ABSTRACT
We measure the black hole mass and investigate the accretion flow around the local (z = 0.0502) quasar PG 1119+120. Spectroscopic monitoring with Calar Alto provides H β lags and linewidths ...from which we estimate a black hole mass of log (M•/M⊙) = 7.0, uncertain by ∼0.4 dex. High cadence photometric monitoring over 2 yr with the Las Cumbres Observatory provides light curves in seven optical bands suitable for intensive continuum reverberation mapping. We identify variability on two time-scales. Slower variations on a 100-d time-scale exhibit excess flux and increased lag in the u′ band and are thus attributable to diffuse bound-free continuum emission from the broad-line region. Faster variations that we attribute to accretion disc reprocessing lack a u′-band excess and have flux and delay spectra consistent with either τ ∝ λ4/3, as expected for a temperature structure of T(R) ∝ R−3/4 for a thin accretion disc, or τ ∝ λ2 expected for a slim disc. Decomposing the flux into variable (disc) and constant (host galaxy) components, we find the disc SED to be flatter than expected with $f_{\nu } \sim \rm {const}$. Modelling the SED predicts an Eddington ratio of λEdd > 1, where the flat spectrum can be reproduced by a slim disc with little dust extinction or a thin disc that requires more dust extinction. While this accretion is super-Eddington, the geometry is still unclear; however, a slim disc is expected due to the high radiation pressure at these accretion rates, and is entirely consistent with our observations.
The purpose of our study was to investigate the safety, pharmacokinetics (PK), and initial antitumor efficacy of HC‐1119 in patients with metastatic castration‐resistant prostate cancer (mCRPC). ...Eligible mCRPC patients were included in our study (NCT 03774056) with two parts. Part A was a dose escalation study in which patients received a dose escalation of HC‐1119 (40, 80, 160 and 200 mg/day). Part B was a dose expansion study in which patients received HC‐1119 at the dose of 80 and 160 mg. Safety assessment and pharmacokinetic samplings were performed for all patients at the given time points; preliminary tumor response was also assessed. Twenty‐four patients were enrolled in part A and 19 patients in part B, respectively. HC‐1119 was safe, well tolerated and no dose‐limiting toxicity was observed. Fatigue was the most common treatment‐related adverse event and no seizures were observed. At the dose levels of 40, 80 and 160 mg, the AUC and Cmax of HC‐1119 in plasma increased almost dose‐proportionally at the steady state in mCRPC patients. Maximum prostate‐specific antigen (PSA) response rates (≥50% reduction from the baseline) in dose escalation and dose expansion cohorts were 77% and 75%, respectively; the overall disease control rate (22 patients available for imaging analysis) was 72.7%, with PR in 4 patients, SD in 12 patients and PD in 6 patients; the 2‐year overall survival rate in patients from Part B was 56.8%. HC‐1119 was safe, well tolerated and efficacious and HC‐1119 at 80 mg/day is recommended for further studies.

The influence of basin and depositional context on the formation and growth of fluid escape pipes in sedimentary basins is not fully understood. While seismic reflection data is commonly used to ...study these structures, direct observation and sampling are scarce. In this study, we investigate the evolution of hundreds of meter-tall fluid escape pipes (100–800 m) in the Canterbury Basin, New Zealand, using open source seismic and borehole data. Our analysis includes seismic interpretation of 19 unconformities within the Neogene-Quaternary successions, characterization of pipes and fluid-related anomalies in the subsurface, modeling of the shale volume (Vsh), lithology characterization, 3D static modeling, and seismic inversion. We identify thirty-one vertical to sub-vertical pipes with three parts consisting of top, main conduit/stem, and root zone on seismic profiles. Our results suggest that lithology plays a crucial role in the initiation and modulation of pipe structures in the Canterbury Basin, with pipes mostly formed post Tarantian (0.113 Ma) and truncated at the U19 unconformity. Furthermore, vertical fluid migration from overpressured units, such as the tops of contourite mounds, was observed in the pipes. Our findings provide new insights into the subsurface fluid flow and sedimentary architecture of the Canterbury Basin and have wider implications for other passive continental margins with similar characteristics.
•Pipes range in height from 110 m to 808 m and are characterized by a top, main conduit, and root zone.•Pipes traverse through a mixture of heterogeneous siliciclastic sediments dominated by mud.•The pipes are rooted in overpressured and multiple layers, including sediment drifts.
The formation of the Antarctic Circumpolar Current (ACC) is critical for the evolution of the global climate, but the timing of its onset is not well constrained. Here, we present new seismic ...evidence of widespread Late Eocene to Oligocene marine diagenetic chert in sedimentary drift deposits east of New Zealand indicating prolonged periods of blooms of siliceous microorganisms starting ~36 million years ago (Ma). These major blooms reflect the initiation of the arrival and upwelling of northern-sourced, nutrient-rich deep equatorial Pacific waters at the high latitudes of the South Pacific. We show that this change in circulation was linked to the initiation of a proto-ACC, which occurred ~6 Ma earlier than the currently estimated onset of the ACC at 30 Ma. We propose that the associated increased primary productivity and carbon burial facilitated atmospheric carbon dioxide reduction contributing to the expansion of Antarctic Ice Sheet at the Eocene-Oligocene Transition.
AIM： To appraise the effect of treatment for diabetic macular edema（DME） in proliferative stage with sufficient panrentinal photocoagulation（PRP） therapy and intravitreal injections（IV） Conbercept ...and posterior subtenon＇s triamcinolone acetonide（STTA） sequential therapy.METHODS： This prospective clinical randomized controlled trial of cross-over design was conducted in three phases. The participants included cases of DME in proliferative stage. They were divided into two groups and treated with PRP before enrollment. Group A were treated with IVConbercept 0.5 mg for one month in the 1^st phase. Group B were treated with STTA 40 mg（twice per two weeks）. The interventions were exchanged in the second phase（2mo） between the two groups. In the third phase（3-6mo） no other treatment was given. Best corrected visual acuity（BCVA）, central macular thickness（CMT） measured by OCT and complications were compared.RESULTS： After phase I： in Group A, BCVA improved from 0.201±0.17 to 0.37±0.24（F=5.88, P=0.004）. CMT changed from 449±155.10 to 304.1±84.70 μm（F=14.9, P〈0.01）. In Group B, BCVA changed from 0.195±0.19 to 0.26±0.20（F=0.76, P=0.41） while CMT changed from 463.82±152.92 to 366.00±115.40 μm（F=3.70, P〈0.03）. The improvement of BCVA was better in Group A（P〈0.05）. After phase II： in Group A, BCVA raised to 0.47±0.27（F=0.26, P〈0.01）, CMT reduced to 260.67±62.97 μm（F=-188.3, P〈0.01）; in Group B, BCVA raised to 0.51±0.26（F=0.31, P〈0.01）, CMT reduced to 261.93±50.15 μm（F=-201.9, P〈0.01）. But there were no difference between two groups（P〉0.05）. After phase III： in Group A, BCVA maintained 0.42±0.25（F=0.22, P=0.001）, CMT maintained 267.8±58.34 μm,（F=-0.27, P〈0.01）; in Group B, BCVA was 0.47±0.25（F=-0.27, P〈0.01）, CMT was 272.71±49.16 μm（F=-191.1, P〈0.01）. No serious complications happened in all phases.CONCLUSION： PRP＋Conbercept is better than PRP＋STTA in DME with proliferative stage but PRP＋Conbercept＋STTA sequential therapy may be a wiser choice for persistent effectiveness on anatomical as well as functional status.
To investigate the effect of microbead iridocorneal angle occlusion on intraocular pressure (IOP) diurnal fluctuation in rat eyes.
Male Dark Agouti (DA) rats, 8-10 week old, were each given a single ...intracameral injection of microbeads, followed by injection of dispersive viscoelastic solution. The right eye served as the experimental eye, while the left eye served as the control. IOP was measured twice daily postoperatively for 3wk and compared between groups. At the end of 3wk, the rats were sacrificed and the eyes were harvested for histological analysis and retinal ganglion cell (RGC) counting.
After microbead injection, experimental eyes had significantly higher dark time IOP than controls from the second week to the third week 2

=5) from the first week to the third week after injection. Comparatively, they were significantly lower in the control eyes, which were 4.69±0.323 mm Hg (
=5), 2.84±1.122 mm Hg (
=5) and 4.98±0.603 mm Hg (
=5) respectively. However, at the end of 3wk, the larger fluctuations in IOP in the experimental eyes was not associated with a significant loss of RGCs.
Microbead occlusion exacerbates diurnal IOP fluctuation in rats. This reported model may serve as a method of investigating the pathological effects of IOP fluctuation. A longer observation period, or repeated injections, may be needed to observe a significant change in RGC density.
The Subtropical Front (STF) is one of the major ocean fronts in the south-west Pacific. The STF migration has a distinct but varying impact on the temperature change throughout the region. To improve ...our understanding on the history of the STF migration and the relationship between STF migration and global climate change, we have generated a 450-kyr record of sea surface temperature (SST) change at IODP Site U1352 located in the Canterbury Basin off New Zealand. Planktonic foraminiferal assemblages from the site were used to derive SST estimates. Comparisons of the planktonic foraminiferal assemblage and SST record of IODP Site U1352 with those of ODP Site 1119, DSDP Site 594 and core MD06-2986 allow us to reconstruct the spatial change of the STF over the last 450 kyr by locating the positions of its Southland Front (SF) section, which runs nearly parallel to the South Island's shoreline. The percentage of Neogloboquadrina pachyderma in the sum of Nq. pachyderma and Nq. incompta was below 40% at Sites IODP U1352, ODP 1119 and DSDP 594 during Marine Isotope Stage (MIS) 11c, MIS 7a–c and MIS 5e. Moreover, the SSTs of the three sites during these periods were similar and close to that of the present-day. This indicates that the three sites were covered by a Subtropical Water–Subantarctic Water (STW–SAW) mixture during these interglacials. IODP Site U1352 recorded a similar scenario during MIS 7e. In comparison with previous studies, the higher resolution record of IODP Site U1352 showed the influence of an STW–SAW mixture was limited to MIS 11c, MIS 7e, MIS 7a–c and MIS 5e at this site. The SSTs of Sites IODP U1352, ODP 1119 and DSDP 594 were similar during MIS 11a–b and MIS 9a–d. At the same time, the temperature differences among the three sites and core MD06-2986 (west of New Zealand) during MIS 11a–b and MIS 9a–d were larger than during interglacials MIS 11c, MIS 7a–c, and MIS 5e. These indicate that the Subantarctic Water (SAW) influenced Sites IODP U1352, ODP 1119 and DSDP 594 during MIS 11a–b and MIS 9a–d. The ~3–4 °C warmer SSTs at Sites IODP U1352 and ODP 1119 than at DSDP Site 594 suggest that the SF is situated between DSDP Site 594 and Sites IODP U1352, ODP 1119 during MIS 7d and glacial periods of MIS 10, MIS 8, and MIS 6. Hence, unlike previous studies in the region, MIS 7d is identified as a glacial period.
•Foraminiferal assemblages of the upper continental slope in the Canterbury Bight are analysed.•450 kyr record of sea surface temperature at IODP Site U1352 is reconstructed.•The Southland Front's migration is controlled by both the bathymetric and the climate changes.•The signals of global climate change are amplified by SF's migration in the south-west Pacific.
Determining the relative influence of eustasy versus local sedimentary processes on strata formation is a fundamental challenge in the study of continental margin stratigraphy. In this paper, the ...relative contribution of these factors on continental margin evolution during the Middle to Late Pleistocene is evaluated using samples from Integrated Ocean Drilling Program Expedition 317. Core‐logging, biostratigraphy and quantitative X‐ray diffraction mineralogy are used to delineate continental shelf sedimentary systems. Major lithological unconformities bound stratigraphic sequences that contain recurring compositional patterns and that resemble other examples of Middle to Upper Pleistocene sequences. However, a preliminary chronology suggests that sequence boundary formation cannot be linked ‘one to one’ with eustatic cycles and therefore these sequences can contain multiple ca 100 ka eustatic cycles. Smaller amplitude, higher frequency transitions in sediment composition are interpreted as stratigraphic sequences driven by more rapid perturbations in the interplay of accommodation and sediment supply; their stratigraphy is variable in time and across the shelf, suggesting a strong influence of local sedimentary forcing in their formation. Changes in sediment composition after the Middle Pleistocene Transition indicate that sediment transfer from onshore sources in the glaciated Southern Alps to the middle‐shelf occurred over a single 100 ka glacio‐eustatic cycle, with an additional 100 ka lag before the mineralogical signal was preserved on the outer‐shelf. This phenomenon is coincident with rapid shelf progradation in this basin, suggesting a causal relation between across‐shelf sediment transport and margin progradation. This is one of very few studies that provide insights at the core scale into the processes driving continental margin evolution during the Middle to Late Pleistocene. This work shows that compositional changes in mud‐dominated successions can lead to a sequence stratigraphic interpretation and the identification of high‐frequency sequences, which may not be possible using a conventional stratigraphic approach.
