The argument that human society can decouple economic growth-defined as growth in Gross Domestic Product (GDP)-from growth in environmental impacts is appealing. If such decoupling is possible, it ...means that GDP growth is a sustainable societal goal. Here we show that the decoupling concept can be interpreted using an easily understood model of economic growth and environmental impact. The simple model is compared to historical data and modelled projections to demonstrate that growth in GDP ultimately cannot be decoupled from growth in material and energy use. It is therefore misleading to develop growth-oriented policy around the expectation that decoupling is possible. We also note that GDP is increasingly seen as a poor proxy for societal wellbeing. GDP growth is therefore a questionable societal goal. Society can sustainably improve wellbeing, including the wellbeing of its natural assets, but only by discarding GDP growth as the goal in favor of more comprehensive measures of societal wellbeing.
This article traces Jonathan Swift’s legacy in the work of three modern Irish poets: Jessica Traynor, Rita Ann Higgins and Derek Mahon. I use two motifs to explore this inheritance: institutions and ...shadows. Using the work of Genevieve Lloyd, I argue that meaningful engagement with Enlightenment’s legacy must recognise its moral ambiguity and emotional complexity through the metaphor of shadow. Institutions, whether in the case of St Patrick’s Hospital, Dublin (Ireland’s first psychiatric hospital, founded by Swift), the residential institutions of the twentieth century or the modern apparatus of Direct Provision, embody a continuing legacy of suffering, control and containment. Imagination and empathy are the difficult but necessary means through which these legacies must be acknowledged and confronted.
The United Nations Sustainable Development Goals (SDGs) comprise an important policy achievement towards identifying shared goals for addressing global challenges such as climate change and ...biodiversity loss. Though policy coherence for sustainable development ultimately depends on coherence among the SDGs – as progress toward one goal should not hamper progress toward others – synergies and trade-offs are emerging among the metrics (indicators) used to measure progress towards the various targets of the SDGs. In this context, the choice of Gross Domestic Product (GDP) per capita as an indicator for SDG 8 (“Decent Work and Economic Growth”) contradicts the evidence that limitless economic growth is not possible on a planet with finite resources. Here, we highlight how pursuing unconditional GDP growth risks failing to achieve the SDGs overall. We show how, in the European Union, GDP is unrelated to other measures of economic performance such as levels of employment, and relates inversely to indicators of environmental sustainability and broader measures of wellbeing. Pursuing SDG 8 through a continuous increase in GDP will therefore hinder the achievement of environmental goals and goals on reducing inequalities. We propose guidelines for selecting alternative indicators for SDG 8 with the aim of improving coherence among all of the SDGs, as well as between the SDGs and other policy initiatives for sustainability. This will inform a better monitoring of SDG 8 and the definition of post-2030 Agendas for sustainable development.
The COVID-19 pandemic has made well-fitting face masks a critical piece of protective equipment for healthcare workers and civilians. While the importance of wearing face masks has been acknowledged, ...there remains a lack of understanding about the role of good fit in rendering protective equipment useful. In addition, supply chain constraints have caused some organizations to abandon traditional quantitative or/and qualitative fit testing, and instead, have implemented subjective fit checking. Our study seeks to quantitatively evaluate the level of fit offered by various types of masks, and most importantly, assess the accuracy of implementing fit checks by comparing fit check results to quantitative fit testing results.
Seven participants first evaluated N95 and KN95 respirators by performing a fit check. Participants then underwent quantitative fit testing wearing five N95 respirators, a KN95 respirator, a surgical mask, and fabric masks.
N95 respirators offered higher degrees of protection than the other categories of masks tested; however, it should be noted that most N95 respirators failed to fit the participants adequately. Fit check responses had poor correlation with quantitative fit factor scores. KN95, surgical, and fabric masks achieved low fit factor scores, with little protective difference recorded between respiratory protection options. In addition, small facial differences were observed to have a significant impact on quantitative fit.
Fit is critical to the level of protection offered by respirators. For an N95 respirator to provide the promised protection, it must fit the participant. Performing a fit check via NHS self-assessment guidelines was an unreliable way of determining fit.
Venetoclax is a highly selective, potent, oral BCL-2 inhibitor, which induces apoptosis in multiple myeloma cells. Venetoclax plus bortezomib and dexamethasone has shown encouraging clinical efficacy ...with acceptable safety and tolerability in a phase 1 trial. The aim of this study was to evaluate venetoclax plus bortezomib and dexamethasone in patients with relapsed or refractory multiple myeloma.
In this randomised, double-blind, multicentre, phase 3 trial, patients aged 18 years or older with relapsed or refractory multiple myeloma, an Eastern Cooperative Oncology Group performance status of 2 or less, who had received one to three previous therapies were enrolled from 90 hospitals in 16 countries. Eligible patients were randomly assigned (2:1) centrally using an interactive response technology system and a block size of three to receive venetoclax (800 mg per day orally) or placebo with bortezomib (1·3 mg/m2 subcutaneously or intravenously and dexamethasone (20 mg orally). Treatment was given in 21-day cycles for the first eight cycles and 35-day cycles from the ninth cycle until disease progression, unacceptable toxicity, or patient withdrawal. Randomisation was stratified by previous exposure to a proteasome inhibitor and the number of previous therapies. Sponsors, investigators, study site personnel, and patients were masked to the treatment allocation throughout the study. The primary endpoint was independent review committee-assessed progression-free survival in the intention-to-treat population. Safety analyses were done in patients who received at least one dose of study drug. This study is registered with ClinicalTrials.gov, NCT02755597.
Between July 19, 2016, and Oct 31, 2017, 291 patients were randomly assigned to receive venetoclax (n=194) or placebo (n=97). With a median follow-up of 18·7 months (IQR 16·6–21·0), median progression-free survival according to independent review committee was 22·4 months (95% CI 15·3–not estimable) with venetoclax versus 11·5 months (9·6–15·0) with placebo (hazard ratio HR 0·63 95% CI 0·44–0·90; p=0·010). The most common grade 3 or worse treatment-emergent adverse events were neutropenia (35 18% of 193 patients in the venetoclax group vs seven 7% of 96 patients in the placebo group), pneumonia (30 16% vs nine 9%), thrombocytopenia (28 15% vs 29 30%), anaemia (28 15% vs 14 15%), and diarrhoea (28 15% vs 11 11%). Serious treatment-emergent adverse events occurred in 93 (48%) patients in the venetoclax group and 48 (50%) patients in the placebo group, with eight (4%) treatment-emergent fatal infections reported in the venetoclax group and none reported in the placebo group. Three deaths in the venetoclax group (two from pneumonia and one from septic shock) were considered treatment-related; no deaths in the placebo group were treatment-related.
The primary endpoint was met with a significant improvement in independent review committee-assessed progression-free survival with venetoclax versus placebo plus bortezomib and dexamethasone. However, increased mortality was seen in the venetoclax group, mostly because of an increased rate of infections, highlighting the importance of appropriate selection of patients for this treatment option.

Protest‐framing ...theory reveals the interlocking rhetorical tactics (injustice, identity, and agency framing) consumers use to mobilize mass audiences against a firm, contributing important insights to our understanding of negative word of mouth. Moreover, an analysis of protest sites reveals that consumers “frame” their corporate betrayal to the public to demonstrate their power to influence others and gain revenge. As a result, a community of discontent may arise in which both individual and social identities appear to be constructed and affirmed.
As the dominant form of urban agriculture (UA) in Australia, existing home food gardens potentially represent a significant resource in the context of future urban food security and sustainability. ...However, a severe lack of in-field data has hindered our understanding of the form and function of home food gardens which in turn may hinder innovation and improvement. We investigated the productivity, resource efficiency and potential financial savings of home food gardens in South Australia. A group of 34 citizen science participants measured and recorded inputs and outputs from their gardens. Inputs included time spent on various gardening activities, financial costs, and water use. Outputs included crop yields, from which retail value and nutritional content were then derived. The paper outlines a field-demonstrated, comprehensive methodology for continued and consistent data collection for all forms of UA. We found smaller gardens to be more intensive than larger gardens, requiring higher inputs, but also returning higher outputs per unit area. Both productivity and resource efficiency varied among the gardens, and labour requirements were significantly lower than previously estimated. Water use efficiency of the gardens were calculated and found to have comparable water use efficiency to commercial horticulture. Of the gardens involved, we calculated that 65% should break even in five or less years and save money. After applying a minimum wage almost one in five gardens were financially viable. The results represent the most comprehensive measurements on home food gardens to date, and allow practical, evidence-based recommendations for diversification, time saving and smart irrigation practices to improve garden productivity and enhance the viability of UA.
The master transcription factors play integral roles in the pluripotency transcription circuitry of embryonic stem cells (ESCs). How they selectively activate expression of the pluripotency network ...while simultaneously repressing genes involved in differentiation is not fully understood. Here, we define a requirement for the INO80 complex, a SWI/SNF family chromatin remodeler, in ESC self-renewal, somatic cell reprogramming, and blastocyst development. We show that Ino80, the chromatin remodeling ATPase, co-occupies pluripotency gene promoters with the master transcription factors, and its occupancy is dependent on OCT4 and WDR5. At the pluripotency genes, Ino80 maintains an open chromatin architecture and licenses recruitment of Mediator and RNA polymerase II for gene activation. Our data reveal an essential role for INO80 in the expression of the pluripotency network and illustrate the coordination among chromatin remodeler, transcription factor, and histone-modifying enzyme in the regulation of the pluripotent state.

This article examines the striking parallel between plans that were discussed in the CIA in 2017 to kidnap and possibly murder Julian Assange, the founder of WikiLeaks, who was living in the ...Ecuadorian Embassy in London and the abduction and murder eighty years earlier in Paris of the head of the White Russian army in exile by the NKVD. The latter, one of the most notorious episodes in the history of espionage and counterespionage in the French capital during the interwar years, was fictionalized by Éric Rohmer in his 2004 film Triple Agent. Fusing the actual events of those years-the Popular Front government in France, the Stalinist Terror in Russia, and the build-up to the Nazi-Soviet Pact in 1939-with a tragic personal story, Rohmer exposed the human dramas without which the rival intelligence services could not have been as effective as they were. The events of the 1930s are long past, and even the Cold War years of the 1960s and 1970s have receded into the distance. But as this article shows, and Éric Rohmer's film reminds us, the cloak- and-dagger game of spies and counterspies has not changed that much, the technologies of contemporary intelligence warfare notwithstanding.
