To explore the origins and consequences of tetraploidy in the African clawed frog, we sequenced the Xenopus laevis genome and compared it to the related diploid X. tropicalis genome. We characterize ...the allotetraploid origin of X. laevis by partitioning its genome into two homoeologous subgenomes, marked by distinct families of 'fossil' transposable elements. On the basis of the activity of these elements and the age of hundreds of unitary pseudogenes, we estimate that the two diploid progenitor species diverged around 34 million years ago (Ma) and combined to form an allotetraploid around 17-18 Ma. More than 56% of all genes were retained in two homoeologous copies. Protein function, gene expression, and the amount of conserved flanking sequence all correlate with retention rates. The subgenomes have evolved asymmetrically, with one chromosome set more often preserving the ancestral state and the other experiencing more gene loss, deletion, rearrangement, and reduced gene expression.
Studies suggest that exercise may be neuroprotective when implemented before the clinical presentation of Parkinson's disease (PD). Levels of brain-derived neurotrophic factor (BDNF), theorized to ...play a role in neuroprotection, are affected by its genotype and exercise. Here we explore this previously unstudied interaction on age at diagnosis and severity of symptoms.
76 participants with PD submitted buccal cells to determine BDNF genotype, completed the modified Lifetime Physical Activity Questionnaire to determine exercise habits, and were assessed using the Movement Disorder Society – Unified Parkinson's Disease Rating Scale III (MDS-UPDRS-III) and the Mini-Balance Evaluations Test (MBT). For aim 1 (age at diagnosis), 60 participants (age = 69.6 ± 7.4; males = 45, females = 15) were analyzed. For aim 2 (severity of symptoms), 54 participants (age = 70.0 ± 7.6; males = 41, females = 13) were analyzed.
The final hierarchical regression model for age at diagnosis produced an R2 = 0.146, p = .033; however, the only significant variable in the final model was average moderate physical activity from ages 20s to 40s (p = .009). The regression for MDS-UPDRS III was not significant; however, the regression for MBT was, p = .0499. In the final model, 23.1% of the variance was explained. Years since diagnosis (p = .014) and average vigorous physical activity from ages 20s to 40s (p = .047) were the only predictors in the final model.
While a strong interaction between BDNF genotype and lifetime physical activity was not observed, our results suggest that lifetime exercise may be neuroprotective in PD. Specifically, higher amounts of moderate PA were associated with an older age at diagnosis.
•This is the first study to show that pre-diagnosis exercise is associated with age at diagnosis.•Higher amounts of moderate physical activity were associated with an older age at diagnosis.•Every hour increase in weekly moderate physical activity from ages 20–40 was associated with a PD diagnosis 3 months later.
Abstract SNAPSHOT USA is a multicontributor, long‐term camera trap survey designed to survey mammals across the United States. Participants are recruited through community networks and directly ...through a website application ( https://www.snapshot-usa.org/ ). The growing Snapshot dataset is useful, for example, for tracking wildlife population responses to land use, land cover, and climate changes across spatial and temporal scales. Here we present the SNAPSHOT USA 2021 dataset, the third national camera trap survey across the US. Data were collected across 109 camera trap arrays and included 1711 camera sites. The total effort equaled 71,519 camera trap nights and resulted in 172,507 sequences of animal observations. Sampling effort varied among camera trap arrays, with a minimum of 126 camera trap nights, a maximum of 3355 nights, a median 546 nights, and a mean 656 ± 431 nights. This third dataset comprises 51 camera trap arrays that were surveyed during 2019, 2020, and 2021, along with 71 camera trap arrays that were surveyed in 2020 and 2021. All raw data and accompanying metadata are stored on Wildlife Insights ( https://www.wildlifeinsights.org/ ), and are publicly available upon acceptance of the data papers. SNAPSHOT USA aims to sample multiple ecoregions in the United States with adequate representation of each ecoregion according to its relative size. Currently, the relative density of camera trap arrays varies by an order of magnitude for the various ecoregions (0.22–5.9 arrays per 100,000 km 2 ), emphasizing the need to increase sampling effort by further recruiting and retaining contributors. Rover to acquire and cache rock core and regolith samples, prepare abraded rock surfaces, and support proximity science instruments.
The SCS consists of the Robotic Arm (RA), the Turret and Corer, and the Adaptive Caching Assembly (ACA). These elements reside and interact both inside and outside of the
Perseverance
Rover to enable surface interactions, sample transfer, and caching. The main body of the Turret consists of the Coring Drill (Corer) with a Launch Abrading Bit initially installed prior to launch. Mounted to the Turret main structure are two proximity science instruments, SHERLOC and PIXL, as well as the Gas Dust Removal Tool (gDRT) and the Facility Contact Sensor (FCS). These work together with the RA to provide the sample acquisition, abraded surface preparation, and proximity science functions. The ACA is a network of assemblies largely inside the front belly of the Rover, which combine to perform the sample handling and caching functions of the mission. The ACA primarily consists of the Bit Carousel, the Sample Handling Assembly (SHA), End Effector (EE), Sample Tubes and their Sample Tube Storage Assembly (STSA), Seals and their Dispenser, Volume, and Tube Assembly (DVT), the Sealing Station, the Vision Station, the Cover Parking Lot, and additional supporting hardware. These components attach to the Caching Component Mounting Deck (CCMD) that is integrated with the Rover interior. This work describes these major elements of the SCS, with an emphasis on the functionality required to perform the set of tasks and interactions required by the subsystem. Key considerations of contamination control and biological cleanliness throughout the development of these hardware elements are also discussed.
Additionally, aspects of testing and validating the functionality of the SCS are described. Early prototypes and tests matured the designs over several years and eventually led to the flight hardware and integrated testing in both Earth ambient and Mars-like environments. Multiple unique testbed venues were developed and used to enable testing from low-level mechanism operation through end-to-end sampling and caching interactions with the full subsystem and flight software. Various accomplishments from these testing efforts are highlighted. These past and ongoing tests support the successful preparations of the SCS on its pathway to operations on Mars.
In response to the critical shortage of Doxil(®), the US Food and Drug Administration (FDA) allowed temporary importation of non-FDA-approved second-generation liposomal doxorubicin, Lipo-Dox(®). ...Lipo-Dox utilizes a different liposomal particle than Doxil and demonstrates different pharmacokinetic properties. Its use has never been evaluated in a North American population. The objective of this study was to evaluate the efficacy and tolerability of Lipo-Dox at Magee-Womens Hospital, University of Pittsburgh Medical Center, for patients with recurrent epithelial ovarian cancer who were treated during the Doxil shortage.
Patients treated with Lipo-Dox from January 2012 to December 2012 were identified retrospectively. Disease response was defined radiographically by RECIST (Response Evaluation Criteria in Solid Tumors) or biochemically by CA-125 level if measurable disease was not present. Survival was defined from the start date of Lipo-Dox until the date of progression or death. Toxicity was assessed by the Gynecologic Oncology Group common toxicity criteria.
Eighteen patients with recurrent epithelial ovarian cancer who received Lipo-Dox were identified. These patients had a median of three prior treatment regimens. The median number of Lipo-Dox cycles given was 3.5 (range 1-8). No patients had a complete or partial response. Two patients had stable disease over a mean follow-up of 144.5 days. Fourteen patients had progressive disease, with a median time to progression of 82 days. Progression was based on CA-125 in four patients and RECIST in the remainder. Nine patients died from the disease.
Although this represents a small, pretreated population, there were no clinical responses to Lipo-Dox, raising the question as to whether it is an equivalent substitute for Doxil. Further evaluation is needed, but if confirmed, these findings raise concerns regarding the use of current stocks of Lipo-Dox, as well as the prudence of managing future drug shortages with pharmacologically similar, but clinically untested drugs.

Onco Targets Ther. 2014;7:1409-1413.It has been brought to our attention that there are two pharmaceutical companies that market drugs with the name 'lipodox' and ...we erroneously refer to the product used in our current study-Outcomes analysis of an alternative formulation of PEGylated liposomal doxorubicin in recurrent epithelial ovarian carcinoma during the drug shortage era. by the trade name Lipo-dox®, a drug produced by TTY Biopharm Co. Ltd, when the Sun Pharma product is Lipodox. Lipo-Dox® utilizes a different liposomal particle than Doxil®, DSCP versus HSCP. Due to the similarity of the drug names, we were under the mistaken impression that Sun Pharma's Lipodox utilized the DSCP liposome. We postulated that this might be related to the poor clinical outcomes seen in our population. It is now clear that Lipodox utilizes the same liposome as Doxil® and this is not a relevant point of discussion. The authors would like to let the readers know that Lipodox uses HSCP as the liposome and this should be corrected in the manuscript. The patients in our current study were all treated with Lipodox, and this drug uses HSCP as the liposome.
Recently, it has been shown that Charcot-Marie-Tooth disease type 1a (CMT1a) is linked with a duplication of a DNA segment that is detected by probe VAW409R3, and that is located on chromosome ...17p11.2. Here, we show that this duplication also contains VAW412R3a, but not A10-41 and EW503. Accounting for the duplication in recombination analysis, we found recombinants between CMT1a and EW301 and EW502, but not with A10-41, VAW409R3, and VAW412R3. Using pulsed-field gel electrophoresis analysis, we estimated the minimal size of the duplicated region in CMT1a patients to be 1100 kb.
