Merging neutron stars offer an excellent laboratory for simultaneously studying strong-field gravity and matter in extreme environments. We establish the physical association of an electromagnetic ...counterpart (EM170817) with gravitational waves (GW170817) detected from merging neutron stars. By synthesizing a panchromatic data set, we demonstrate that merging neutron stars are a long-sought production site forging heavy elements by r-process nucleosynthesis. The weak gamma rays seen in EM170817 are dissimilar to classical short gamma-ray bursts with ultrarelativistic jets. Instead, we suggest that breakout of a wide-angle, mildly relativistic cocoon engulfing the jet explains the low-luminosity gamma rays, the high-luminosity ultraviolet-optical-infrared, and the delayed radio and x-ray emission. We posit that all neutron star mergers may lead to a wide-angle cocoon breakout, sometimes accompanied by a successful jet and sometimes by a choked jet.
We create squeezed light by exploiting the quantum nature of the mechanical interaction between laser light and a membrane mechanical resonator embedded in an optical cavity. The radiation-pressure ...shot noise (fluctuating optical force from quantum laser amplitude noise) induces resonator motion well above that of thermally driven motion. This motion imprints a phase shift on the laser light, hence correlating the amplitude and phase noise, a consequence of which is optical squeezing. We experimentally demonstrate strong and continuous optomechanical squeezing of 1.7±0.2dB below the shot-noise level. The peak level of squeezing measured near the mechanical resonance is well described by a model whose parameters are independently calibrated and that includes thermal motion of the membrane with no other classical noise sources.
ABSTRACT We apply a number of statistical and machine learning techniques to classify and rank gamma-ray sources from the Third Fermi Large Area Telescope Source Catalog (3FGL), according to their ...likelihood of falling into the two major classes of gamma-ray emitters: pulsars (PSR) or active galactic nuclei (AGNs). Using 1904 3FGL sources that have been identified/associated with AGNs (1738) and PSR (166), we train (using 70% of our sample) and test (using 30%) our algorithms and find that the best overall accuracy (>96%) is obtained with the Random Forest (RF) technique, while using a logistic regression (LR) algorithm results in only marginally lower accuracy. We apply the same techniques on a subsample of 142 known gamma-ray pulsars to classify them into two major subcategories: young (YNG) and millisecond pulsars (MSP). Once more, the RF algorithm has the best overall accuracy (∼90%), while a boosted LR analysis comes a close second. We apply our two best models (RF and LR) to the entire 3FGL catalog, providing predictions on the likely nature of unassociated sources, including the likely type of pulsar (YNG or MSP). We also use our predictions to shed light on the possible nature of some gamma-ray sources with known associations (e.g., binaries, supernova remnants/pulsar wind nebulae). Finally, we provide a list of plausible X-ray counterparts for some pulsar candidates, obtained using Swift, Chandra, and XMM. The results of our study will be of interest both for in-depth follow-up searches (e.g., pulsar) at various wavelengths and for broader population studies.
We recently derived mouse expanded potential stem cells (EPSCs) from individual blastomeres by inhibiting the critical molecular pathways that predispose their differentiation. EPSCs had enriched ...molecular signatures of blastomeres and possessed developmental potency for all embryonic and extra-embryonic cell lineages. Here, we report the derivation of porcine EPSCs, which express key pluripotency genes, are genetically stable, permit genome editing, differentiate to derivatives of the three germ layers in chimeras and produce primordial germ cell-like cells in vitro. Under similar conditions, human embryonic stem cells and induced pluripotent stem cells can be converted, or somatic cells directly reprogrammed, to EPSCs that display the molecular and functional attributes reminiscent of porcine EPSCs. Importantly, trophoblast stem-cell-like cells can be generated from both human and porcine EPSCs. Our pathway-inhibition paradigm thus opens an avenue for generating mammalian pluripotent stem cells, and EPSCs present a unique cellular platform for translational research in biotechnology and regenerative medicine.
Soluble aggregates of the microtubule-associated protein tau have been challenging to assemble and characterize, despite their important role in the development of tauopathies. We found that ...sequential hyperphosphorylation by protein kinase A in conjugation with either glycogen synthase kinase 3β or stress activated protein kinase 4 enabled recombinant wild-type tau of isoform 0N4R to spontaneously polymerize into small amorphous aggregates in vitro. We employed tandem mass spectrometry to determine the phosphorylation sites, high-resolution native mass spectrometry to measure the degree of phosphorylation, and super-resolution microscopy and electron microscopy to characterize the morphology of aggregates formed. Functionally, compared with the unmodified aggregates, which require heparin induction to assemble, these self-assembled hyperphosphorylated tau aggregates more efficiently disrupt membrane bilayers and induce Toll-like receptor 4-dependent responses in human macrophages. Together, our results demonstrate that hyperphosphorylated tau aggregates are potentially damaging to cells, suggesting a mechanism for how hyperphosphorylation could drive neuroinflammation in tauopathies.
Control over ferroelectric polarization variants in BiFeO3 films through the use of various vicinal SrTiO3 substrates is demonstrated. The revolution of domain formation as a function of vicinality ...is characterized, and the ferroelectric polarization variants in these films and the corresponding structural variants are carefully analyzed. The piezo/ferroelectric properties of the BiFeO3 films, in turn, can be tailored through this approach.
Aim
The current study aimed to visualize the damage caused by enterolysin A to the cells of sensitive strains and to find out cleavage site within the peptidoglycan moiety of bacterial cell walls.
...Methods and Results
Enterolysin A produced by a local isolate, Enterococcus faecalis B9510 was found to rapidly kill cells of the sensitive strain Lactococcus lactis ssp. cremoris 2144 during 120 min of treatment as compared to the untreated control where no such effect was observed. Transmission electron microscopy of the enterolysin A‐treated cells revealed leaking of the cytoplasmic contents ultimately resulting in complete lysis of cell walls. To find the cleavage site, purified cell walls of L. lactis ssp. cremoris 2144, Pediococcus pentosaceus 43201 and Lactobacillus delbrueckii ssp. bulgaricus ATCC 11842 were treated with enterolysin A, and liberated amino acids were derivatized for N and C terminals and analysed using thin layer chromatography on silica gel with isopropanol as solvent. The results showed that enterolysin A cleaves the peptide bonds at two locations within peptidoglycan subunits. The first location is between L‐alanine and D‐glutamic acid of the stem peptide and the other location is between L‐lysine of the stem peptide and D‐aspartic acid of the interpeptide bridge.
Conclusions
Enterolysin A cleaves the peptide bonds within the stem peptide as well as in the interpeptide bridge of Gram‐positive bacterial cell walls. This gives a possible reason for the broad spectrum of enterolysin A activity.
Significance and Impact of the Study
This is the first report identifying the cleavage site of enterolysin A within the cell walls of sensitive bacteria. This will help in identifying potential applications for enterolysin A.
