U ovoj disertaciji konstruiran je prostorno-vremenski ...glikomski profil mozga, istražujući N-glikanske strukture četiriju funkcionalno različitih regija mozga, fronatalna dorsalna kora velikog mozga, hipokampus, strijatum te mali mozak, i njihovu raspodjelu u mozgu odraslog čovjeka; kao i u mozgu čovjeka tijekom fetalnog razvoja. Također, s namjerom da se dobije evolucijska perspektiva N-glikanskih struktura unutar različitih regija mozga, napravljena je usporedba odgovarajućih regija mozga između glikoma mozga čovjeka, čimpanzi, makaki majmuna i štakora. Određivanje glikanskih profila uzoraka mozga je provedeno korištenjem dvaju analitičkih pristupa: tekućinske kromatografije ultravisoke djelotvornosti i spektrometrije masa. Uvidom u N-glikom analiziranih uzoraka, uočena je značajna razlika između različitih regija, vrsta i razvoja mozga. Razlika između N-glikoma je bila veća što su vrste bile evolucijski udaljenije. N-glikanski profil malog mozga se najviše razlikovao od ostalih analiziranih regija. Razvijena HILIC-UPLC metoda za analizu N-glikana tkiva mozga se pokazala reproducibilnom i robusnom.- Protein N-glycosylation is crucial for the development and function of the nervous system. In this dissertation, a spatio-temporal profile of the brain glycome was constructed by investigating N-glycan structures of four functionally distinct brain regions, the dorsal fronatal cortex, the hippocampus, the striatum and the cerebellum, and their distribution within the adult human brain as well as the fetal. Moreover, in order to obtain an evolutionary perspective of the corresponding brain regions, a cross-species comparison was made between the human, the chimpanzee, the macaque and the rat. Two analytical approaches were used for glycosylation profiling of brain samples: ultra-performance liquid chromatography and mass spectrometry. Significant difference was noticed between the regions, the species and the developmental stages in the analyzed N-glycomes. The difference between species was higher as they were evolutionary more distinct. The cerebellar N-glycom was the most distinctive of all the analyzed brain regions. Crystal structure of the truncated form of the HpFlgD protein revealed that spatial orientation of the two domains differs from that of the homologous FlgD family members. This fact together with the observation that truncated HpFlgD assembles into tetramers, both in the solution and in the crystal form, strongly suggests that significant differences exist in the molecular organization of the flagella in different bacterial species. It was shown that incubation of the putative copper binding CrdA protein with Cu2+ ions favours formation of monomeric species in solution and that CrdA binds Cu2+ with very low affinity which is a property of copper trafficking proteins. Functional assays of the HP1026 protein demonstrated for the first time its ATPase activity. While proteins that belong to the class of AAA+ proteins usually form hexamers, HP1026 was found to form dimers.- U sklopu ove disertacije strukturno su okarakterizirani proteini iz bakterije H. pylori koji su važni za njezino preživljavanje (HpFlgD, CrdA, HP1026). Riješena kristalna struktura skraćenog proteina HpFlgD ukazala je na drugačiju međusobnu orijentaciju dviju domena nego što je nađeno u ostalim homolozima proteina FlgD. Također je pokazano da je skraćeni HpFlgD i u otopini i kristalu prisutan kao tetramer što ukazuje na značajnu razliku u molekularnoj organizaciji biča u različitih bakterijskih vrsta. Uočeno je da dodatak iona Cu2+ u otopinu CrdA, proteina koji pretpostavljeno veže bakrove ione, potiče stvaranje monomernih vrsta u otopini te da CrdA veže ione Cu2+ sa slabim afinitetom što je karakteristika proteina koji prenose bakrove ione. Funkcionalna karakterizacija proteina HP1026 je po prvi put demonstrirala da HP1026 obavlja ATPaznu aktivnost. Istraživanje je usmjereno na optimiranje procesnih parametara priprave YSZ filmova. Određen je utjecaj udjela stabilizatora kristalne faze, broja slojeva, brzine nanošenja slojeva, vremena kvašenja te temperature i atmosfere toplinske obrade. Za karakterizaciju sol-gel filmova ZrO2 i njegovih praškastih oblika korištene su sljedeće analitičke tehnike: difrakcija rentgenskog zračenja u polikristalnom uzorku (PXRD), infracrvena spektroskopija s Fourierovom transformacijom (FTIR), Ramanova spektroskopija, diferencijalna toplinska analiza/termogravimetrija (DTA/TG) i diferencijalna pretražna kalorimetrija/termogravimetrija (DSC/TG), analiza oslobođenih plinova sa spektrometrijom masa (EGAMS), optička emisijska spektrometrija s tinjajućim izbojem (GDOES), pretražna elektronska mikroskopija uz energijski razlučujuću rentgensku spektroskopjiu (SEM/EDS) te mikroskopija atomskih sila (AFM). Istraživanja korozijske otpornosti nezaštićenog nehrđajućega čelika te čelika prevučenog nanostrukturnim sol-gel ZrO2 filmovima provedena su elektrokemijskim mjernim tehnikama potenciodinamičke polarizacije (Tafelova ekstrapolacija) i elektrokemijske impedancijske spektroskopije (EIS) u mirujućem simuliranom morskom okolišu (3,5%-tna vodena otopina NaCl). Dugotrajnost zaštite koju pružaju keramički filmovi ispitana je elektrokemijskom impedancijskom spektroskopijom (EIS). Elektrokemijskim mjerenjima utvrđeno je da nanostrukturni sol-gel ZrO2 filmovi, posebice troslojni filmovi toplinski obrađeni u vakuumu na temperaturi 400 C, značajno povećavaju korozijsku postojanost nehrđajućega čelika u 3,5%-tnoj vodenoj otopini NaCl.- Abstract In this thesis nanostructured films of zirconia stabilized with yttria were prepared by sol-gel method and deposited on the austenitic stainless steel AISI 316L (X2CrNiMo17-2-2) surface by dip coating method. Process parameters optimisation for YSZ films was the aim of the study. Influence of the amount of crystal structure stabiliser, number of layers, withdrawal speed, soaking time, temperature and atmosphere of thermal processing were determined. For the characterization of sol-gel ZrO2 films and its powders following analytical techniques were used: powder X-ray diffraction (PXRD), Fourier transform infrared (FTIR) and Raman spectroscopies, differential thermal/thermogravimetric analysis (DTA/TG), differential scanning calorimetry/thermogravimetric analysis (DSC/TG), evolved gas analysis-mass spectrometry (EGAMS), glow-discharge optical emission spectrometry (GDOES), scanning electron microscopy with energy dispersive X-ray spectroscopy (SEM/EDS), atomic force microscopy (AFM). Electrochemical corrosion behaviour of the sol-gel ZrO2 films coated and uncoated stainless steel substrates was evaluated by electrochemical tests such as Tafel extrapolation and electrochemical impedance spectroscopy (EIS) in simulated marine environment (3.5 wt.% aqueous NaCl solution).
