We evaluated the impact of a participatory training of pig farmers on knowledge, attitude and practices (KAP) of biosecurity relating to ASF control in two districts of Uganda using a randomized ...control trial (RCT). A total of 830 pig farmers from 32 villages were included in the study, with 425 farmers receiving training, while 405 did not. An item response theory model was used to assess the impact of the training on farmer's KAP. Logistic regression models were used to assess the factors that affected knowledge gain and change in attitude and practices after training. Focus group discussions (FGD) were carried out with selected farmers from the treatment group at the end of the intervention, to share their experiences and discuss potential factors that could hinder adoption of biosecurity in their communities. Results of the regression analyses showed that there was a significant effect of biosecurity training (p = .038) on gain in knowledge after 12 months, but there were limited changes in farmer's attitude and practice at 12 and 28 months after training. Pig production domain (peri‐urban/urban production), group membership, gender (male) and education of the farmer positively influenced knowledge gain and attitude of farmers towards biosecurity. This paper provides empirical evidence on the impact of training intervention on biosecurity practices for disease prevention or control. In addition, it breaks down the components of the biosecurity practices and documents the specific challenges to its uptake by the farmers. It therefore relaxes the assumption of knowledge constraint as a barrier to uptake. The results clearly show that knowledge is not the binding constraint to uptake of the biosecurity interventions.
Distillers dried grains with solubles (DDGS) may be included in diets fed to pigs in all phases of production. The concentrations of DE and ME in DDGS are similar to those in corn. Phosphorus in DDGS ...is highly digestible to pigs and apparent total tract digestibility values of approximately 60% have been reported. The concentration of starch in DDGS is low (i.e., between 3 and 11%), but the concentration of fat in DDGS is approximately 10% and the concentrations of ADF, NDF, and total dietary fiber in DDGS are approximately 3 times greater than those in corn (9.9, 25.3, and 42.1%, respectively). The apparent total tract digestibility of dietary fiber is less than 50%, which results in reduced digestibility values for DM and energy in DDGS. The concentrations of most AA in DDGS are approximately 3 times greater than those in corn, but the standardized ileal digestibility of most AA is approximately 10 percentage units less than in corn. Nursery pigs from 2 to 3 wk postweaning, and growing and finishing pigs may be fed diets containing up to 30% DDGS without any negative impact on growth performance. However, the carcass fat in pigs fed diets containing DDGS has a greater iodine value than the carcass fat in pigs not fed DDGS. It may, therefore, be necessary to withdraw DDGS from the diet of finishing pigs during the final 3 to 4 wk before slaughter to achieve the desired pork fat quality. Lactating sows can also be fed diets containing up to 30% DDGS, and DDGS can replace all the soybean meal in diets fed to gestating sows without negatively affecting sow or litter performance. Inclusion of DDGS in diets fed to pigs may improve immune system activation, but more research is needed to elucidate the mechanisms responsible for these effects. Manure volume will increase when DDGS is included in the diets because of the reduced digestibility of DM in DDGS. Nitrogen excretion may also increase, but this can be prevented by the use of crystalline AA in diets containing DDGS. In contrast, P excretion can be reduced in diets containing DDGS if the total dietary concentration of P is reduced to compensate for the greater digestibility of P in DDGS. In conclusion, DDGS can be included in diets fed to growing pigs in all phases of production, beginning at 2 to 3 wk postweaning, in concentrations of up to 30% DDGS, and lactating and gestating sows can be fed diets containing up to 30 and 50%, respectively, without negatively affecting pig performance.

African swine fever (ASF) is a major threat to domestic pigs and wild boars. Since 2018, ASF outbreaks have been ongoing in China. As of August 3, 2019, a total of 151 ASF clusters of outbreaks reported in China have caused severe economic losses for the industry, the pig farmers and pork producers, due to the lack of an efficacious vaccine. The present study aims to analyze the epidemiologic characteristics of ASF outbreak that occurred in several regions across China during the period August 2018- August 2019. Particular focus was on the epidemic distribution, main transmission routes, incidence/fatality, impact on pig production capacity, and the main preventive measures adopted to mitigate the risk of ASF spread in pig farming systems by Chinese government. Results show that anthropogenic factors, spatial distribution, efficient measures taken by China,and good response timely in implementation of preventive measures are important on the transmission of ASF and these suggest that effective ASF risk management in China will require a comprehensive and integrated approach linking science and implemented by all relevant stakeholders. This provides an empirical basis to optimize current interventions as well as develop new tools and strategies to reduce the risk transmission of African swine fever virus (ASFV) to domestic pigs and wild boars.
Although classical swine fever (CSF) has been well known for decades and epidemics still occur, clinical diagnosis continues to cause problems for veterinary practitioners. This is due to the ...extensive differential diagnosis, further complicated by the emergence of new diseases such as porcine reproductive and respiratory syndrome (PRRS) and porcine dermatitis and nephropathy syndrome (PDNS). In addition, acute, chronic and prenatal courses of CSF have to be distinguished. As a cause of considerable economical losses within the EU, control of CSF requires knowledge of the primary outbreaks and spread of the disease.
Genetic typing of CSF virus isolates has proved to be a potent method of supporting epidemiological investigations. Phylogenetic analysis of CSF virus strains and isolates originating from different continents has allowed three genetic groups and several subgroups within these groups to be distinguished. Whereas isolates belonging to group 3 seem to occur solely in Asia, all CSF virus isolates of the 1990s isolated in the EU belonged to one of the subgroups within group 2 (2.1, 2.2, or 2.3) and were clearly distinct from former CSF reference viruses, which belong to group 1.
Within the EU, different strategies are followed for the eradication of CSF in domestic pigs and in wild boar. While a strict non-vaccination policy is followed for domestic pigs, eradication of the disease in wild boar is more complex, and oral immunisation together with special hunting strategies have been applied. Recently, marker vaccines with a companion discriminatory test designed to allow differentiation between vaccinated animals and animals having recovered from field virus infection have been developed. Preliminary studies indicated that the discriminatory tests had a reduced sensitivity and specificity. Further improvements are therefore necessary before marker vaccines can be considered for emergency use in EU Member States. Prevention of CSF remains the main objective within the EU.
African swine fever is a viral hemorrhagic disease of pigs (Sus scrofa) caused by a double-stranded DNA virus, African swine fever virus (ASFV). With up to 100% mortality in pigs and no vaccine, the ...socioeconomic impacts of this disease are immense. In sub-Saharan Africa, warthogs (Phacochoerus africanus) are the original vertebrate host for the virus, which is transmitted among wild suids via the soft tick Ornithodoros moubata. In Malawi, O. moubata is thought to be widely distributed, with the potential to spread ASFV beyond its historical enzootic zone in the Central Region. We surveyed 76 domestic pig farms, 18 active warthog burrows, and three bushpigs (Potamochoerus larvatus) resting sites for O. moubata in the Central Region of Malawi, and tested the ticks for ASFV using PCR. We found a limited distribution for O. moubata: 75 ticks were discovered at a single farm in the Mchinji District. Eleven percent of these ticks were ASFV positive. This suggests that wildlife and O. moubata play a limited role in the epidemiology of ASF in Malawi; thus, other factors for disease spread, such as fomites and pig-to-pig infection, need to be considered.

Porcine deltacoronavirus (PDCoV) has caused enormous economic losses to the global pig industry. However, the immune escape mechanism of PDCoV remains to be fully clarified. Transcriptomic analysis ...revealed a high abundance of interferon (IFN)-induced protein with tetratricopeptide repeats 3 (IFIT3) transcripts after PDCoV infection, which initially implied a correlation between IFIT3 and PDCoV. Further studies showed that PDCoV nsp5 could antagonize the host type I interferon signaling pathway by cleaving IFIT3. We demonstrated that PDCoV nsp5 cleaved porcine IFIT3 (pIFIT3) at Gln-406. Similar cleavage of endogenous IFIT3 has also been observed in PDCoV-infected cells. The pIFIT3-Q406A mutant was resistant to nsp5-mediated cleavage and exhibited a greater ability to inhibit PDCoV infection than wild-type pIFIT3. Furthermore, we found that cleavage of IFIT3 is a common characteristic of nsp5 proteins of human coronaviruses, albeit not alphacoronavirus. This finding suggests that the cleavage of IFIT3 is an important mechanism by which PDCoV nsp5 antagonizes IFN signaling. Our study provides new insights into the mechanisms by which PDCoV antagonizes the host innate immune response.IMPORTANCEPorcine deltacoronavirus (PDCoV) is a potential emerging zoonotic pathogen, and studies on the prevalence and pathogenesis of PDCoV are ongoing. The main protease (nsp5) of PDCoV provides an excellent target for antivirals due to its essential and conserved function in the viral replication cycle. Previous studies have revealed that nsp5 of PDCoV antagonizes type I interferon (IFN) production by targeting the interferon-stimulated genes. Here, we provide the first demonstration that nsp5 of PDCoV antagonizes IFN signaling by cleaving IFIT3, which affects the IFN response after PDCoV infection. Our findings reveal that PDCoV nsp5 is an important interferon antagonist and enhance the understanding of immune evasion by deltacoronaviruses.
Smallholder subsistence pig production is common in Uganda and African swine fever (ASF) is endemic in the country, with its spread driven by human activities along the smallholder value chain. ...Previous research in the study area has revealed that many stakeholders are aware of how ASF is spread, its prevention and control, and have a generally positive attitude towards biosecurity. Despite this, even basic biosecurity is largely lacking. Costs, as well as a lack of adaptation to the local context, culture and traditions have been identified as factors hindering biosecurity implementation. Community engagement and local ownership of disease problems are increasingly recognised as important for improving disease prevention and control. The objective of this study was to investigate the capacity of participatory action at community level with broad inclusion of stakeholders to improve biosecurity in the smallholder pig value chain. Specific attention was paid to participants’ perceptions and experiences of implementing the biosecurity measures included in their co-created community contracts. The study was conducted in Northern Uganda in villages purposively selected on the basis of previous occurrences of ASF. In each village, farmers and traders were also purposively selected. At a first meeting, basic information about ASF was shared and participants presented with a list of biosecurity measures adapted for farmers and traders respectively. Participants discussed each measure in farmer and trader subgroups, decided on the measures to implement for one year, and signed a community contract to this effect. The following year, interviews were again undertaken and implementation support given. Interview data were coded and thematically analysed. Each subgroup chose a minimum of three and a maximum of nine measures, with wide variations between villages in their selection of measures. At the follow-ups, none of the subgroups had fully implemented what had been agreed in their contract, but all had changed some of their biosecurity routines. Some frequently recommended biosecurity measures, such as not borrowing breeding boars, were not considered feasible. Relatively simple and cheap biosecurity measures were rejected for reasons of cost, highlighting the participants’ general level of poverty and the relevance of poverty as a specific factor governing disease control results. The participatory methodology allowing for discussions, co-creation and the option to refuse measures seemed to facilitate the implementation of measures that had initially been thought to be controversial. The broad community approach was deemed to be positive for strengthening community identity, cooperation and implementation.
To verify whether porcine deltacoronavirus infection induces disease, we inoculated gnotobiotic pigs with 2 virus strains (OH-FD22 and OH-FD100) identified by 2 specific reverse transcription PCRs. ...At 21-120 h postinoculation, pigs exhibited severe diarrhea, vomiting, fecal shedding of virus, and severe atrophic enteritis. These findings confirm that these 2 strains are enteropathogenic in pigs.

An overexpression of CD2v and p54 proteins does not alter the characteristics of PRV-∆TK/∆gE. Moreover, PRV-∆TK-(CD2v)-∆gE-(p54) successfully induces antibody production via intramuscular (IM) vaccination and confers effective protection for vaccinated mice upon challenge. Thus, PRV-∆TK-(CD2v)-∆gE-(p54) demonstrates good immunogenicity and safety, providing highly effective protection against PRV and ASFV. It potentially represents a suitable candidate for the development of a bivalent vaccine against both PRV and ASFV infections.
