Polysaccharides were identified, which allow for colon targeting in human Inflammatory Bowel Disease (IBD) patients, as well as in rats and dogs (which are frequently used as animals in preclinical ...studies). The polysaccharides are degraded by colonic enzymes (secreted by bacteria), triggering the onset of drug release at the target site. It has to be pointed out that the microbiota in rats, dogs and humans substantially differ. Thus, the performance of this type of colon targeting system observed in animals might not be predictive for patients. The aim of this study was to limit this risk. Different polysaccharides were exposed to culture medium inoculated with fecal samples from IBD patients, healthy dogs and “IBD rats” (in which colonic inflammation was induced). Dynamic changes in the pH of the culture medium were used as an indicator for the proliferation of the bacteria and, thus, the potential of the polysaccharides to serve as their substrate. Fundamental differences were observed with respect to the extent of the pH variations as well as their species-dependency. The most promising polysaccharides were used to prepare polymeric film coatings surrounding 5-aminosaliciylic acid (5-ASA)-loaded starter cores. To limit premature polysaccharide dissolution/swelling in the upper gastro intestinal tract, ethylcellulose was also included in the film coatings. Drug release was monitored upon exposure to culture medium inoculated with fecal samples from IBD patients, healthy dogs and “IBD rats”. For reasons of comparison, also 5-ASA release in pure culture medium was measured. Most film coatings showed highly species-dependent drug release kinetics or limited colon targeting capacity. Interestingly, extracts from aloe vera and reishi (a mushroom) showed a promising potential for colon targeting in all species.

Wetlands are complex ecosystems, considered among the most productive environments in the world. They contain important biodiversity hotspots, provide the resources on which countless plant and ...animal species depend and perform important environmental and economic functions. Wetlands reduce flooding events, improve water quality and represent a valuable cultural and natural heritage. The European Directive (2000/60/EC) requires fostering an integrated approach to wetland management through collaborative governance processes. The Wetland Contract is a tool that has been developed and implemented in order to promote the sustainable governance of water systems. Among the Mediterranean countries, Italy and Croatia count on a rich variety of coastal wetlands that, together with the plants and animals that inhabit and pass through them, constitute an extremely valuable natural heritage. This paper presents and discusses the results of the Interreg Project CREW that, between 2018–2021, contributed to the drafting of seven new Wetland Contracts. Through a comparative reading, the aim is to understand, on the one hand, how the different Contracts have responded individually to a shared programme based on European requirements; on the other hand, the aim is also to bring similar experiences closer together to assess their overall potential on an international scale. The article shows how the constant exchange between CREW partners and the continuous updating on ongoing activities, methodologies and proposed solutions in the seven target areas facilitated the construction of a shared cross-border strategy for wetland governance. The comparison also highlights some principles considered fundamental by all partners (protection and enhancement; integrated governance; awareness-raising and learning) that have served the construction of a transnational Observatory, aimed at monitoring, sharing practices and able to guide the formation of future Wetland Contracts.
European Territorial Cooperation programmes (also known as Interreg) are designed to promote integration by encouraging organizations in multiple regions to work together on projects to address ...shared challenges. While some key aspects of these programmes have been studied (territorial scope, thematic areas, types of initiatives), research into participant organizations and their logic within partnerships (groups of organizations working together on projects) and the reasons for their participation in multiple projects, individually or in clusters, has been less thorough. This article presents a quantitative analysis of programmes under Interreg B and C, investigating the variables that influence organizations' involvement in partnerships and the logic behind repeated collaborations. The aim is to supplement existing research, complementing it and supporting providing a better understanding of this framework to programme planners, enabling them to make more informed decisions when aiming to maximize the impact of their programmes when considering partners and partnerships.
Despite decades of spending, Cohesion Policy appears unable to fully address growing national disparities and increasing 'roll-out' nationalism. In the present study we discuss regional effects of ...'fuzzy' policy concepts, such as EU's policy for Territorial Cohesion, in Central European borderlands from a stakeholder perspective. Identifying how key policy documents have framed the discussion of Territorial Cohesion, we furthermore demonstrate the differing ways regional stakeholders have interpreted this vision. By showing how multiple translations have produced dynamics that create a circular process, we conclude that this process leads towards non-comparable outcomes, increased misunderstanding, while fuelling EU scepticism.
This paper estimates the impacts of opening of borders and intensity of cross-border cooperation (CBC) on regional growth across European border regions. The difference-in-difference estimations show ...that the opening of national borders for free movement of people is associated with a 2.7% increase in regional gross value added (GVA) per capita for the border regions of Europe. Using an extension of this estimation based on European Commission-supported CBC projects data for the 2007-13 period (through the INTERREG programme), the results suggest that the doubling of the number of CBC project partners (per 100,000 population) is correlated with an increase of 2.3% in regional GVA per capita. These results confirm the positive economic benefits of European integration by means of CBC. For future research efforts, these findings also encourage the research community to investigate more deeply the channels through which CBC affects the socioeconomic development of border regions.

The Cartagena-La Unión mountain range was the focus of an intense mining activity between early XIX and late XX centuries. Most of Spanish national production of lead and zinc was extracted from its ...mines. During the ore concentration process, contaminated wastes containing heavy metal minerals, cyanides and sulfates were produced and deposited in earth dams. The Spanish National Institute of Geology and Mining had catalogued 75 earth dams in the councils of Cartagena and La Unión. These deposits pose a potential risk for the environment and nearby populations. Without suitable and precautionary measures, contaminated particles can be transported far away due to the wind action and runoff water, and may be incorporated to the food chain. This risk is increase due to the fact that it is a seismically active area, and breakage of these dams can lead to the dumping of thousands of tons of contaminated wastes. The SOIL TAKE CARE Project is an international project co financed by the European Regional Development Fund (ERDF) through the Interreg Sudoe Cooperation Programme. It aims to improve the management and rehabilitation of contaminated soils in South-Western Europe that includes Spain, Portugal and south of France. The University of Oviedo takes part of that Project by the instrumentation and monitoring of two of those earth dams. Among the work realized so far highlights the perforation of two boreholes and the installation of several sensors. It aims a double objective: to analyze the erosion and infiltration capacity of rainfall into the dams and to detect possible symptoms of slope instability. Although the investigation is still in course, preliminary results shows fast rainfall infiltration into the superficial soil layers, being discharge curves much more extended. This water retention capacity, coupled with the existence of impermeable layers into the dams, could lead to a complete saturation of superficial soil layers and trigger slope instability processes.
The use of territorial impact assessment procedures is gaining increasing relevance in the European Union policy evaluation processes. However, no concrete territorial impact assessment tools have ...been applied to evaluating EU cross-border programmes. In this light, this article provides a pioneering analysis on how to make use of territorial impact assessment procedures on cross-border programmes. More specifically, it assesses the main territorial impacts of the Inner Scandinavian INTERREG-A sub-programme, in the last 20years (1996–2016). It focuses on its impacts on reducing the barrier effect, in all its main dimensions, posed by the presence of the administrative border. The results indicate a quite positive impact of the analysed cross-border cooperation programme, in reducing the barrier effect in all its main dimensions. The obtained potential impact values for each analysed dimension indicate, however, that the ‘economy-technology’ dimension was particularly favoured, following its strategic intervention focus in stimulating the economic activity and the attractiveness of the border area.
•A territorial impact assessment method to assess cross-border cooperation is proposed.•This method rationale is based on the main dimensions of the barrier effect.•This method identified positive impacts in all analysed dimensions.•The economy-technological dimension was the most positively impacted one.
Fully automated at-line terahertz time-domain spectroscopy in transmission mode is used to measure tablet porosity for thousands of immediate release tablets. The measurements are rapid and ...non-destructive. Both laboratory prepared tablets and commercial samples are studied. Multiple measurements on individual tablets quantify the random errors in the terahertz results. These show that the measurements of refractive index are precise, with the standard deviation on a single tablet being about 0.002, with variation between measurements being due to small errors in thickness measurement and from the resolution of the instrument. Six batches of 1000 tablets each were directly compressed using a rotary press. The tabletting turret speed (10 and 30 rpm) and compaction pressure (50, 100 and 200 MPa) were varied between the batches. As expected, the tablets compacted at the highest pressure have far lower porosity than those compacted at the lowest pressure. The turret rotation speed also has a significant effect on porosity. This variation in process parameters resulted in batches of tablets with an average porosity between 5.5 and 26.5%. Within each batch, there is a distribution of porosity values, the standard deviation of which is in the range 1.1 to 1.9%. Destructive measurements of disintegration time were performed in order to develop a predictive model correlating disintegration time and tablet porosity. Testing of the model suggested it was reasonable though there may be some small systematic errors in disintegration time measurement. The terahertz measurements further showed that there are changes in tablet properties after storage for nine months in ambient conditions.

This study aimed at developing the Diamod® as a dynamic gastrointestinal transfer model with physically interconnected permeation. The Diamod® was validated by studying the impact of the intraluminal ...dilution of a cyclodextrin-based itraconazole solution and the negative food effect for indinavir sulfate for which clinical data are available demonstrating that the systemic exposure was strongly mediated by interconnected solubility, precipitation, and permeation processes. The Diamod® accurately simulated the impact of water intake on the gastrointestinal behavior of a Sporanox® solution. Water intake significantly decreased the duodenal solute concentrations of itraconazole as compared to no intake of water. Despite this duodenal behavior the amount of permeated itraconazole was not affected by water intake as observed in vivo. Next to this, the Diamod® accurately simulated the negative food effect for indinavir sulfate. Different fasted and fed state experiments demonstrated that this negative food effect was mediated by an increased stomach pH, entrapment of indinavir in colloidal structures and the slower gastric emptying of indinavir under fed state conditions. Therefore, it can be concluded that the Diamod® is a useful in vitro model to mechanistically study the gastrointestinal performance of drugs.

This study aimed to evaluate and better understand the potential impact that a layer of surrounding hydrogel (mimicking living tissue) can have on the drug release from PLGA microparticles. ...Ibuprofen-loaded microparticles were prepared with an emulsion solvent extraction/evaporation method. The drug loading was about 48%. The surface of the microparticles appeared initially smooth and non-porous. In contrast, the internal microstructure of the particles exhibited a continuous network of tiny pores. Ibuprofen release from
microparticles was measured into agarose gels and well-agitated phosphate buffer pH 7.4. Optical microscopy, scanning electron microscopy, differential scanning calorimetry, X-ray powder diffraction, and X-ray μCT imaging were used to characterize the microparticles before and after exposure to the release media. Importantly, ibuprofen release was much slower in the presence of a surrounding agarose gel, e.g., the complete release took two weeks vs. a few days in well agitated phosphate buffer. This can probably be attributed to the fact that the hydrogel sterically hinders substantial system swelling and, thus, slows down the related increase in drug mobility. In addition,
, the convective flow in agitated bulk fluid likely damages the thin PLGA layer at the microparticles' surface, giving the outer aqueous phase more rapid access to the inner continuous pore network: Upon contact with water, the drug dissolves and rapidly diffuses out through a continuous network of water-filled channels. Without direct surface access, most of the drug "has to wait" for the onset of substantial system swelling to be released.
