Xenotransplantation is a promising strategy to alleviate the shortage of organs for human transplantation. In addition to the concerns about pig-to-human immunological compatibility, the risk of ...cross-species transmission of porcine endogenous retroviruses (PERVs) has impeded the clinical application of this approach. We previously demonstrated the feasibility of inactivating PERV activity in an immortalized pig cell line. We now confirm that PERVs infect human cells, and we observe the horizontal transfer of PERVs among human cells. Using CRISPR-Cas9, we inactivated all of the PERVs in a porcine primary cell line and generated PERV-inactivated pigs via somatic cell nuclear transfer. Our study highlights the value of PERV inactivation to prevent cross-species viral transmission and demonstrates the successful production of PERV-inactivated animals to address the safety concern in clinical xenotransplantation.

The oxygen reduction reaction (ORR) is one of the most important reactions in life processes and energy conversion systems. To alleviate global warming and the energy crisis, the development of ...high‐performance electrocatalysts for the ORR for application in energy conversion and storage devices such as metal–air batteries and fuel cells is highly desirable. Inspired by the biological oxygen activation/reduction process associated with heme‐ and multicopper‐containing metalloenzymes, iron and copper‐based transition‐metal complexes have been extensively explored as ORR electrocatalysts. Herein, an outline into recent progress on non‐precious‐metal electrocatalysts for the ORR is provided; these electrocatalysts do not require pyrolysis treatment, which is regarded as desirable from the viewpoint of bioinspired molecular catalyst design, focusing on iron/cobalt macrocycles (porphyrins, phthalocyanines, and corroles) and copper complexes in which the ORR activity is tuned by ligand variation/substitution, the method of catalyst immobilization, and the underlying supporting materials. Current challenges and exciting imminent developments in bioinspired ORR electrocatalysts are summarized and proposed.

Cushing's disease results from corticotroph adenomas of the pituitary that hypersecrete adrenocorticotropin (ACTH), leading to excess glucocorticoid and hypercortisolism. Mutations of the ...deubiquitinase gene USP8 occur in 35-62% of corticotroph adenomas. However, the major driver mutations in USP8 wild-type tumors remain elusive. Here, we report recurrent mutations in the deubiquitinase gene USP48 (predominantly encoding p.M415I or p.M415V; 21/91 subjects) and BRAF (encoding p.V600E; 15/91 subjects) in corticotroph adenomas with wild-type USP8. Similar to USP8 mutants, both USP48 and BRAF mutants enhance the promoter activity and transcription of the gene encoding proopiomelanocortin (POMC), which is the precursor of ACTH, providing a potential mechanism for ACTH overproduction in corticotroph adenomas. Moreover, primary corticotroph tumor cells harboring BRAF V600E are sensitive to the BRAF inhibitor vemurafenib. Our study thus contributes to the understanding of the molecular mechanism of the pathogenesis of corticotroph adenoma and informs therapeutic targets for this disease.
A nitrogen-phosphorus dual-doped porous spore carbon (NP-PSC) positive electrode matrix was prepared using native auricularia auricula as solid medium based on the principle of biomass rot. The freeze-drying and carbonization activation processes made the materials present a three-dimensional porous spore carbon aerogel properties. Yeast fermentation transformed auricularia auricula from blocky structure to porous structure and introduced nitrogen-phosphorus dual-doping. The physical and chemical properties of the prepared materials were characterized in detail. Electrochemical performance of NP-PSC in Li-S batteries was systematically investigated. Porous structure and heteroatom-doping improved the electrochemical performance, which is much superior to conventional activated carbon materials.
Many cities and countries are now striving to create intelligent transportation systems that utilize the current abundance of multisource and multiform data related to the functionality and the use ...of transportation infrastructure to better support human mobility, interests, and lifestyles. Such intelligent transportation systems aim to provide novel services that can enable transportation consumers and managers to be better informed and make safer and more efficient use of the infrastructure. However, the transportation domain is characterized by both complex data and complex problems, which calls for visual analytics approaches. The science of visual analytics is continuing to develop principles, methods, and tools to enable synergistic work between humans and computers through interactive visual interfaces. Such interfaces support the unique capabilities of humans (such as the flexible application of prior knowledge and experiences, creative thinking, and insight) and couple these abilities with machines' computational strengths, enabling the generation of new knowledge from large and complex data. In this paper, we describe recent developments in visual analytics that are related to the study of movement and transportation systems and discuss how visual analytics can enable and improve the intelligent transportation systems of the future. We provide a survey of literature from the visual analytics domain and organize the survey with respect to the different types of transportation data, movement and its relationship to infrastructure and behavior, and modeling and planning. We conclude with lessons learned and future directions, including social transportation, recommender systems, and policy implications.
