The metagenomic dataset profiled in this research is built on bacterial 16S rRNA gene amplicon of DNA mined from barley rhizosphere under chemical (CB) and organic (OB) fertilization. Amplicon-based ...sequencing was prepared by the Illumina platform, and the raw sequence dataset was examined using Metagenomic Rast Server (MG-RAST). The metagenome comprised sixteen samples that include CB1 (494,583 bp), CB2 (586,532 bp), CB3 (706,685 bp), CB4 (574,606 bp), CB5 (395,460 bp), CB6 (520,822 bp), CB7 (511,729 bp), CB8 (548,074 bp), OB1 (642,794 bp), OB2 (513,767 bp), OB3 (461,293 bp), OB4 (498,241 bp), OB5 (689,497 bp), OB6 (423,436 bp), OB7 (478,657 bp) and OB8 (279,186 bp). Recent advances in high-volume sequencing technology and mining of genomes from metagenomic samples call for rapid and reliable genome quality evaluation. The current release of the PATRIC database ...contains over 220,000 genomes, and current metagenomic technology supports assemblies of many draft-quality genomes from a single sample, most of which will be novel.
We have added two quality assessment tools to the PATRIC annotation pipeline. EvalCon uses supervised machine learning to calculate an annotation consistency score. EvalG implements a variant of the CheckM algorithm to estimate contamination and completeness of an annotated genome.We report on the performance of these tools and the potential utility of the consistency score. Additionally, we provide contamination, completeness, and consistency measures for all genomes in PATRIC and in a recent set of metagenomic assemblies.
EvalG and EvalCon facilitate the rapid quality control and exploration of PATRIC-annotated draft genomes.
Anaerobic performance is considered an indicator of performance for short-term muscle activities of high intensity. It is important to determine whether different anaerobic field tests performed to ...measure anaerobic performance can be used as an alternative to WAnT performed in the laboratory. The study aimed to compare the anaerobic performance with laboratory and field tests in trained children. One-hundred four athletes between the ages of 10 and 16, minimum age of training of one year in different sports voluntarily participated in the study. Wingate Anaerobic Test (WAnT) was performed as the reference test. Besides, Running-based Anaerobic Sprint Test (RAST) and Pediatric RAST (PRAST) were performed to determine anaerobic performance. Peak power (PP), average power (AP), minimum power (MP), fatigue index (FI) and total exercise time (TED) were determined for each test. All variables of WAnT, RAST, and PRAST were significantly different (p<0.01). According to test-retest results of all tests, ICC 95% CI values have a high-reliability coefficient for all variables. It was found there is a high correlation significantly between WAnT and RAST for all variables (p<0.01). Besides, there were also high correlations significantly between WAnT-PRAST and RAST-PRAST excluding fatigue index (p<0.01). As a result of this study, it was determined all tests have high reliability. Considering that WAnT requires complex, expensive device and tools, trained staff and is performed in the form of cycling in the laboratory, RAST and PRAST performed with body weight in field conditions can be used to determine anaerobic performance in trained children. High correlations between tests support this determination.

sp. strain AN1) have been whole genome sequenced and annotated. In the present study we have mapped and compared the virulence profile of
strain CD08_7 along with other reference genomes which showed some characteristic features that could contribute to pathogenicity. The RAST (Rapid Annotation using Subsystem Technology) based genome mining revealed more genes responsible for pathogenicity in strain CD08_7 when compared with the other four sequenced strains. The studied categories were resistance to antibiotic and toxic compounds, invasion and intracellular resistance, membrane transport, stress response, osmotic stress, oxidative stress, phages and prophages and iron acquisition. A total of 1431 protein-encoding genes were identified in the genome of strain CD08_7 among which 163 were predicted to contribute for pathogenicity. Out of 163 genes only 59 were common to other genome, which shows the higher levels of genetic richness in strain CD08_7 that may contribute to its functional versatility. This study provides a comprehensive analysis on genome of
strain CD08_7 and possibly indicates its importance as a clinical pathogen.
In this study, rapid diagnostic of multidrug-resistant (MDR) sepsis pathogens, directly from positive blood culture (BC) bottles, was evaluated by combining MALDI-TOF and the EUCAST Rapid ...Antimicrobial Susceptibility Testing (RAST). Carbapenemase production in Escherichia coli and Klebsiella pneumoniae isolates was also evaluated by RAST. From 171 positive BC bottles analyzed, 79 (46 %) MDR species, including E. coli (4/34, 12 %), K. pneumoniae (33/48, 69 %), Pseudomonas aeruginosa (12/12, 100 %), Acinetobacter baumannii (15/15, 100 %), and Staphylococcus aureus (14/37, 38 %) displaying resistance to beta-lactams, fluoroquinolones, aminoglycosides, and/or trimethoprim/sulphamethoxazole, were identified. In this regard, turnaround time of direct MALDI-TOF identification and RAST was < 7 h, which was significantly (p< 0.05) lower than our routine method. Carbapenemase detection by RAST displayed 100% sensitivity and 88.7 % specificity at 8 h. This protocol could offer advantages for the treatment and clinical outcomes of septic patients, improving the rapid diagnostic of sepsis by MDR pathogens.

Bez energije nema ni gospodarskog rasta. Stoga je za neometan razvoj gospodarstva nužno osigurati dovoljne količine energije po pristupačnim cijenama. Sve učestalije energetske krize determiniranjem ...dostupnosti i cijena energenata snažno utječu na potrebu implementacije energetskih i ekonomskih mjera i politika koje će omogućiti neometan razvoj gospodarstava. U navedenim okvirima, zadaća je znanstvenika ponuditi nositeljima politika jasan teorijski i empirijski koncept odnosa energije i gospodarskog rasta kako bi se usvojile energetske politike koje će minimizirati ranjivost gospodarstava i omogućiti održivi gospodarski rast i međunarodnu konkurentnost. Cilj ovoga istraživanja upravo je ukazati na proturječnosti ekonomske teorije i postojećih empirijskih istraživanja međuovisnosti energije i gospodarskog rasta. Dok teorije rasta u potpunosti zanemaruju ili, u najbolju ruku, marginaliziraju ulogu energije, istraživanja uzročnosti gospodarskog rasta i potrošnje energije sve su brojnija iako, još uvijek, inkonkluzivna. Političke implikacije postojećih istraživanja stoje na klimavim nogama te bi se trebale uzeti s velikom dozom opreza. Jedino robusni i konzistentni rezultati istraživanja mogu poslužiti kao čvrsta baza donošenju adekvatnih i pravodobnih mjera ekonomske i energetske politike. Iako je dostupnost podataka jedno od ključnih ograničenja postojećih istraživanja, autori bi u svojim budućim istraživanjima trebali uvažavati multivarijantni okvir, primarno uvažavanjem cijena energije u modelima, te iskoristiti napredne ekonometrijske tehnike.
This paper presents the steady state analysis of the Rostov-II benchmark using the conventional two-step approach. It involves the STREAM/RAST-K and CASMO-5/PARCS code systems. This paper documents a ...comprehensive code-to-code comparison between Serpent 2, CASMO-5, and STREAM at the lattice level for the different fuel assemblies (FAs) loaded in the Rostov-II core; and between Serpent 2, PARCS, and RAST-K at the core level in 2D. Finally, the 3D results of both deterministic models are compared to the steady state measurements of the Rostov-II benchmark. With respect to the measurements available in the Rostov-II benchmark, comparable accuracy (30 ppm difference in boron concentration, 2% assembly power) with an industrial calculation scheme (BIPR8) are reported up to 36.73 EFPDs. The calculations reported in the paper showed that the modeling of the resonance self-shielding in the lattice code as well as the geometrical modeling of the reflector are key for an accurate solution (reducing the in-out power tilt). At the core simulator level, a fairly crude 1D reflector model appears to be enough. Overall, this paper provides the detailed models and conditions used in STREAM/RAST-K and CASMO-5/PARCS, and accurate calculation solution for the Rostov-II benchmark with STREAM/RAST-K and CASMO-5/PARCS compared with measurement.
Patients with food-induced allergic disorders may be first seen with a variety of symptoms affecting the skin, respiratory tract, gastrointestinal tract, and/or cardiovascular system. The skin and ...respiratory tract are most often affected by IgE-mediated food-induced allergic reactions, whereas isolated gastrointestinal disorders are most often caused by non-IgE-mediated reactions. When evaluating possible food-induced allergic disorders, it is often useful to categorize disorders into IgE- and non-IgE-mediated syndromes. The initial history and physical examination are essentially identical for IgE- and non-IgE-mediated disorders, but the subsequent evaluation differs substantially. Proper diagnoses often require screening tests for evidence of food-specific IgE and proof of reactivity through elimination diets and oral food challenges. Once properly diagnosed, strict avoidance of the implicated food or foods is the only proven form of treatment. Clinical tolerance to food allergens will develop in many patients over time, and therefore follow-up food challenges are often indicated. However, a number of novel immunomodulatory strategies are in the developmental stage and should provide more definitive treatment for some of these food-induced allergic disorders in the next several years. , using total lab automation. Zone diameters from RAST were compared with MIC results. Furthermore, its influence on time to report was investigated. RAST was performed to all positive aerobic and anaerobic blood culture bottles by subculturing them, i.e. onto Mueller-Hinton agar and adding six antibiotic discs covering Gram-negative and Gram-positive therapy (cefoxitin, ampicillin, vancomycin, piperacillin/tazobactam, meropenem and ciprofloxacin). RAST was automatically imaged after 6 h. Zone sizes were measured using a TLA software tool and interpreted according to EUCAST clinical breakpoints. Bacteria were identified using MALDI-TOF MS and MIC results were determined using Vitek2 panels. Categorial agreement between agar diffusion and MIC results was investigated. Additionally, time to RAST and time to Vitek were compared for 100 isolates (20 per species). Between November 2018 and April 2019, 3313 positive mono-bacterial blood culture bottles were collected of which 894 bottles with RAST-validated species were investigated. Among these bottles, 2029 individual antibiotic measurements were compared with MIC results from Vitek2 and 14 very major, 28 major and 12 minor errors were found. A median reduction of 17:30 h in time to report was observed. Introduction of RAST with automatic TLA imaging function could reduce time to report by 17:30 h. Excellent accordance between zone diameter and MIC results, particularly for cefoxitin, vancomycin and meropenem, was observed, but drawbacks due to ATU were seen.
Based on experimental pure component data for the characterization of the isostructural imidazolate framework Potsdam (IFP) series reported in Part I, a model for the simulation of non‐isothermal ...dynamic adsorption of CO2/CH4‐mixtures in fixed‐bed columns is presented in this Part II. The robustness of the model is examined and validated, by comparison to experimental breakthrough data at different process conditions, such as varying concentration, temperature, and pressure. Thereby, different predictive methods for the estimation of adsorption equilibria of mixtures are compared (RAST, IAST, ML). The results show that ideal behaviour can be assumed with good accuracy for the system under consideration, except for IFP‐2, which shows significant deviations at increased pressures and temperatures. A detailed kinetic analysis reveals that mass transfer is significantly influenced by micropore diffusion. Thus, only for IFP‐1 the dynamic separation of CO2 and CH4 is equilibrium‐driven, while for the remaining IFPs the kinetic regime dominates the process, which in some cases increases the separation efficiency (IFP‐2 to ‐7) but can also inhibit it (IFP‐8). The determined intracrystalline diffusion coefficients show very good agreement with values for metal organic framework (MOF) compounds of similar structure reported in the literature.
