CYP enzyme induction is a sensitive biomarker for phenotypic metabolic competence of in vitro test systems; it is a key event associated with thyroid disruption, and a biomarker for toxicologically ...relevant nuclear receptor-mediated pathways. This paper summarises the results of a multi-laboratory validation study of two in vitro methods that assess the potential of chemicals to induce cytochrome P450 (CYP) enzyme activity, in particular CYP1A2, CYP2B6, and CYP3A4. The methods are based on the use of cryopreserved primary human hepatocytes (PHH) and human HepaRG cells.
The validation study was coordinated by the European Union Reference Laboratory for Alternatives to Animal Testing of the European Commission's Joint Research Centre and involved a ring trial among six laboratories. The reproducibility was assessed within and between laboratories using a validation set of 13 selected chemicals (known human inducers and non-inducers) tested under blind conditions. The ability of the two methods to predict human CYP induction potential was assessed. Chemical space analysis confirmed that the selected chemicals are broadly representative of a diverse range of chemicals.
The two methods were found to be reliable and relevant in vitro tools for the assessment of human CYP induction, with the HepaRG method being better suited for routine testing. Recommendations for the practical application of the two methods are proposed.

Background: In pregnant women, changes in body posture and higher pelvic anteversion and increase lumbar lordosis are observed. In this study, we present applications of tensegrity massage combined with point manipulation of selected muscle attachments.
Aim: The aim of the study was to determine the effectiveness of tensegrity massage combined with point manipulation of selected muscle attachments in pregnant women.
Participants and intervention: Two 30-year-old pregnant women received tensegrity massage. The patients were subjected to a series of nine massage sessions of 45 minutes each, performed three times a week.
Research Design: A case study
Main Outcomes Measures: Pain levels were assessed in both patients using the VAS
Results: The procedure involved normalization of the tone of the system of ligaments, fascia, and muscles in the lumbar spine region, the pelvic girdle, and the lower limbs. The therapy brought pain relief and reduced muscle tone in the lumbosacral region of the spine, as well as in the pelvic girdle and the lower limbs.
Conclusion: Tensegrity massage combined with point massage is an effective therapy for pain problems and incorrect resting muscle tone caused by pregnancy-related posture changes within the pelvis and spine.

This study examined the effects of a 5-min massage session on the muscle stiffness and tone reduction in healthy young adults.
Participants aged between 19 and 24 were randomly assigned to an intervention group (IG) (n = 40) or a control group (CG) (n = 40). The IG participated in a 5-min massage session of the lateral and medial gastrocnemius muscle (LGCM and MGCM respectively). The muscle contractile properties were assessed by tensiomyography (TMG). It measured maximal displacement (Dm), delay time (Td) and contraction time (Tc). Oscillation frequency (F) and stiffness (S) were used to assess muscle passive mechanical properties. These measurements were collected by MytonPro.
There were no differences between the groups in the pre-test measurements. The results of mixed-model ANOVA showed significant (p < 0.05) time × group interactions, with greater alteration in the IG compared to the CG in the LGCM for Dm (%diff = 22.5, ƞp2 = 0.41), Td (%diff = 3.63, ƞp2 = 0.02), Tc (%diff = 13.38, ƞp2 = 0.1), F (%diff = 5.55, ƞp2 = 0.07) and in the MGCM for Dm (%diff = −14.34, ƞp2 = 0.06), F (%diff = −7.53, ƞp2 = 0.12). No difference (p > 0.05) was noticed in other variables.
A 5-min massage session impacts the contractile and passive mechanical properties of the LGCM and MGCM. The size and direction of changes may depend on the muscle type, its geometry as well as other factors.

Abstract Objective The purpose of this study was to compare the clinical outcomes of classic massage to massage based on the tensegrity principle for patients with chronic idiopathic shoulder pain. ...Methods Thirty subjects with chronic shoulder pain symptoms were divided into 2 groups, 15 subjects received classic (Swedish) massage to tissues surrounding the glenohumeral joint and 15 subjects received the massage using techniques based on the tensegrity principle. The tensegrity principle is based on directing treatment to the painful area and the tissues (muscles, fascia, and ligaments) that structurally support the painful area, thus treating tissues that have direct and indirect influence on the motion segment. Both treatment groups received 10 sessions over 2 weeks, each session lasted 20 minutes. The McGill Pain Questionnaire and glenohumeral ranges of motion were measured immediately before the first massage session, on the day the therapy ended 2 weeks after therapy started, and 1 month after the last massage. Results Subjects receiving massage based on the tensegrity principle demonstrated statistically significance improvement in the passive and active ranges of flexion and abduction of the glenohumeral joint. Pain decreased in both massage groups. Conclusions This study showed increases in passive and active ranges of motion for flexion and abduction in patients who had massage based on the tensegrity principle. For pain outcomes, both classic and tensegrity massage groups demonstrated improvement.
