopic charcoal and pollen analyses were used to reconstruct the fire and vegetation history of the Willamette Valley for the last 1200
years. Presented in this paper are three ...new paleoecological reconstructions from Lake Oswego, Porter Lake, and Warner Lake, Oregon, and portions of previous reconstructions from Battle Ground Lake, Washington, and Beaver Lake, Oregon. The reconstructions show that prior to Euro-American settlement vegetation and fire regimes were influenced by a combination of natural and anthropogenic factors. Battle Ground Lake shows a stronger influence from climate, while Lake Oswego, Beaver Lake, Porter Lake, and Warner Lake were more controlled by human activity. However, human-set fires were also modulated by regional climate variability during the Medieval Climate Anomaly and the Little Ice Age. Fire reconstructions from Battle Ground Lake, Lake Oswego, Beaver Lake, and Porter Lake imply that fires were infrequent in the Willamette Valley 200–300
years prior to Euro-American settlement. The decline of Native American populations due to introduced disease may have led to this reduction in fire activity. The prehistoric record from Warner Lake, however, indicates that fires in the foothills of the Cascade Range were more frequent than on the valley floor, at least until ca. AD 1800. The historic portions of the reconstructions indicate that Euro-American land clearance for agriculture and logging produced the most dramatic shifts in vegetation and fire regimes. All five records indicate that few fires in the Willamette Valley have occurred since ca. AD 1930, and fires today are predominantly grass fires.
